[Expert consensus on orbital MRI examination and diagnostic evaluation in thyroid eye disease (2026 edition)].

Thyroid eye disease (TED) is the most common orbital disease in adults, with substantial impacts on patients' appearance, visual function, and quality of life. Magnetic resonance imaging (MRI) demonstrates distinct advantages in depicting characteristic changes in orbital soft tissue structures and serves as one of the primary imaging tools for evaluating disease extent and characterizing pathological features, playing an important role in differential diagnosis, disease classification, and activity assessment of TED. However, current orbital MRI examination protocols for TED lack unified stan

Astrocytic Neurovascular Signalling Dysfunction in Glaucoma: Neurochemical Mechanisms and Translational Implications.

Glaucoma is increasingly conceptualized as a chronic neurodegenerative disorder in which retinal ganglion cell loss cannot be fully attributed to intraocular pressure dependent mechanisms alone. Mounting evidence implicates dysfunction of the neurovascular unit, with astrocytes playing a central role through their neurochemical regulation of vascular tone, metabolic coupling, and redox homeostasis. Under physiological conditions, astrocytes mediate neurovascular coupling via calcium dependent signalling and controlled release of vasoactive and metabolic mediators, ensuring alignment between ne

The Role of Neutrophils and NETosis in Diseases: The Implications for Therapy.

Neutrophils are the most abundant innate immune cells and are pivotal first responders in host defense, playing an important role in maintaining body homeostasis and regulating pathological conditions. Growing evidence indicates that neutrophils exhibit functional heterogeneity and participate in immune regulation through processes such as neutrophil extracellular trap formation (NETosis). These findings have expanded the traditional view of neutrophils as short-lived effector cells. Nevertheless, how distinct neutrophil states are temporally coordinated during disease development, and how thi

Mechanisms and clinical research progress of IL-6-mediated crosstalk between the alveolar microenvironment and the immune system in patients with severe pneumonia undergoing mechanical ventilation.

Severe pneumonia is a common critical illness encountered in clinical practice. Although mechanical ventilation is essential for life support, it can paradoxically result in ventilator-induced lung injury and exacerbate pulmonary inflammation. Interleukin-6 (IL-6) is a key pleiotropic cytokine with both proinflammatory and immunometabolic effects, playing a crucial role in regulating the alveolar microenvironment and systemic immune responses in patients with severe pneumonia. Current evidence suggests that IL-6 overexpression is closely associated with increased alveolar inflammation, immune

Robust Heterojunction Nanoconstructs Enable Precise Decipherment of Aqueous Humor Metabolic Profiles in Multiple Retinal Diseases.

Retinal diseases (RD), which often cause irreversible vision impairment and lack effective treatments, necessitate more attention than common ocular conditions like cataract (CA), and aqueous humor metabolic analysis has proven an invaluable tool for their investigation. In this work, a robust, high-performance heterojunction nanoconstruct (MZ-D) with cost-effectiveness (∼2.7 yuan per gram), streamlined two-step synthesis, high reproducibility, was developed. Combined with high-throughput mass spectrometry and machine learning, aqueous humor metabolic profiles were extracted and analyzed for m

Melatonin Promotes Human Hair Follicle Stem Cell Proliferation by Suppressing 5-AR: Implications for Treating Androgenetic Alopecia.

OBJECTIVE: This study explores the influence of melatonin and SRD5A2 on the proliferation and migration of human hair follicle stem cells (hHFSCs), which are vital in the treatment of Androgenic alopecia (AGA), wound healing, and regenerative medicine. Melatonin interacts with various stem cells, including hHFSCs, in multiple ways. METHODS: In this study, SRD5A2-silenced hHFSCs were established using shRNA and subsequently cultured with melatonin. The effects on cell proliferation and migration were assessed using Cell Counting Kit-8, EdU-555, and the Transwell Migration Assay, while stemness

A prokaryotic-like cardiolipin synthase is essential to maintain mitochondrial function, lipid homeostasis, and survival of the blood stage malaria parasite Plasmodium falciparum.

Mitochondrion is essential for the propagation of the malaria parasites in the human host. The structure, biogenesis, and function of mitochondria depend on the synthesis of a key mitochondrial membrane-specific phospholipid, cardiolipin (CL), which is typically synthesized by cardiolipin synthase (CLS). Unlike most eukaryotic cell models, the synthesis of CL in the malaria parasite remains unresolved. Here, we show that Plasmodium falciparum harbors a prokaryotic-like CLS (PfCLS), which is functional in the parasite mitochondrion. Inducible knockdown of PfCLS disrupts parasite development and

Single-Chain Anti-IL-1β Antibody Carried by Outer Membrane Vesicles of Bacteroides fragilis Alleviates Tubular Inflammation in Chronic Kidney Disease.

Microinflammation is a key driver of chronic kidney disease (CKD) progression, with interleukin-1β (IL-1β) playing a pivotal role. However, current anti-IL-1β antibody therapies face critical limitations, such as systemic side effects and substantial production costs, which hinder their therapeutic efficacy and clinical translation for CKD intervention. To address this, we developed Bacteroides fragilis-derived outer membrane vesicles (OMVs) encapsulating anti-IL-1β single-chain variable fragment (scFv) and conjugated with kidney-targeting peptides (KKEEE)3K. Engineered OMV-(KKEEE)3K-scFv's sa

CHTOP regulates the NRF2/HO-1 axis to modulate chemoresistance in colorectal cancer.

Chemotherapy resistance remains a significant challenge in colorectal cancer (CRC) treatment, with disrupted redox balance playing a central role. Here we identify CHTOP as a key regulator of oxidative stress and chemoresistance in CRC. Mechanistically, CHTOP promotes NRF2 transcriptional activity by recruiting the SENP3-containing 5FMC complex to deSUMOylate NRF2, thereby sustaining HO-1 expression and redox balance. Notably, CHTOP expression itself is tightly controlled by a feedback mechanism. The p52 isoform of PSIP1 inhibits CHTOP expression by interfering with HNRNPH1-mediated splicing,

Enhancer of Zeste Homolog 2 (EZH2): From Glioblastoma Biology to Potential Epigenetic Therapy.

Epigenetic dysregulation is a hallmark of glioblastoma (GBM) pathogenesis, with histone methylation playing a central role in chromatin remodeling and gene expression regulation. Enhancer of zeste homolog 2 (EZH2) is a key epigenetic regulator responsible for histone methylation. It primarily functions as a transcriptional repressor and regulates signaling pathways associated with tumor progression. Importantly, EZH2 is frequently recruited to long non-coding RNA (lncRNA) scaffolds, including HOTAIR and AGAP2-AS1, thereby enabling coordinated gene silencing by establishing repressive chromatin

Autophagy-dependent sensitization effects of PARP inhibitors on recurrent nasopharyngeal carcinoma treated with carbon ion and photon irradiation.

Tumor radioresistance and severe toxicity make reirradiation for recurrent nasopharyngeal carcinoma (NPC) a significant clinical challenge. This study aims to investigate the ability of the poly(ADP-ribose) polymerase (PARP) inhibitor olaparib to sensitize recurrent NPC cells irradiated with photon or carbon ion (C-ion), and to explore the underlying mechanism of the synergistic promotion of cell death by olaparib and ionizing radiation. The results show that olaparib has significant X-ray and C-ion radiosensitization effects on recurrent NPC cells and the associated HK-RR photon-resistant mod

SLC6A14-mediated glutamine promotes SYTL4-CXCL8 axis activation to drive gemcitabine resistance and immune evasion in pancreatic cancer.

Chemoresistance remains a major challenge in pancreatic ductal adenocarcinoma (PDAC). Glutamine sustains drug resistance and shapes the immunosuppressive tumor microenvironment; however, the underlying mechanisms remain unclear. Identifying key regulators that drive both gemcitabine resistance and immune evasion is crucial for improving theapeutic outcomes in PDAC. Here we identified solute-carrier family 6 member 14 (SLC6A14) as the central regulator of glutamine metabolism that drives gemcitabine resistance. SLC6A14-mediated glutamine metabolism facilitated α-ketoglutarate production, activa

Glycosylphosphatidylinositol biosynthesis functions as a conserved host defense pathway against coronaviruses via regulation of LY6E.

Coronaviruses, including SARS-CoV-2, rely on host factors for their replication and pathogenesis, while hosts deploy defense mechanisms to counteract viral infections. Although numerous host proviral factors have been identified, the landscape of host restriction factors and their underlying mechanisms remain less explored. Here, we conducted genome-wide CRISPR knockout screens using three distinct coronaviruses-SARS-CoV-2, HCoV-OC43 (a common cold human virus from the genus Betacoronavirus) and porcine epidemic diarrhea virus (Alphacoronavirus) to identify conserved host restriction factors.

FAcTs on fire: Exploring thermogenesis.

Thermoregulation is a fundamental biological process that allows birds and mammals to maintain a stable internal temperature despite environmental fluctuations, a mechanism shaped by millions of years of evolution. Non-shivering thermogenesis (NST), primarily driven by brown adipose tissue (BAT), plays a central role in thermoregulation by not only helping maintain energy homeostasis but also influencing broader metabolic and physiological processes. Recent research has revealed that BAT thermogenesis is regulated by peripheral hormones and at a central level, with key hypothalamic energy-sens

Pain integration of multimodal noxious stimulation based on the perceptual instruction.

Individuals encounter various somatosensory inputs in their daily lives, often in multiple forms at once. When exposed to noxious stimuli, the intensity of these inputs can lead to a complex experience of pain. However, the mechanisms by which these harmful stimuli combine into a unified pain experience remain unclear. Understanding this process is essential for comprehending pain perception in challenging situations. Here, we investigated pain integration through four experiments involving multimodal noxious stimuli-heat, pressure, and electrical inputs-applied to the feet, instructed by perc

Advances in essential oils for metabolic diseases.

Metabolic diseases are a group of complex disorders caused by abnormal metabolic processes, such as diabetes, obesity, and non-alcoholic fatty liver disease. Long-term uncontrolled metabolic diseases will significantly increase the risk of cardiovascular disease, renal damage, and neurologic complications, leading to shorter survival and reduced quality of life for patients. While conventional treatments rely on pharmacological interventions, essential oils have shown significant therapeutic potential as a natural treatment modality. Essential Oils are concentrated aromatic substances extracte
