Effect of replacing main-chain ureas with thiourea and guanidinium surrogates on the bactericidal activity of membrane active oligourea foldamers.

Membrane-active foldamers have recently emerged as potential mimics of antimicrobial peptides (AMPs). Amphiphilic cationic helical N,N'-linked oligoureas are one such class of AMP mimics with activities in vitro against a broad range of bacteria including Bacillus anthracis, a Gram-positive sporulating bacillus and causing agent of anthrax. Here we have used site-selective chemical modifications of the oligourea backbone to gain additional insight into the relationship between structure and function and modulate anthracidal activity. A series of analogues in which urea linkages at selected pos…

Roles of a 20 kDa protein associated with a carbocycle-forming enzyme involved in aminoglycoside biosynthesis in primary and secondary metabolism.

2-Deoxy-scyllo-inosose (DOI) synthase participates in the biosynthesis of 2-deoxystreptamine (DOS)-containing aminoglycoside antibiotics. The enzyme is expected to be of industrial use, because it converts a sustainable resource (glucose 6-phosphate) into carbocycle (DOI), which easily aromatizes to yield catechol. In the present study, we clarified the physiological role of a non-catalytic 20 kDa protein, BtrC2, associated with DOI synthase from the butirosin-producer Bacillus circulans. Based on the results of complementation analysis using btrC2 disruptant and western analysis against catal…

Spatial localization of bacteria controls coagulation of human blood by 'quorum acting'.

Blood coagulation often accompanies bacterial infections and sepsis and is generally accepted as a consequence of immune responses. Though many bacterial species can directly activate individual coagulation factors, they have not been shown to directly initiate the coagulation cascade that precedes clot formation. Here we demonstrated, using microfluidics and surface patterning, that the spatial localization of bacteria substantially affects coagulation of human and mouse blood and plasma. Bacillus cereus and Bacillus anthracis, the anthrax-causing pathogen, directly initiated coagulation of b…

[Whipple's disease--a challenging diagnosis].

Whipple's Disease is a rare multisystemic disease caused by Gram-positive bacillus, Tropheryma whippelii, family of Actinobacterias and group of Actinomycetes. Because the disease is rare and has different forms of presentation the diagnosis comes frequently late. The authors present the case of a white man, 50 years old, admitted with a clinical picture characterized by weight loss, abdominal pain, ascites, diarrhea. He suffered of arthralgias for four years. The diagnosis was made by biopsy of mesenteric adenopathies and liver, during the laparotomy. The biopsy specimens showed numerous aggr…

Predictive value of isolating Pseudomonas aeruginosa from aerobic and anaerobic blood culture bottles.

Pseudomonas aeruginosa is a particularly virulent pathogen when it causes bacteraemia and early diagnosis is essential to reduce morbidity a … f 73.8 %. However, for all episodes of Gram-negative bacteraemia, the likelihood of a Gram-negative bacillus occurring in the aerobic bottle first being P. aeruginosa was only 39 %. The converse finding of a Gram-negative bacillus isolated first in the anaerobic bottle or from both bottles together was clinically helpful, having a negative predictive value …

Robert Koch: Nobel laureate and controversial figure in tuberculin research.

Tuberculosis has been a major cause of death for centuries. Likewise, anthrax has posed a deadly threat to both farm animals and humans and today poses a threat as a weapon of biological warfare. Cholera, which wreaked havoc in the East and threatened to enter Europe, also posed a deadly threat. The causes of these diseases remained mysteries for centuries. Nobel laureate Robert Koch (1843-1910), often called the founder of medical bacteriology, is credited with discovering the tubercle bacillus, Mycobacterium tuberculosis; with demonstrating for the first time in history the life cycle of the…

Detection of pathogenic and non-pathogenic bacteria by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry.

The proteins isolated from the whole cells of bacterial pathogens and related non-pathogenic simulants were analyzed directly, with minimal sample preparation by matrix-assisted laser desorption/ionization (MALDI) time-of-flight mass spectrometry. Inspection of mass spectrometric profiles obtained from direct MALDI-MS analysis of the protein extracts revealed specific biomarkers for individual bacterial cells. The observed biomarkers enabled us not only to detect pathogenic bacteria (Bacillus anthracis, Yersinia pestis and Brucella meliteusis), but also to distinguish them from the correspondi…

[Activities and isozymes of adenosine deaminase and lactate dehydrogenase in tuberculous pleural effusion with special reference to the presence of mycobacterium tuberculosis].

We have previously shown that tuberculous pleurisy possesses a high level of adenosine deaminase (ADA) which is predominantly composed of ADA2. In this paper, we report the cases of tuberculous pleural effusion which contained mainly ADA1. In these cases, mycobacterium tuberculosis was positive by smear examination and/or culture and granulocytes were found to be major components. Analysis of lactate dehydrogenase (LDH) revealed that its activity was high and LDH5 occupied about 50% of total activity. In the tubercle bacillus negative cases, lymphocytes were the main components and the levels …

MALDI-TOF misidentification of Sanguibacteroides justesenii: confirmation by 16S rRNA sequencing.

Sanguibacteroides justesenii is a rarely reported Gram-negative anaerobic bacillus of uncertain clinical significance. Its identification is challenging using conventional methods. We report the case of a 58-year-old woman admitted for acute heart failure who developed septic shock following colonic perforation. Blood cultures grew an obligate anaerobic Gram-negative bacillus. MALDI-TOF MS (Bruker®) identified the isolate as Gabonibacter massiliensis (score 2.19). Given the unexpected result, the strain was referred to a reference laboratory, where 16S rRNA gene sequencing identified S. justes…

Rapid quality evaluation of Pu-erh raw tea storage years via hyperspectral imaging coupled with machine learning and microbial profiling.

This study developed a non-destructive approach for identifying storage years and quality evolution of Pu-erh raw tea by integrating sensory evaluation, chemical analysis, electronic nose/tongue, hyperspectral imaging, and 16S rRNA sequencing. Results revealed that Bacillus and Pantoea shifts drive chemical transformations; notably, Bacillus abundance reached 40.44% in Hangzhou-stored samples after six years, correlating with significant degradation of polyphenols and ester-type catechins. Specific wavelengths at 1633, 1085, and 1250 nm were intrinsically linked to alterations in caffeine, pol…

Hydrogen peroxide (H2O2)-independent microbial advanced oxidation process for treatment of azo compound-contaminated water.

Azo compounds have adverse effects on the environment and public health, requiring effective treatment methods. Advanced oxidation processes (AOPs) are promising but hindered due to their toxic byproducts and high energy demands. Here, we report the development of magnesium-induced microbial AOP facilitated by biogenic iron nanoparticles. The novel strain, Bacillus sp. YN-1, capable of degrading azo compounds, was first isolated and used to synthesize iron nanoparticles, facilitating a Fenton-like reaction. Magnesium was used as an electron donor, replacing hydrogen peroxide to generate reacti…

Determination of the X-ray structure of the snake venom protein omwaprin by total chemical synthesis and racemic protein crystallography.

The 50-residue snake venom protein L-omwaprin and its enantiomer D-omwaprin were prepared by total chemical synthesis. Radial diffusion assays were performed against Bacillus megaterium and Bacillus anthracis; both L- and D-omwaprin showed antibacterial activity against B. megaterium. The native protein enantiomer, made of L-amino acids, failed to crystallize readily. However, when a racemic mixture containing equal amounts of L- and D-omwaprin was used, diffraction quality crystals were obtained. The racemic protein sample crystallized in the centrosymmetric space group P2(1)/c and its struct…

Coaggregation Strengthened Nanozymes for the Identification of Oral Cariogenic Bacteria.

Dental caries, caused by oral cariogenic bacteria, underscore the critical importance of early diagnosis for effective prevention and treatment. The identification of oral cariogenic bacteria depends on the development of the recognition elements. In this work, coaggregation between bacteria has been subtly mined for exploration of the recognition elements, which has been further used to construct a colorimetric sensor array for the identification of nine oral cariogenic bacteria. Three fermentative bacteria (Kocuria sp. RXG-8, Bacillus sp. 7578-18, and Bacillus sp. LTZ-12) can coaggregate wit…
