Utilization of Ultrasound Guided Tissue-directed Cellular Implantation for the Establishment of Biologically Relevant Metastatic Tumor Xenografts.

Preclinical testing of anticancer therapies relies on relevant xenograft models that mimic the innate tendencies of cancer. Advantages of st … ment. In comparison, orthotopic xenograft models at native tumor sites have been shown to mimic the tumor microenvironment and replicate important disease characteristics such as distant metastatic spread. These models often require tedious surgical procedures with prolonged anesthetic time and recovery periods. To address this, cancer researchers have recently utilized …

Breaking through to the Other Side: Microenvironment Contributions to DCIS Initiation and Progression.

Refinements in early detection, surgical and radiation therapy, and hormone receptor-targeted treatments have improved the survival rates for breast cancer patients. However, the ability to reliably identify which non-invasive lesions and localized tumors have the ability to progress and/or metastasize remains a major unmet need in the field. The current diagnostic and therapeutic strategies focus on intrinsic alterations within carcinoma cells that are closely associated with proliferation. However, substantial accumulating evidence has indicated that permissive changes in the stromal tissues

Understanding the epigenetic regulation of tumours and their microenvironments: opportunities and problems for epigenetic therapy.

The tumour microenvironment plays an instrumental role in cancer development, progression and treatment response/resistance. Accumulating evidence is underscoring the fundamental importance of epigenetic regulation in tumour immune evasion. Following many pioneering discoveries demonstrating malignant transformation through epigenetic anomalies ('epimutations'), there is also a growing emphasis on elucidating aberrant epigenetic mechanisms that reprogramme the milieu of tumour-associated immune and stromal cells towards an immunosuppressive state. Pharmacological inhibition of DNA methylation

Acidic extracellular pH neutralizes the autophagy-inhibiting activity of chloroquine: implications for cancer therapies.

Acidic pH is an important feature of tumor microenvironment and a major determinant of tumor progression. We reported that cancer cells upregulate autophagy as a survival mechanism to acidic stress. Inhibition of autophagy by administration of chloroquine (CQ) in combination anticancer therapies is currently evaluated in clinical trials. We observed in 3 different human cancer cell lines cultured at acidic pH that autophagic flux is not blocked by CQ. This was consistent with a complete resistance to CQ toxicity in cells cultured in acidic conditions. Conversely, the autophagy-inhibiting activ…

Before the seeds arrive: pre-metastatic niche orchestration in bladder cancer.

Metastatic relapse remains a major cause of mortality in bladder cancer (BCa), yet the biological processes that prepare distant organs for tumor colonization before overt metastasis remain incompletely understood. The pre-metastatic niche (PMN) provides a framework for viewing metastasis as a systemic process in which tumor-derived signals reshape distant immune, stromal, vascular, lymphatic, and metabolic environments before cancer-cell arrival. In BCa, some of the strongest disease-specific evidence implicates extracellular vesicle (EV)-mediated communication, including effects on lymphangi

Ablation-immunotherapy synergy in cancer: mechanisms of the abscopal effect, emerging clinical applications, and translational roadmaps for next-generation onco-immunology.

Tumor ablation is rapidly being appreciated as not only a locoregional cytoreductive tool but also an important modulator of antitumor immunity. Many ablative methods (including, but not limited to, radiofrequency ablation, microwave ablation, cryoablation, high-intensity focused ultrasound, and photothermal and photodynamic methods) have been shown to induce immunogenic cell death, release tumor-associated antigens, and produce danger-associated molecular signals that can remodel the tumor microenvironment. While such effects can trigger innate and adaptive immune responses, they are usually …

From fermentation to pharmabiotics: engineering kefir-derived Lactobacillus for targeted intratumoral immunotherapy in colorectal Cancer.

Colorectal cancer (CRC) is a leading cause of cancer-related death, and most patients benefit little from immune checkpoint inhibitors (ICIs) because microsatellite-stable (MSS) tumors are largely resistant to them. In this context, this review aims to address a fermentation-to-pharmabiotic pipeline to convert Lactobacillus strains in kefir into engineered Live Biotherapeutic Products (LBPs). Scientists have discovered an effective way to isolate beneficial bacteria from kefir, which can be used to develop new medical treatments through laboratory-controlled engineered LBP. Metabolism of myeloid cells in brain tumors.

Primary and recurrent brain tumors are aggressive malignancies with high mortality rates and limited treatment options. Glioblastoma (GBM) in particular are largely refractory to immunotherapies despite harboring a significant proportion of immune cells in the tumor microenvironment (TME). Increasing evidence suggests that the immunosuppressive TME in brain tumors is driven by functional and metabolic reprogramming of resident and infiltrating myeloid cells. Here, we examine the determinants of immunometabolic landscape in brain tumors and distinct features of heterogeneous myeloid cell popula…

Nanomedicine for colorectal cancer therapy by remodeling the immune microenvironment.

Colorectal cancer (CRC), a malignant tumor with high incidence and mortality rates, poses a significant threat to human health. Although immunotherapeutic strategies such as immune checkpoint inhibitors (ICIs) have shown promising progress, limited efficacy, autoimmune side effects, systemic toxicity, and drug resistance remain major challenges in CRC immunotherapy. The complex tumor immune microenvironment (TIME), shaped by long-term interactions between cancer cells and microenvironmental components, is a key factor contributing to the poor response of most CRC patients to immunotherapy. Computational identification of migrating T cells in spatial transcriptomics data.

T cells are the central players in antitumor immunity, and effective tumor killing depends on their ability to infiltrate into the tumor microenvironment (TME) while maintaining normal cytotoxicity. However, late-stage tumors develop immunosuppressive mechanisms that impede T cell movement and induce exhaustion. Investigating T cell migration in human tumors in vivo could provide insights into tumor immune escape, although it remains a challenging task. In this study, we developed ReMiTT, a computational method that leverages spatial transcriptomics data to track T cell migration patterns with…

Immune cell delivery platforms for tumor therapy: From empirical approaches to AI-integrated frameworks.

The heterogeneity and immunosuppressive characteristics of the tumor microenvironment present significant challenges to traditional treatment strategies, including inadequate targeting and variable drug resistance. In recent years, immune cell-based delivery systems have emerged, leveraging the innate homing capabilities of immune cells alongside advanced engineering techniques to offer novel approaches for precise targeting and the enhancement of anti-tumor immune responses. However, the rational design and optimization of such complex biological systems remain empirically driven and resource…

The Gut-Brain-Immune Axis in Glioma: Emerging Mechanisms and Therapeutic Opportunities.

Gliomas, particularly glioblastomas, represent some of the most treatment-resistant cancers due to their profoundly immunosuppressive tumor microenvironment (TME) and the restrictive nature of the blood–brain barrier. While recent advances in immunotherapy have transformed the treatment landscape for peripheral tumors, success in gliomas remains limited. Emerging evidence suggests that the gut microbiota—through the gut-brain-immune axis—may play a significant role in shaping systemic and central immune responses. Gut-derived microbial metabolites, immune cell modulation, and neuro-immune sign…

Vaginal Microbiota and Ovarian Cancer: A New Frontier in Immunomodulation and Diagnosis.

Ovarian cancer remains one of the deadliest gynaecological cancers due to its often-late diagnosis and the absence of specific symptoms in the early stages. Beyond genetic alterations, the tumour microenvironment, including the vaginal microbiota, plays a decisive role in tumour progression. Recent studies have highlighted the involvement of microbial imbalance 'dysbiosis' in ovarian carcinogenesis, particularly through interactions with the immune system and the modulation of local inflammatory pathways such as Th17-related pathways. In a state of 'eubiosis,' the vaginal microbiota, dominated

Neutrophil extracellular traps in colorectal cancer: a paradoxical nexus of tumor defense and disease progression.

Neutrophil extracellular traps (NETs) are extracellular networks of decondensed chromatin and granular proteins generated by neutrophils in response to activating stimuli. Growing evidence indicates that NETs participate in colorectal cancer (CRC) development through their interactions with chronic inflammation, immune regulation, coagulation, and tumor progression. However, their biological effects vary across disease settings, and the regulatory mechanisms specific to CRC have not been fully clarified. This review examines the roles of NETs at different stages of CRC, including the transitio

Spatiotemporal dynamics of tumor-associated neutrophils: bridging the gap between cancer progression and immunotherapy.

Neutrophils, traditionally regarded as short-lived first responders of innate immunity, have emerged as pivotal regulators within the tumor microenvironment (TME). Recent advances reveal that tumor-associated neutrophils (TANs) and neutrophil extracellular traps (NETs) exhibit remarkable spatiotemporal heterogeneity, with their phenotypes and functions dynamically evolving across tumor developmental stages and anatomical niches. TANs and NETs display dual and context-dependent roles: they can promote tumor progression via immune suppression, angiogenesis, extracellular matrix remodeling, and m…

The non-bacterial oncobiome: the role of the mycobiome and virome in tumor plasticity.

Tumor plasticity, the capacity of malignant cells to undergo reversible phenotypic switching, is a fundamental driver of lineage diversion and therapeutic resistance. While the bacterial microbiome is a recognized modulator of the tumor microenvironment (TME), the non-bacterial oncobiome, comprising the mycobiome (fungi) and virome (viruses), represents a critical but under-explored frontier in cellular adaptability. This review synthesizes current evidence regarding the mechanistic contributions of fungal and viral constituents to tumor plasticity and characterizes the molecular cross-talk th…
