A low dose of recombinant interleukin 1 protects granulocytopenic mice from lethal gram-negative infection.

Natural and synthetic immunomodulators that increase nonspecific resistance to infection induce interleukin 1 (IL-1) production. Therefore, we investigated the effect of the administration of IL-1 on the survival of lethally infected granulocytopenic mice. Mice with cyclophosphamide-induced granulocytopenia were injected with approximately 10(7) Pseudomonas aeruginosa in the thigh muscle at time 0; gentamicin was administered 6 hr and 23 hr later. When recombinant human IL-1 beta (one of the two forms of IL-1) was given as a single i.p. injection 24 hr before the infection, survival was increa

Expression of HLA class II antigens and secretion of interleukin-1 by monocytes and macrophages from adults and neonates.

HLA class II antigen expression and IL-1 production by mononuclear phagocytes are important for antigen-stimulated T-cell activation. We examined these surface antigens and a monocyte marker antigen on fresh cord and adult blood monocytes, macrophages (M phi) derived from monocytes in vitro, human placental (fetal) M phi, from adult women. By FACS analysis, we found less DR on cord blood monocytes (80 +/- 7) than on adult monocytes (96 +/- 1) with greater heterogeneity in density of DR due to a weakly staining subpopulation of cord monocytes. There were markedly fewer DR and DQ positive placen

Malignant granular lymphoproliferation after Epstein-Barr virus infection: partial immunologic reconstitution with interleukin-2.

This report describes a patient who developed a malignant proliferation of granular lymphocytes following Epstein-Barr virus (EBV) infection. For many months, his illness resembled prolonged infectious mononucleosis with persistent fatigue, fever, leukocytosis, and serologic evidence of recent primary EBV infection. After approximately 1 year, however, he developed progressive granular lymphocytosis and extensive lymphocytic infiltration of the bone marrow and liver. Tests for EBV DNA in pre- and postmortem tissue samples using a sensitive DNA hybridization technique were negative. Southern bl

Monoclonal anti-interleukin 2 (15-2) antibody binding to granular layer keratinocytes of human skin.

Among several monoclonal antibodies (moABs) directed against human interleukin 2 (IL-2), the 15-2 moAB raised in our laboratory against unglycosylated recombinant IL-2 (produced in Escherichia coli) cross-reacted with a human skin epitope. This moAB gave a strong staining on the cell-surface membranes of keratinocytes from the granular layer of the epidermis. In addition, the 15-2 moAB stained 15% of epidermal cell suspensions obtained from suction blisters and reacted with cells from the spinous layer in parakeratosis and psoriasis, as well as with spinous epithelioma cells. The arrangement of immunoglobulin, T cell antigen receptor, and interleukin 2 receptor genes in human lymphoid neoplasms.

Ig and T cell antigen receptor genes in their germ line form are separated DNA segments that are joined by recombinations during lymphocyte development. The analysis of Ig and T cell receptor gene arrangements has been of value in the study of lymphoid neoplasms. The identification of T cell receptor gene rearrangements taken in conjunction with studies of Ig gene rearrangements aids in the elucidation of the lineage (T cell or B cell) and the clonality of lymphoid populations of all series. The application of this molecular genetic approach has great potential for complementing conventional m

The interleukin 2 secretion defect in vitro in systemic lupus erythematosus is reversible in rested cultured T cells.

Interleukin 2 (IL 2) secretion in response to mitogenic stimulation in vitro is strongly reduced in circulating T lymphocytes from patients with SLE. It is still not clear how this abnormality relates to the B cell hyperactivity in the disease. Some investigators proposed that an intrinsic T helper cell defect could lead to suppressor cell dysfunction and autoimmunity. Others have found that in fact increased suppressor cell activity can cause IL 2 hyposecretion. In the present study we report that the IL 2 secretion in response to PHA plus PMA by T cells from patients with SLE, which initiall

Comparison of the immunosuppressive effects of cyclosporine, lipid-soluble anesthetics, and calmodulin antagonists. Response to exogenous interleukin 2.

The purpose of these investigations was to compare the immunosuppressive mechanism of cyclosporine (CsA) with those of lipid-soluble local anesthetics and calmodulin antagonists. Chlorpromazine (CPZ) and pentobarbital (PB) both inhibit lymphocyte activation by attenuating sodium and potassium ion potentials. CPZ and N-(6-aminohexyl)-5-chloro-1-naphthalene sulfonamide (W-7) can also block calcium-dependent activation processes by inhibition of calmodulin and protein kinase C. All four compounds were found to suppress human and murine lymphoproliferation to both alloantigen or mitogen in a dose-

Quantitative measurement of human interleukin 2 receptor levels with intact and detergent-solubilized human T-cells.

2A3 monoclonal antibody (gamma 1, kappa) is a novel high-affinity reagent for detecting the human interleukin 2 (IL-2) receptor. The antibody inhibits IL-2 binding to its receptor and is an antagonist of IL-2 action. Detailed analysis of the mechanism of binding of the IgG, (Fab')2 and Fab' of 2A3 antibody shows that the bivalent species cross-link on the cell surface when bound. Measurements of IL-2 receptor expression on digitonin-permeabilized cells suggest that the intracellular pool of receptors is small. The antibody will bind to IL-2 receptors on glutaraldehyde-fixed cells in the presen

Quantitative absorption of interleukin-2 by peripheral blood and synovial fluid lymphocytes from patients with rheumatoid arthritis.

Peripheral blood (PBL) and synovial fluid lymphocytes (SFL) from 18 patients with definite rheumatoid arthritis (RA) and from one patient with Reiter's disease (RD) were examined for their capacity to absorb quantitatively interleukin-2 (IL-2) from a standardized, lectin-free IL-2 source. For comparison normal ConA blasts and PBL from various inflammatory and noninflammatory diseases as well as from healthy control persons were studied. IL-2 activity was quantitated by measuring 3H-thymidine-2-deoxyriboside uptake in the IL-2 dependent murine T cell line CTL6. ConA blasts exhibited a high IL-2

Interleukin 2-mediated events in gamma-interferon production are calcium dependent at more than one site.

Both leukotrienes and dibutyryl cyclic GMP can replace the interleukin 2 (IL 2) requirement for gamma-interferon (gamma-IFN) production. In this study, the Ca dependence of the IL 2 help was demonstrated by blockage of gamma-IFN production by the Ca blocker Mn, and the competitive reversal of this block by Ca. Neither leukotriene C4 nor dibutyryl cyclic GMP could reverse the Mn block, which suggests that arachidonic acid release from phospholipids is not the only Ca-dependent event in IL 2 help for gamma-IFN production. A role for calmodulin or protein kinase C in the IL 2-mediated events was

Abrogation of anti-Pichinde virus cytotoxic T cell memory by cyclophosphamide and restoration by coinfection or interleukin 2.

Previously, we demonstrated that memory cell-mediated immune responses can be generated in Pichinde virus (PV)-primed mice after secondary challenge in vivo with homologous virus. Further, treatment of mice with cyclophosphamide (CY) before primary infection with PV abrogated the generation of H-2-restricted, virus-specific cytotoxic T lymphocytes (CTL), and rechallenge of these mice was followed by neither a primary nor a secondary CTL response. Here, we demonstrate that this CY-induced block in memory anti-PV CTL generation was not due to establishment of a persistent infection. Interestingl

Differences in the production of and/or the response to interleukin-2 by T lymphocytes from patients with the various connective tissue diseases.

We have studied the production of and the response to interleukin-2 (IL-2) by blood T lymphocytes from 83 untreated patients with six connective tissue diseases, each patient with a healthy age/sex matched control. SLE patients had markedly decreased production of IL-2, both when elicited with phytohemagglutinin (PHA) and when promoted by autologous mixed lymphocyte reaction (AMLR). They also had decreased response to IL-2. Conversely, patients with scleroderma had normal production of IL-2 with both stimuli and their lymphocytes responded to IL-2 similarly to, or even better than, controls. P

Interleukin 2 augmentation of natural killer cell activity in homosexual men with acquired immune deficiency syndrome.

Natural killer cell activity of peripheral blood mononuclear cells against K562 myeloid cells was studied in 8 normal heterosexual men, 5 healthy homosexual men, and 11 homosexual men with acquired immune deficiency syndrome. The overall natural killer cell activity was lower in healthy homosexual men and in homosexual men with acquired immune deficiency syndrome. Peripheral blood mononuclear cells from four normal heterosexual men were preincubated overnight in complete medium supplemented with various concentrations of lectin-free interleukin 2 lacking interferon. Cells from these cultures e

Investigation of the correlation between serum levels of interleukin-6 and clinical and paraclinical findings in COVID-19 patients.

BACKGROUND: COVID-19 frequently presents with pneumonia. Impaired immune function, particularly in older adults and immunocompromised individuals, increases susceptibility to infection and contributes to higher morbidity and mortality. Among inflammatory cytokines, Interleukin-6 (IL-6) plays a pivotal role in immune dysregulation and cytokine storm. Therefore, this study aimed to investigate the association between serum IL-6 levels and clinical, laboratory, radiological, and outcome-related findings among hospitalized patients with COVID-19. METHODS: This descriptive-analytical cross-section

Targeting Interleukin-8-Mediated Cellular Crosstalk Reverses Hypertrophic Cardiomyopathy and Cardiac Fibrosis in Noonan Syndrome.

BACKGROUND: Genetic variants in components or regulators of the RAS-mitogen-activated protein kinase (RAS-MAPK) signaling pathway are causative for severe and early-onset hypertrophic cardiomyopathy (HCM) in patients with Noonan syndrome (NS). Despite paracrine communication being considered to play a pivotal role in the etiology of cardiomyopathies, there is a paucity of knowledge about the underlying pathomechanism that leads to the development of HCM and cardiac fibrosis in NS. METHODS: To dissect the impact of noncardiomyocytes in the development of NS, we employed two-dimensional and thr

Evaluation of serum semaphorin-3A and interleukin 6 levels in patients with neovascular age-related macular degeneration.

PURPOSE: To determine whether serum semaphorin 3 A (Sema3A) and interleukin 6 (IL-6) play a role in the common etiopathogenesis of nAMD and nAMD-associated systemic diseases by evaluating these mediators levels. METHODS: This prospective case-control study included 74 patients divided into nAMD (n = 33) and a control group (n = 41). Serum Sema3A and IL-6 levels were analyzed using the enzyme-linked immunosorbent assay. RESULTS: The nAMD group had a statistically higher IL-6 level [5,9 (1-16,9) pg/mL], compared to the control group [1,5 (0,38 - 2,96) pg/mL] (p < 0.001). On the other hand, the
