Artificial Intelligence-Assisted Structural Analysis of Bones with Paget's Disease of Bone and Osteoporosis: Lessons from Mouse Models.

Background/Objectives: Paget's disease of bone (PDB) and osteoporosis are chronic metabolic bone disorders characterized by disrupted bone remodeling and increased skeletal fragility; however, the underlying mechanism of PDB remains poorly understood. Artificial intelligence (AI) has emerged as a transformative tool in medical imaging, enabling automated feature extraction and improved diagnostic classification of skeletal disorders. This study aimed to investigate whether AI could distinguish subtle variations in bone morphology between PDB and osteoporotic bone. Methods: C57BL/6 mice femurs

Top-Down Proteomics of Zebrafish Brain Regions Using Capillary Zone Electrophoresis-Tandem Mass Spectrometry.

Understanding region-specific proteoform profiles in the brain is crucial for deciphering neural function and identifying therapeutic targets. Zebrafish (Danio rerio) is a valuable vertebrate model for neuroscience research due to its substantial conservation of brain structure and function with that of mammals. We present a label-free quantitative top-down proteomics (TDP) analysis of distinct zebrafish brain regions using microdissection and capillary zone electrophoresis-tandem mass spectrometry (CZE-MS/MS). We analyzed four anatomically distinct regions─telencephalon (Tele), combined haben

Drug-induced gastric motility disorders: A disproportionality analysis from the FAERS and CVARD databases.

BACKGROUND: Delayed gastric emptying and gastroesophageal reflux represent critical yet frequently underrecognized complications in hospitalized patients, particularly in the context of polypharmacy. While multiple medication classes have been implicated in disrupting gastrointestinal motility, the comprehensive risk spectrum of individual drugs remains poorly characterized. This study aimed to conduct a comprehensive disproportionality analysis to identify drugs associated with delayed gastric emptying and reflux using large-scale pharmacovigilance data. METHODS: We analyzed adverse event re

Single-cell RNA sequencing reveals compartment-specific immune and tumor cell states in a contiguous spinal metastasis.

BACKGROUND: Metastatic disease commonly exhibits differential treatment responses across anatomical sites, including visceral versus osseous lesions and, within the spine, osseous, epidural, and intradural compartments. Intradural spinal metastases, though rare, portend rapid neurologic deterioration and worse outcomes than epidural disease, yet the biological mechanisms underlying this differential behavior remain poorly understood. METHODS: We report a 61-year-old woman with metastatic ER+/PR+ breast cancer who developed bilateral leg weakness and loss of walking due to severe thoracic spin

Spatial correspondence between altered brain function and normative neurotransmitter density maps in Type 1 diabetes mellitus.

PURPOSE: Type 1 diabetes mellitus (T1DM) is associated with cognitive impairment, yet the underlying neurobiological mechanisms remain poorly understood. While resting-state functional magnetic resonance imaging (rs-fMRI) has revealed aberrant brain activity in T1DM, the spatial correlation between these functional alterations and normative neurotransmitter receptor/transporter distributions has not been systematically explored. METHODS: We enrolled 43 T1DM patients and 52 well-matched healthy controls (HCs). All participants underwent rs-fMRI and comprehensive neuropsychological assessments.

Avacopan is an oral C5a receptor antagonist approved for severe active granulomatosis with polyangiitis and microscopic polyangiitis. However, its real-world safety profile, particularly hepatobiliary safety signals, remains incompletely characterized. We analyzed avacopan-related adverse event reports from FAERS, JADER, and EudraVigilance. Disproportionality analyses were performed using reporting odds ratio and Bayesian Confidence Propagation Neural Network. Subgroup, sensitivity analysis, time-to-onset, Bradford Hill, network pharmacology, molecular docking, machine learning, and SHAP analy

Patient-Reported Feedback Suggests an Alternative Sweet Spot for Deep Brain Stimulation Programming in Essential Tremor.

BACKGROUND: Deep brain stimulation (DBS) of the ventral intermediate nucleus (VIM) and caudal zona incerta is an established therapy for essential tremor (ET). Clinical outcomes depend on precise electrode placement and optimal stimulation parameters. Effective programming must balance tremor suppression with side-effect risk, yet systematic incorporation of patient-reported feedback remains limited. OBJECTIVE: The aim was to assess whether subjective patient feedback, quantified via a Visual Analogue Scale (VAS), can guide DBS programming for effective tremor control. METHODS: In 15 VIM-DBS

Sex differences in brain functional connectivity changes to different energy densities of laser acupuncture at PC6 (Neiguan): a pilot resting-state fMRI study.

Purpose previous studies have suggested that laser acupuncture (LA) may influence autonomic nervous system (ANS) activity and brain functional connectivity (FC); however, the underlying neural mechanisms and potential sex-dependent responses remain to be further investigated. Using resting-state functional magnetic resonance imaging (fMRI), we examined the effects of LA at different energy densities (EDs), hypothesizing that varying dosages elicit differential brain responses influenced by biological sex. Methods we enrolled 60 healthy adults of both sexes aged 18-55 years with a BMI of 18.5-2

Altered Neural Oscillations and Hemispheric Asymmetry in Single-Sided Deafness.

OBJECTIVES: Single-sided deafness (SSD) disrupts binaural auditory input and can lead to cortical reorganization affecting neural oscillatory activity. Whether the side of hearing loss (left versus right) yields distinct electroencephalography (EEG) spectral and lateralization patterns remains unclear. We compared spectral resting-state EEG power and hemispheric asymmetry among patients with left SSD (LSSD), right SSD (RSSD), and normal-hearing controls (NH). DESIGN: Resting-state EEG was recorded from 100 participants, including 32 with LSSD, 31 with RSSD, and 37 NH. A power spectrum was com

Gut-brain axis mechanisms of remote brain dysfunction after traumatic spinal cord injury: immune inflammation, the vagus nerve, and neuroendocrine pathways.

Traumatic spinal cord injury (SCI) has traditionally been regarded as a central nervous system injury mainly confined to the injured segment. However, increasing evidence indicates that SCI can also induce neuroinflammation, cognitive decline, and emotional disorders in remote brain regions, suggesting that its pathological impact is systemic rather than purely local. Remote brain dysfunction after SCI is unlikely to be driven by a single pathway, but may arise from the combined effects of systemic inflammation, autonomic imbalance, neuroendocrine dysregulation, and disruption of intestinal ho

Germanium-doped calcium phosphate ceramics in combination with mesenchymal stem cells derived from the neural crest, obtained from rabbit vibrissae: an experimental model of rabbit jaw regeneration.

BACKGROUND: Jaw bone regeneration remains a clinical challenge in maxillofacial surgery. Bioactive ceramics and neural crest-derived multipotent mesenchymal stem cells (NC-MSCs) show regenerative potential, but their combined effect in repairing jaw defects has not been fully investigated. To our knowledge, no previous studies have evaluated germanium-doped biphasic calcium phosphate ceramics combined with NC-MSCs from vibrissae in a critical-size mandibular defect model. This study aimed to evaluate biphasic calcium phosphate ceramics doped with 1% germanium (BCPdGe 1%) alone versus BCPdGe 1%

Role of the WNT signalling pathway in physiological and pathological blood-brain barrier.

BACKGROUND: The blood-brain barrier (BBB) is essential for maintaining central nervous system (CNS) homeostasis and protecting neural tissue. The wingless-type MMTV integration site family (WNT) signalling pathway has emerged as a key regulator of BBB development, maintenance, and repair. Dysregulation of this pathway is implicated in BBB dysfunction associated with various neurological disorders. METHODS: We conducted a comprehensive review of recent literature integrating data from animal models, human induced pluripotent stem cell (iPSC)-derived BBB systems, and disease-specific mechanisti
