Screening of compounds in ginseng that activate CYP19A1 enzyme promoters for the treatment of postmenopausal osteoporosis using molecular docking and affinity ultrafiltration technology.

The primary etiology of postmenopausal osteoporosis is estrogen deficiency. The enzyme aromatase (CYP19A1) serves as the key rate-limiting enzyme in the conversion of androgens to estrogens. In this study, ginseng was selected as the subject of investigation. The study identified that the 50 % ethanol extract of ginseng exhibited the most potent activity in the determination of CYP19A1 enzyme activity. This extract was subsequently analyzed using UHPLC-QE Orbitrap-MS in conjunction with mass spectrometry molecular network technology. To identify the active components effective against postmeno

Neuro-skeletal crosstalk: Brain-derived 5-HT mediates the bone-protective effects of β-Sitosterol against postmenopausal osteoporosis.

BACKGROUND: Postmenopausal osteoporosis (PMOP) is frequently accompanied by depression, and the underlying neuro-skeletal crosstalk remains unclear. Serotonin and sympathetic nervous system (SNS) activity are implicated in both mood and bone regulation. OBJECTIVE: To investigate whether β-sitosterol (βS) alleviates PMOP-associated depression and bone loss through modulation of central 5-hydroxytryptamine (5-HT) synthesis and SNS activity. DESIGN: An integration of in vivo and in vitro studies using mouse models and cellular assays. METHODS: Ovariectomized (OVX) and 5-HT-deficient mice were

Apelin-13 treats postmenopausal osteoporosis by reducing mitochondrial oxidative phosphorylation and modulating the OPG/RANK/RANKL/IGF-1 pathway.

OBJECTIVE: Postmenopausal osteoporosis (PMOP) is a prevalent metabolic bone disorder characterized by decreased bone mineral density and an increased risk of fractures. This study aims to investigate the molecular mechanisms underlying the therapeutic effects of Apelin-13 in PMOP. METHODS: A PMOP model was established using mouse embryonic pre-osteoblast MC3T3-E1 cells treated with dexamethasone (DEX). A variety of experimental techniques-including MTT assays, Western blotting, flow cytometry, immunofluorescence staining, mitochondrial membrane potential assays, and ELISA-were employed to elu

Risk factors for refracture after vertebral augmentation in postmenopausal women with osteoporotic vertebral compression fractures: a systematic review and meta-analysis.

BACKGROUND: Postoperative refracture is a significant complication that may arise after percutaneous vertebral augmentation (PVA). While several studies have explored the risk factors associated with refracture following PVA for osteoporotic vertebral compression fractures (OVCF), there is currently no meta-analysis specifically focusing on postmenopausal women. QUESTIONS/PURPOSES: To assess the risk factors for refracture after PVA in postmenopausal women with OVCF and to explore the underlying mechanisms. METHODS: We systematically searched Cochrane Library, PubMed, EMBASE, and Web of Scienc

Diosgenin activates ERα-dependent PI3K/AKT/GSK3β signaling to enhance brain-derived serotonin and alleviate postmenopausal bone loss.

INTRODUCTION: Postmenopausal osteoporosis (PMOP) stems from estrogen deficiency, which leads to reduced central serotonin (5-HT) levels, induces depressive-like behaviors, enhances sympathetic nervous system (SNS) activity, and disrupts bone homeostasis. However, the underlying mechanisms and potential therapeutic strategies targeting this pathway remain incompletely understood. OBJECTIVE: To test whether phytoestrogens diosgenin (DG) can inhibit bone loss in PMOP by restoring the 5-HT-SNS-bone metabolism circuit. METHODS: Ovariectomized (OVX) mice and a pharmacological 5-HT-depletion model

Anti-resorptive and anabolic therapies improve Falls Risk Assessment Score (FRAS) in postmenopausal women with type 2 diabetes mellitus.

UNLABELLED: Zoledronate, denosumab and teriparatide improve Falls Risk Assessment Score (FRAS) and reduce the number of participants experiencing falls over the last 12 months when administered over a period of 72 weeks to postmenopausal women with type 2 diabetes mellitus at a high risk of fragility fractures. PURPOSE: Type 2 diabetes mellitus (T2D) increases the risk of falls, which also exacerbates the risk of fragility fractures. We aimed at exploring the effects of anti-osteoporotic therapies (zoledronate, denosumab or teriparatide) on the risk of falls in postmenopausal women with T2D.

Comparative outcomes of sphincteroplasty and sacral neuromodulation in postmenopausal women with late-onset fecal incontinence following obstetric trauma: a retrospective study.

Fecal incontinence (FI) is a debilitating condition that commonly affects postmenopausal women, particularly those with a history of obstetric trauma. To date, there are few studies directly comparing SP and SNM, which present many discrepancies in the inclusion criteria, such as age, etiology, and elapsed time from the onset of the condition. This retrospective study aims to compare the clinical outcomes of the two procedures most widely performed in the management of FI: sphincteroplasty and sacral neuromodulation (SNM). The inclusion criteria were established to ensure a representative samp

Case Report: Serous borderline ovarian tumor and extensive abdominopelvic endometriosis mimicking advanced epithelial ovarian cancer in a postmenopausal patient.

The lifetime risk of ovarian cancer is 1 in 75, and it is often diagnosed at an advanced stage due to the lack of effective screening. Epithelial ovarian cancer (EOC) is the most common type of ovarian cancer in older patients, with borderline ovarian tumors (BOTs) accounting for 15% of EOC cases. BOTs are usually of low malignant potential. Since platinum-based chemotherapy has not been shown to improve survival rates, cytoreductive surgery (CRS) is recommended as a treatment option. Additionally, endometriosis affects 10.5% of patients with EOC. It often progresses to clear cell and endometr

Evaluation of romosozumab's effects on bone marrow adiposity in postmenopausal osteoporotic women: results from the FRAME bone biopsy sub-study.

Romosozumab, a humanized monoclonal antibody that binds and inhibits sclerostin, produces a marked increase in bone formation with a concomitant decreased bone resorption. This transient rise in bone formation in the first 2 months of treatment is mainly due to an increased modeling-based bone formation. This requires the recruitment and differentiation of osteoblasts, one possibility being a preferential switch in commitment of precursors to osteoblasts over adipocytes. The purpose of this study was to analyze the marrow adiposity in transiliac bone biopsies at months 2 or 12 from the FRAME b

Acute impact of inorganic nitrate supplementation after ischemia and during small muscle mass exercise in postmenopausal females: A pilot study.

Menopause is associated with reduced endothelial-dependent vasodilation and increased cardiovascular disease (CVD) risk. Dietary nitrate, a non-pharmacological approach, may increase vasodilatory capacity consequentially reducing CVD risk. We investigated macro- and microvascular function after acute nitrate supplementation in postmenopausal females (PMF). Vascular function was studied with flow-mediated vasodilation (FMD) and near-infrared post occlusive reactive hyperemia (PORH). Incremental handgrip exercise was performed to investigate blood flow and tissue oxygenation. We hypothesized acu

Psychological Symptom Cluster Severity Profiles Are Associated With Higher Levels of Stress and Worse Quality of Life in Postmenopausal Women With Breast Cancer.

BACKGROUND: Although common, the severity of the psychological symptom cluster (PSC) (ie, co-occurrence of anxiety, depression, fatigue, sleep disturbance) varies among women with breast cancer. In addition, the impact of various types of stress and other risk factors on its severity are understudied, and the influence of the PSC on quality of life (QOL) outcomes remains poorly characterized. OBJECTIVE: To identify subgroups of postmenopausal women with breast cancer with distinct PSC severity profiles following surgery and evaluate whether these subgroups differed in terms of demographic and
