Internalization-related host factors of common respiratory viruses.

Respiratory viruses impose a substantial health burden worldwide, with viral internalization into host cells being the initial step for infection establishment. This process is tightly regulated by the host cellular machinery through two major pathways: receptor-mediated endocytosis and direct membrane fusion. To clarify the role of host factors in these steps, we present human adenovirus and respiratory syncytial virus as representative non-enveloped and enveloped viruses, respectively, as models to elucidate their life cycles, focusing on how host factors mediate their distinct internalizati

Effects of Carbon Dioxide on Hemolymph and Brain Proteomes in Honey Bee Workers (Apis mellifera L.).

Carbon dioxide (CO2) is essential in beekeeping practices but its optimal dosage and physiological effects on honey bees remain unclear. This study examined CO2 tolerance and molecular responses across three developmental stages: newly emerged, nurse, and forager bees, using gradient exposure and proteomic analysis. Newly emerged bees demonstrated the highest CO2 tolerance. Hemolymph and brain exhibited distinct responses: the cytochrome P450 pathway dominated in hemolymph, while the brain displayed stage-specific strategies. Newly emerged bees activated metabolic reorganization and clearance

Stratified assessment of platelets in sepsis: from dynamic counts to functional phenotypes.

Sepsis is a major global cause of critical illness with high mortality. Sepsis-associated thrombocytopenia (SAT) is linked to markedly increased death risk and is a key indicator of poor prognosis. Although platelets are recognized as central players in the "inflammation-coagulation-immunity" network, most studies emphasize platelet counts rather than functional heterogeneity and underlying regulatory mechanisms, limiting the development of specific biomarkers and targeted therapies. Here, we characterize the "double-edged" role of platelets in sepsis. On the one hand, platelets recognize path

Metabolic Reprogramming in Endometrium During Menstrual Cycle and Its Relationship to Infertility.

The human endometrium, a complex and dynamic tissue, experiences cyclical shedding, repair, regeneration, and modification in anticipation of embryo implantation. During the mid-secretory phase, influenced by ovarian steroid hormones, it becomes briefly receptive. The functional layer of the endometrium, consisting of several cell types including epithelial, stromal, vascular, and immunological cells, is essential for reproduction as it prepares for implantation, sustains pregnancy, and enables menstruation. Progesterone generated during the secretory phase prepares the endometrium for blastoc

Integrative Biology Approaches Uncover Plasticizers' Pro-Carcinogenic Role in Prostate Cancer via Molecular Docking and Network Toxicology.

Diethyl phthalate (DEP), dimethyl phthalate (DMP), and dioctyl phthalate (DOP) are common plasticizers. This study aimed to evaluate the potential roles of these plasticizers in the development of prostate cancer (PCa) via integrative biology approaches. We employed network toxicology analyses to examine interactions between these plasticizers and proteins critical to PCa development. Enrichment analysis and protein-protein interaction network analysis were conducted to identify relevant key proteins and pathways. Molecular docking was further employed to calculate binding affinity. Enrichment

The Role of Leukemia Inhibitory Factor in Attenuating Skeletal Muscle Atrophy: Mechanisms to Exercise Interventions.

Leukemia inhibitory factor (LIF), a member of the interleukin-6 (IL-6) cytokine family, is a well-characterized myokine with pleiotropic regulatory effects on skeletal muscle. LIF modulates several fundamental cellular processes, including myoblast proliferation, apoptosis, angiogenesis, and energy metabolism. Exercise upregulates LIF expression in skeletal muscle, thereby promoting satellite cell activation, proliferation, myoblast differentiation, and angiogenesis, facilitating physiological muscle hypertrophy, and suppressing myocyte apoptosis and muscle atrophy. In addition, LIF plays a cr

Role of the Gut-brain Axis in the Cancer-immunity Cycle.

Antitumor immunity depends on a self-reinforcing cancer-immunity cycle, yet many tumors evade critical steps and respond poorly to checkpoint blockade. Emerging evidence indicates that gut microbiota-derived systemic cues could influence these therapeutic outcomes, highlighting the gut-brain axis (GBA) as a potentially important regulatory framework in cancer immunity. In this review, we summarize how GBA-related signals may influence multiple stages of the cycle through interconnected neural, endocrine, microbial, and immune pathways. Current findings suggest that these impacts are highly con

Oxidative Stress: The Role of Aluminum Ions in Copper Homeostasis Within the Neurons.

Aluminum (Al) and copper (Cu) homeostasis is critical for neuronal health. Disruption of their balance contributes to oxidative stress, mitochondrial dysfunction, and neurodegenerative diseases such as Alzheimer's disease and Parkinson's disease. Existing studies have reported a positive correlation between aluminum and copper concentrations in specific neuronal models. Such homeostatic imbalance may contribute to the activation of pathways related to apoptosis or cuproptosis through mechanisms including oxidative stress, mitochondrial dysfunction, and glutathione depletion. Aluminum-copper dy

Tau-ing and fro-ing: the tanycytic shuttle in neurodegeneration.

After more than a century since Alzheimer's disease (AD) was described and decades of research into β-amyloid and Tau proteins, mechanisms underlying pathogenic protein clearance from brain remain poorly understood. Recent research identifies tanycytes-specialized hypothalamic cells lining the third ventricle-as a previously unrecognized clearance system for brain Tau. These cells actively transport Tau from cerebrospinal fluid to blood via pituitary portal circulation but are dramatically fragmented in AD brains. Single-nucleus RNA sequencing reveals altered stress and transport gene expressi

Cardiac involvement in systemic sclerosis: mechanisms, manifestations and management.

Cardiovascular disease remains a leading cause of mortality in systemic sclerosis (SSc). This review summarizes key mechanisms in the pathophysiology of SSc primary heart involvement (SSc-PHI), presents the main clinical manifestations and their management and discusses unmet needs in SSc-PHI with potential solutions. The pathophysiology of SSc-PHI involves both vascular and immune-mediated pathways, exacerbated by the NLRP3 inflammasome and pro-fibrotic cytokines, leading to myocardial fibrosis as the main cardiac manifestation. Regular screening is recommended to detect SSc-PHI, incorporatin

Integrating frailty into decision-making for disease-modifying therapies in Alzheimer's disease: a proposed expert-opinion based approach.

The introduction of disease-modifying therapies for Alzheimer's disease (AD-DMTs) is reshaping clinical practice, raising critical questions about patient selection, diagnostic pathways, treatment appropriateness, and equity of access. Frailty, a multidimensional condition of reduced physiological reserve and increased vulnerability to stressors, is common in older adults with AD, yet has not been systematically assessed in AD-DMTs trials, limiting the generalizability of trial findings to real-world populations. In this review, we examine the role of frailty in the emerging era of AD-DMTs, su

Gut metabolite indole-3-acetic acid aggravates neuropsychiatric lupus via the AHR/STAT3 pathway in microglia.

Neuropsychiatric systemic lupus erythematosus (NPSLE) is a severe complication of systemic lupus erythematosus (SLE). Despite its high morbidity, the exact pathogenesis of NPSLE remains poorly understood, and effective therapeutic options remain unavailable. Here we show gut bacterium Lactobacillus reuteri (L. reuteri) and its metabolite indole-3-acetic acid (IAA) play key roles in the progression of NPSLE. L. reuteri and IAA induce behavioral deficits, microglial activation, pro-inflammatory cytokine secretion, neuronal loss, and blood brain barrier (BBB) disruption in female lupus-prone mice

Integrating Metabolic Modeling and Targeted Supplementation for the Rapid Detection of Clostridium tyrobutyricum in Dairy Products.

Clostridium tyrobutyricum is a major cause of late blowing defects (LBDs) in cheese, resulting in substantial economic losses. Early detection is critical for maintaining product quality. In this study, we developed a rapid detection approach integrating genome-scale metabolic modeling (GEM) with systematic culture optimization. Three media were evaluated, identifying RCM at 38.5 °C as the optimal condition for reducing the lag phase. Flux Balance Analysis (FBA) revealed that targeted supplementation with magnesium, zinc, Vitamin B6, and L-tryptophan significantly enhanced metabolic flux throu

The pathogenic role of double-negative B cells in autoimmune diseases.

Double-negative (DN) B cells have emerged as pivotal players in autoimmune diseases, transitioning from an overlooked exhausted subpopulation to a lineage with potent pathogenic potential. Based on surface marker expression, DN B cells are subdivided into four distinct subsets: DN1, DN2, DN3, and DN4. This review provides a comprehensive overview of these subsets, focusing on their origins, phenotypes, and developmental trajectories. In disorders such as systemic lupus erythematosus (SLE), rheumatoid arthritis (RA), primary Sjögren's syndrome (pSS), and multiple sclerosis (MS), expanded DN B c

PI5P4Kα promotes glucose and iron acquisition to support metabolic fitness in pancreatic cancer.

Phosphoinositide kinases generate distinct phosphoinositides that regulate pathways to support tumorigenesis. Phosphatidylinositol 5-phosphate 4-kinases (PI5P4Ks) have garnered interest for their role in cancer metabolism; however, their function in pancreatic ductal adenocarcinoma (PDAC) remains unexplored. We identify PI5P4Kα as a critical dependency to support the unique metabolic demands of PDAC cells through its key role in the acquisition of essential metabolic substrates, including glucose and iron. Our data show that inhibition of PI5P4Kα creates a metabolic bottleneck that PDAC cells

Dopaminergic signalling in gastrointestinal health and disease.

Enteric dopaminergic signalling has a critical role in gastrointestinal motility, maintaining mucosal integrity and modulating the gut microbiome. In this Review, we provide an overview of dopamine metabolism and signalling pathways in the central nervous system and periphery and their effects on gastrointestinal health and disease. We describe the physiological role of enteric dopamine, including a discussion of therapeutic opportunities and future research needs. With rising interest in the gut-first hypothesis of Parkinson disease, an area of great interest is how dopaminergic depletion in

DJ-1/PARK7 in Parkinson's disease: mechanisms of pathogenesis and therapeutic potential.

Parkinson's disease (PD) is the second most prevalent neurodegenerative disorder, characterized by the substantial degeneration of nigral dopaminergic neurons, the accumulation of alpha-synuclein, and the intraneuronal formation of Lewy bodies. DJ-1 is a multifaceted protein that has been linked to the pathogenesis of neurodegenerative diseases. Recent research has highlighted a potential role for DJ-1 in PD. This protein plays a critical role in maintaining mitochondrial homeostasis, modulating apoptosis, facilitating chaperone-mediated autophagy (CMA), and stabilizing dopamine levels by inte

Prenatal BPA exposure disrupts androgen synthesis in testicular Leydig cells via epigenetic modifications of steroidogenic factor 1.

Prenatal exposure to bisphenol A (BPA) adversely affects testicular development and interferes with androgen synthesis in male offspring. This study explored the epigenetic mechanisms underlying BPA-induced testicular dysfunction using TM3 Leydig cells and offspring exposed to BPA during gestation. High-dose BPA treatment led to decreased testicular weight, disorganization of seminiferous tubules, and reduced testosterone production. Transcriptomic profiling demonstrated downregulation of Steroidogenic Factor 1 (Sf1) and suppression of critical steroidogenic pathways. These molecular alteratio

Distinct type I and II interferon responses direct cortical and medullary thymic epithelial cell development.

Advances in genomics have redefined our understanding of thymic epithelial heterogeneity and architecture, yet signals driving thymic epithelial differentiation remain incompletely understood. Here, we elucidated pathways instructing human thymic epithelial cell development in the context of other anterior foregut-derived organs. Activation of interferon response gene regulatory networks distinguished epithelial cells of the thymus from those of other anterior foregut-derived organs. Thymic cortex and medulla epithelia displayed distinctive interferon-responsive signatures defined by lineage-s

Known yet underdiagnosed: Invasive assessment of coronary microvascular disease and its implications.

Coronary microvascular disease (CMD) is one of the commonest causes of cardiac chest pain. The condition is more prevalent in women, and incidence is known to increase with age, hypertension, and diabetes. The pathophysiological pathways are heterogenous and related to intrinsic vascular and endothelial dysfunction. Furthermore, this entity is known to be associated with adverse cardiovascular outcomes. Despite this, there is inertia amongst cardiologists to further evaluate patients with non-critical coronary artery disease and suspected CMD. With refinement in technology, we have now better
