Etiology of posterior subcapsular cataracts based on a review of risk factors including aging, diabetes, and ionizing radiation.

PURPOSE: Since the exact development of posterior subcapsular cataracts (PSCs) is poorly understood, we review various risk factors and propose a two-stage etiology for PSCs. METHODS: The biological mechanisms associated with age-related cataracts (primarily nuclear cataracts, cortical cataracts and PSCs) were reviewed in relation to selected risk factors that induce PSCs (including atopy, diabetes, hypoparathyroidism, myopia, retinitis, solar radiation, steroid use, uveitis, vitrectomy and ionizing radiation). We particularly focused on ionizing radiation, as this is known to be a risk facto

Enhancement of Uterine Cancer Development after Oocyte Depletion by Juvenile Exposure
to Gamma Radiation in Rats: - A Comparative Study on Sensitivity of Damage to
Female Reproductive Organs by Radiation
between Juvenile and Adulthood Using a Rat Model.

To clarify sensitivities of juvenile exposure to radiation on uterine carcinogenesis, female Donryu rats, a high yield strain of uterine corpus cancer, were exposed to 0.2 and 1.0 Gy of gamma radiation at postnatal day 14. Sequential changes in their reproductive organs and hematology, and the effects on uterine tumor development were compared to those in adult rats exposed to the same doses. Half number of the rats in each group was treated with N-ethyl-N'-nitro-N-nitrosoguanidine (ENNG) after the radiation to accelerate the development of uterine cancer. Severe apoptosis and depletion of ooc

Targeted Metabolomics of Nonhuman Primate Serum after Exposure to Ionizing Radiation: Potential Tools for High-throughput Biodosimetry.

There is a need for research to rapidly determine an individual's absorbed dose and its potential health effects after a potential radiological or nuclear event that could expose large portions of a population to ionizing radiation (IR). Studies on biomarker identification after radiation exposure could aid in biodosimetry, identifying individual dose absorbed, as well as biologic response, and administering immediate and proper medical care. Metabolomics on easily accessible biofluids is an emerging field with potential for high-throughput biodosimetry. While tremendous effort has been put in

SU-E-I-47: Comparison of Risks for Two Medical Imaging Procedures.

PURPOSE: Radiologists may need to decide which type of image procedure is most appropriate for a particular patient. One factor relevant in making this decision is the relative risk of secondary cancers due to each relevant procedure. Differences in the risk posed by each method are not just due to the total radiation dose imparted by each procedure, but also the distribution of absorbed dose across various organs in the imaging procedure. Two imaging procedures with the same total radiation dose may pose different risks of differential sensitivity to radiation across organs. METHODS: New met

Loss of Nrf2 accelerates ionizing radiation-induced bone loss by upregulating RANKL.

Radiation therapy is an integral part of treatment for cancer patients; however, major side effects of this modality include aberrant bone remodeling and bone loss. Ionizing radiation (IR) is a major external factor that contributes to a significant increase in oxidative stress such as reactive oxygen species (ROS), has been implicated in osteoporotic phenotypes, and has been implicated in osteoporotic phenotypes, bone loss, and fracture risk. One of the major cellular defenses against heightened oxidative stress is mediated by nuclear factor (erythroid-derived 2)-like 2 (Nrf2), a master trans

Neovascular glaucoma after stereotactic radiotherapy for juxtapapillary choroidal melanoma: histopathologic and dosimetric findings.

PURPOSE: Enucleation after stereotactic radiotherapy (SRT) for juxtapapillary choroidal melanoma may be required because of tumor progression (TP) or the development of intractable radiation-induced neovascular glaucoma (NVG). We compare pathologic changes and dosimetric findings in those eyes enucleated secondary to NVG as opposed to TP to better understand potential mechanisms. METHODS AND MATERIALS: Patients with juxtapapillary choroidal melanoma treated with SRT (70 Gy in 5 fractions, alternate days over a total of 10 days) at the Princess Margaret Hospital, Toronto, Ontario, Canada, who

Human ghrelin ameliorates organ injury and improves survival after radiation injury combined with severe sepsis.

In the terrorist radiation exposure scenario, radiation victims are likely to suffer from additional injuries such as sepsis. Our previous studies have shown that ghrelin is protective in sepsis. However, it remains unknown whether ghrelin ameliorates sepsis-induced organ injury and mortality after radiation exposure. The purpose of this study is to determine whether human ghrelin attenuates organ injury and improves survival in a rat model of radiation combined injury (RCI) and, if so, the potential mechanism responsible for the benefit. To study this, adult male rats were exposed to 5-Gy who

Genotoxic and cytotoxic effects of 60Co gamma-rays and 90Sr/90Y beta-rays on Chinese hamster ovary cells (CHO-K1).

Among various types of ionizing radiation, the beta emitter radionuclides are involved in many sectors of human activity, such as nuclear medicine, nuclear industries and biomedicine, with a consequently increased risk of accidental, occupational or therapeutic exposure. Despite their recognized importance, there is little information about the effect of beta particles at the cellular level when compared to other types of ionizing radiation. Thus, the objective of the present study was to evaluate the genotoxic and cytotoxic effects of (90)Sr/(90)Y-a pure, highly energetic beta source-on Chine

Pharmacologic approaches to protection against radiation-induced lethality and other damage.

Studies on mechanisms of radioprotection are leading to a more rational use of protectors for different applications. In considering the feasibility of radioprotectors that act through various mechanisms, it is necessary to distinguish the application needed, e.g., protection against accidental external or internal exposures, acute high-dose radiation injury or low doses over a long period, high-LET radiation exposures during space flight, and protection of normal tissues of cancer patients who are undergoing therapy. Protectors generally are classified as either sulfhydryl compounds, other an

Local and systemic consequences of acute, low-dose ultraviolet B radiation are mediated by different immune regulatory mechanisms.

Acute, low-dose ultraviolet B (UVB) radiation alters the local skin site such that epicutaneous application of hapten fails to induce contact hypersensitivity (CH), but induces tolerance in UVB-susceptible mice. Although the inability of irradiated skin to support CH induction may be a strictly local effect, there may also be systemic immune consequences of UVB radiation delivered in this manner. To examine this matter, abdominal skin of C57BL/6 mice was exposed to acute, low-dose UVB radiation. Dinitrofluorobenzene was immediately painted directly on the irradiated site, or at a distant (unir

Schneeberg lung disease and uranium mining in the Saxon Ore Mountains (Erzgebirge).

The so-called Schneeberg lung disease is a form of bronchial or alveolar carcinoma caused by the effects of the radioactive gas radon and of its radioactive short half-life daughter products. This type of radiation-induced occupational cancer is the most common and the most important radiation injury among workers occupationally exposed to ionizing radiation. There have been many deaths from lung cancer, especially in the Soviet uranium mines in the Erzgebirge of Saxony in the former German Democratic Republic. The history of disease in these miners extends over five centuries; the first obser

Photocarcinogenesis, skin cancer, and aging.

Nonmelanoma skin cancers, like most malignancies, increase in incidence with increasing age. However, in general they are not due to the aging process but are primarily due to solar radiation. Clinically, squamous cell carcinomas and basal cell epitheliomas are the most common cancers that occur in the Caucasian population in the United States. The role of radiation from the sun was suggested by a number of astute clinical observations reported around 1900 and subsequently has been established by epidemiologic and experimental studies. Action spectrum evaluations indicate that the ultraviolet

Nintedanib enhances tumor cell radiosensitivity by promoting ferroptosis and modulating the ATF4/SLC7A11/GSH axis.

OBJECTIVE: Tumor cell radio-resistance and radiation-induced fibrosis of normal tissues hinder the efficacy of radiotherapy. Nintedanib, a promising therapeutic agent for radiation-induced pulmonary fibrosis and solid tumors, has yet to be investigated in combination with radiotherapy. This study aimed to evaluate the antitumor efficacy of nintedanib in conjunction with radiotherapy. METHODS: Tumor-bearing models were utilized to assess the antitumor effects and safety of treatment with nintedanib and radiotherapy in vivo. JS-K enhances renal cell carcinoma radiosensitivity through the Wnt/β-catenin signaling pathway and inhibits DNA damage repair.

The study investigated the effect of the glutathione S-transferase-activated nitric oxide donor JS-K [O2-(2,4-dinitrophenyl)1-[(4-ethoxycarbonyl)piperazin-1-yl]diazen-1-ium-1,2-diolate] to enhance the sensitivity of renal cancer cells to radiation therapy. JS-K has shown promising anti-tumor effects in various types of tumors; however, research on its role in cancer radioresistance is limited. The traditional concept is that renal cell carcinoma (RCC) has inherent resistance to radiation. Therefore, this study aimed to explore whether JS-K could enhance the sensitivity of renal cancer cells to

CD8+ T cells in patients with hypopharyngeal squamous cell carcinoma are susceptible to radiation-induced damage.

Radiotherapy (RT) is a commonly used clinical management for hypopharyngeal squamous cell carcinoma (HPSCC), which represents the most unfavorable prognosis among all subtypes of head and neck squamous cell carcinoma. However, radiation may cause lymphopenia, a significantly adverse event with detrimental prognostic implications for patients. While CD8+ T cells are vital in tumor immunity, the specific effects of RT on CD8+ T cells as well as the underlying mechanisms have not been clearly elucidated. Here we found that subpopulations of peripheral T lymphocytes exhibited differential profiles

Potential of Lactoferrin Against the Radiation-Induced Brain Injury.

The purpose of this work was to study the effects of lactoferrin (Lf) on acute (days 3 and 15) and early-delayed (day 30) changes in the dentate gyrus of mouse hippocampus caused by whole-body gamma-irradiation. Male C57BL/6 mice received Lf (4 mg per mouse, i.p. injection) immediately after whole-body gamma-irradiation at a dose of 7.5 Gy from a 60Co source. The effect of Lf on mouse behavior was evaluated using "Open field" and "Elevated plus-maze" tests. The proportion of cells with DNA replication was determined by 5-ethynyl-2'-deoxyuridine incorporation (thymidine analog) and detected by

Quantifying the Risk of Technology-Driven Health Disparities in Radiation Oncology.

PURPOSE: New technologies are continuously emerging in radiation oncology. Inherent technological limitations can result in health care disparities in vulnerable patient populations. These limitations must be considered for existing and new technologies in the clinic to provide equitable care. MATERIALS AND METHODS: We created a health disparity risk assessment metric inspired by failure mode and effects analysis. We provide sample patient populations and their potential associated disparities, guidelines for clinics and vendors, and example applications of the methodology. RESULTS: A dispar
