Escherichia coli isolates from commercial chicken meat and eggs cause sepsis, meningitis and urinary tract infection in rodent models of human infections.

The zoonotic potential of Escherichia coli from chicken-source food products is important to define for public health purposes. Previously, genotypic and phenotypic screening of E. coli isolates from commercial chicken meat and shell eggs identified some E. coli strains that by molecular criteria resembled human-source extraintestinal pathogenic E. coli (ExPEC). Here, to clarify the zoonotic risk of such chicken-source E. coli, we compared selected E. coli isolates from chicken meat and eggs, stratified by molecularly defined ExPEC status, to human-source ExPEC and to laboratory E. coli for vi

A novel human leiomyoma tissue derived matrix for cell culture studies.

BACKGROUND: The composition of the matrix molecules is important in in vitro cell culture experiments of e.g. human cancer invasion and vessel formation. Currently, the mouse Engelbreth-Holm-Swarm (EHS) sarcoma-derived products, such as Matrigel®, are the most commonly used tumor microenvironment (TME) mimicking matrices for experimental studies. However, since Matrigel® is non-human in origin, its molecular composition does not accurately simulate human TME. We have previously described a solid 3D organotypic myoma disc invasion assay, which is derived from human uterus benign leiomyoma tumor

Comparative genomic and phylogenetic analysis of the first Usutu virus isolate from a human patient presenting with neurological symptoms.

Usutu virus (USUV) is a mosquito-borne flavivirus, belonging to the Japanese encephalitis antigenic complex, that circulates among mosquitoes and birds. We describe and analyze the complete genome sequence of the first USUV strain isolated from an immunocompromised patient with neuroinvasive disease. This USUV isolate showed an overall nucleotide identity of 99% and 96%, respectively, with the genomes of isolates from Europe and Africa. Comparison of the human USUV complete polyprotein sequence with bird-derived strains, showed two unique amino acid substitutions. In particular, one substituti

Effects of aripiprazole and its active metabolite dehydroaripiprazole on the activities of drug efflux transporters expressed both in the intestine and at the blood-brain barrier.

The inhibition potencies of aripiprazole and its active metabolite, dehydroaripiprazole, on the activities of human multidrug resistance protein 1 (MDR1/ABCB1; P-glycoprotein), breast cancer resistance protein (BCRP/ABCG2) and multidrug resistance-associated protein 4 (MRP4/ABCC4), that are drug efflux transporters expressed both in the intestine and at the blood-brain barrier (BBB), were investigated. Aripiprazole and dehydroapripiprazole showed relatively strong inhibitory effects on human MDR1 with IC(50) values of 1.2 and 1.3 µm in human MDR1-transfected Mardin-Darby canine kidney (MDCKII-

Widespread transposable element dysregulation in human aging brains with Alzheimer's disease.

INTRODUCTION: Transposable element (TE) dysregulation is associated with neuroinflammation in Alzheimer's disease (AD) brains. Yet, TE quantitative trait loci (teQTL) have not been well characterized in human aged brains with AD. METHODS: We leveraged large-scale bulk and single-cell RNA sequencing, whole-genome sequencing (WGS), and xQTL from three human AD brain biobanks to characterize TE expression dysregulation and experimentally validate AD-associated TEs using CRISPR interference (CRISPRi) assays in human induced pluripotent stem cell (iPSC)-derived neurons. RESULTS: We identified 26,

Tropism of and innate immune responses to the novel human betacoronavirus lineage C virus in human ex vivo respiratory organ cultures.

Since April 2012, there have been 17 laboratory-confirmed human cases of respiratory disease associated with newly recognized human betacoronavirus lineage C virus EMC (HCoV-EMC), and 7 of them were fatal. The transmissibility and pathogenesis of HCoV-EMC remain poorly understood, and elucidating its cellular tropism in human respiratory tissues will provide mechanistic insights into the key cellular targets for virus propagation and spread. We utilized ex vivo cultures of human bronchial and lung tissue specimens to investigate the tissue tropism and virus replication kinetics following exper

A call for renewed research on tick-borne Francisella tularensis in the Arkansas-Missouri primary national focus of tularemia in humans.

Arkansas-Missouri has emerged as the primary U.S. focus of tularemia, which is caused by the National Institute of Allergy and Infectious Diseases Category A Priority Pathogen Francisella tularensis, over the past 30 yr. There are several pieces of indirect evidence suggesting that a key role of ticks in the transmission of F. tularensis to humans in Arkansas-Missouri is the primary reason why tularemia has remained a prominent disease of humans in this two-state area while fading away from other central or eastern states after a general decline in rabbit-associated tularemia cases. The primar

Human babesiosis: an emerging tick-borne disease.

Human babesiosis is an important emerging tick-borne disease. Babesia divergens, a parasite of cattle, has been implicated as the most common agent of human babesiosis in Europe, causing severe disease in splenectomized individuals. In the US, Babesia microti, a babesial parasite of small mammals, has been the cause of over 300 cases of human babesiosis since 1969, resulting in mild to severe disease, even in non-splenectomised patients. Changing ecology has contributed greatly to the increase and expansion of human babesiosis in the US. A relatively recently described babesial parasite, the W

Advances and challenges in experimental models of posttraumatic epilepsy for therapeutic interventions.

Epilepsy affects over 80 million people worldwide, with approximately 40% experiencing refractory seizures. Despite some progress in deciphering the complex process of epileptogenesis, which transforms a healthy brain into one susceptible to epilepsy, there are still no therapies available to prevent this condition. Traumatic brain injury (TBI) is a leading cause of epilepsy in both military and civilian populations, often leading to the complex condition of posttraumatic epilepsy (PTE). Defined as recurrent seizures following TBI, PTE lacks effective treatment options, highlighting the need f

Human induced pluripotent stem cells and organoid models in space for investigating severe pediatric metabolic and congenital disorders.

As biomedical research in space expands, microgravity and spaceflight-associated stressors are revealing cellular responses that may provide new insights into rare and severe metabolic and congenital disorders. Human induced pluripotent stem cells (hiPSCs) and organoids are particularly promising as they enable patient-specific modelling in the space environment. In this review, we examine the current literature on space-based hiPSC experimental models, highlighting their relevance to pediatric disorders with well-defined genetic backgrounds, namely mitochondrial diseases and congenital heart

Validated computational modeling to evaluate spatial inactivation of airborne pathogens by Far-UVC irradiation.

Recent experimental studies have indicated the potential for ultraviolet C irradiation at 222 nm (Far-UVC) to be used in occupied rooms to safely reduce exposure to airborne pathogens. We present simulations coupling Monte Carlo radiation transfer with computational fluid dynamics to predict the spatial variation in airborne microorganism inactivation. Our simulations effectively reproduce data from steady-state experiments in a room-sized bioaerosol chamber for the reduction of aerosolized Staphylococcus aureus. Application of the validated model suggests that germicidal Far-UVC lamps could r

Uranium Isotopes: From Geosciences to Biomedical Sciences.

PURPOSE OF REVIEW: Current biomarkers of uranium (U) exposure do not distinguish among exposure sources or provide information on uranium accumulation within target organs. This review evaluates two complementary uranium isotope systems developed in geosciences and examines their potential as biomarkers of uranium exposure and biological processing. RECENT FINDINGS: Advances in multi-collector ICP-MS have established δ238U and uranium activity ratios or (234U/238U) as independent isotope tracers. Experimental studies show that δ238U fractionates during biological uptake, transport, and tissue

An algorithm underlying directional hearing in fish.

Humans and other land vertebrates localize sound by comparing the signals in each ear. Even though these differences are virtually absent underwater, fish are still able to sense the direction of sound. In 1975, Arie Schuijf proposed that this ability could arise from a comparison of the particle motion phase and the pressure phase of sound, a prediction that was recently confirmed experimentally for near-field sounds. However, in natural environments, sounds arrive from variable distances, altering the motion-pressure-phase relationship. Thus, directional hearing and distance hearing are pote

Characterization of a Heme-Binding Radical SAM Enzyme HemW from Moraxella catarrhalis.

Moraxella catarrhalis is an emerging human respiratory pathogen responsible for various infectious diseases. Recent transposon sequencing analysis identified yggW (renamed hemW) as essential for M. catarrhalis growth under iron-limiting conditions, mimicking host-imposed nutritional immunity. HemW is annotated as a putative radical S-adenosylmethionine (SAM) enzyme and belongs to the HemN-like subfamily, but its biochemical properties remain unclear. Here, we report on the first experimental characterization of M. catarrhalis HemW (McHemW). Our bioinformatic analysis confirmed its evolutionary

Crocins Ameliorate Experimental Immune Checkpoint Inhibitor-Related Myocarditis by Targeting the Hpx/Nrf2/HO-1 Pathway.

Immune checkpoint inhibitors (ICIs) for cancer therapy may induce immune-related adverse events including myocarditis, which occurs infrequently but carries a high mortality rate. Crocins are the active constituents derived from Crocus sativus L. (saffron), and have demonstrated various bioactivities including anti-tumor, anti-inflammation, antioxidation, anti-ischemia, anti-aging, and neuroprotective effects. This study established a subcutaneous xenotransplanted tumor model of human liver cancer in nude mice to better mimic ICI-related myocarditis. Animal experimental results revealed that c

Beyond Hyperglycaemia in Diabetic Bone Disease: Refining Experimental Models of Diabetes-Induced Osteopathy.

Diabetes-induced osteopathy represents a clinically significant but under recognised complication of both type 1 and type 2 diabetes, characterised by increased fracture risk despite normal or modestly reduced bone mineral density. This paradox reflects deterioration in bone quality driven by advanced glycation end products, oxidative stress, chronic inflammation, impaired osteoblast function, enhanced osteoclast activity and disrupted osteocyte signalling. Although experimental models have provided valuable mechanistic insights, current chemically induced, diet-induced, and genetic systems ea
