New clues for the role of cerebellum in schizophrenia and the associated cognitive impairment.

Schizophrenia (SZ) is a complex neuropsychiatric disorder associated with severe cognitive dysfunction. Although research has mainly focused on forebrain abnormalities, emerging results support the involvement of the cerebellum in SZ physiopathology, particularly in Cognitive Impairment Associated with SZ (CIAS). Besides its role in motor learning and control, the cerebellum is implicated in cognition and emotion. Recent research suggests that structural and functional changes in the cerebellum are linked to deficits in various cognitive domains including attention, working memory, and decisio

Clinical, cognitive and neural correlates of self-monitoring deficits in schizophrenia: an update.

BACKGROUND: An area of recent interest in schizophrenia research is to investigate specific neural and cognitive abnormalities associated with symptoms of this disorder. OBJECTIVE: To establish clinical, cognitive and neural correlates of self-monitoring deficits in schizophrenia, which according to various theoretical models can account for the first-rank symptoms of this disorder. METHODS: Relevant data were identified from PubMed and PsycInfo searches up to July 2006 using combinations of keywords including 'self-monitoring', 'symptoms', 'self-agency', 'neuropsychological', 'cognitive', '

[Subjective memory loss--a sign of cognitive impairment in the elderly? An overview of the status of research].

According to the growing clinical interest in early indicators of dementia, numerous studies have examined the association between subjective memory complaints and cognitive performance in old age. Their results are contradictory. In this paper, studies carried out over the last 10 years are compared with regard to the study design and the assessment instruments used. The results are discussed with particular reference to the diagnostic validity of subjective memory complaints. The majority of case-control studies and cross-sectional studies of non-representative samples could not demonstrate

Molecular and cellular correlates of human cortical lateralization.

Hemispheric specialization is a fundamental feature of human brain organization, with most individuals showing left hemisphere dominance for language and right hemisphere dominance for visuospatial attention. Although lateralized functions exist in other species, humans exhibit a pronounced, species-wide bias. Despite its evolutionary and functional significance, the molecular and cellular basis of this asymmetry remains unclear. Here, we identify neurochemical and cellular features associated with cortical lateralization. We show lateralized gradients in neurotransmitter receptor densities al

Cerebellar volumes' selective association with MoCA over MMSE: a diagnostic insight into mild cognitive impairment and mild dementia.

INTRODUCTION: Linked to motor control, cerebellum is increasingly recognized for its role in cognition and neurodegenerative disorders. METHODS: This retrospective study investigates associations between cerebellar volumes and cognitive screening tools-the Mini-Mental State Examination (MMSE) and the Montreal Cognitive Assessment (MoCA)-in individuals with mild cognitive impairment (MCI) or mild dementia. RESULTS: MoCA scores showed significant positive correlations with cognitive-related cerebellar regions, particularly the left Crus I lobule (r = 0.40, p = 0.02) and total Crus I volume (r

Tau-ing and fro-ing: the tanycytic shuttle in neurodegeneration.

After more than a century since Alzheimer's disease (AD) was described and decades of research into β-amyloid and Tau proteins, mechanisms underlying pathogenic protein clearance from brain remain poorly understood. Recent research identifies tanycytes-specialized hypothalamic cells lining the third ventricle-as a previously unrecognized clearance system for brain Tau. These cells actively transport Tau from cerebrospinal fluid to blood via pituitary portal circulation but are dramatically fragmented in AD brains. Single-nucleus RNA sequencing reveals altered stress and transport gene expressi

Cellular mechanisms of serotonergic psychedelics - apical hypercontextualisation.

Classical serotonergic psychedelics primarily exert their profound effects through agonism at the serotonin 2A (5-HT2A) receptor, which is abundantly expressed in many cortical regions, particularly in layer V pyramidal neurons of associative and visual areas. At the cellular level, these receptors are predominantly localized postsynaptically on the soma and apical dendrites. This review synthesizes evidence from distinct subfields of psychedelic cognitive neuroscience to illuminate how 5-HT2A agonism at apical dendrites of layer V pyramidal neurons disrupts the usual boundaries of conscious m

Autism spectrum disorder and personality disorders: differential diagnosis or comorbidity?

OBJECTIVE: The clinical overlap between autism spectrum disorder (ASD) and personality disorders (PDs) poses significant diagnostic challenges. Shared features - such as social communication difficulties, rigidity, and emotional dysregulation - often lead to misdiagnosis or delayed diagnosis, especially in adults and underrepresented populations (e.g., women). METHODS: This narrative review synthesizes current evidence on the differential diagnosis and comorbidity of ASD and PDs to guide clinical practice. RESULTS: Findings highlight substantial symptom overlap, particularly in social cognit

Cognitive-Developmental Mechanisms in Hallucinations.

Hallucinations figure prominently in a range of psychiatric disorders but, to date, their developmental origins are not well understood. The aim of the present article is to explore how ideas from mainstream developmental psychology can enhance understanding of how hallucinations develop in different modalities across the lifecourse. Hallucinations vary in their clinical significance depending on at what point they occur in the lifetime of the individual. Key cognitive-developmental processes include engaging with imaginary entities, exposure to adverse events, executive functioning, social co

Grey-matter correlates of empathy in 4-Repeat Tauopathies.

Loss of empathy is an early and central symptom of frontotemporal lobar degeneration spectrum diseases. We aimed to investigate the topographical distribution of morphometric brain changes associated with empathy in Progressive Supranuclear Palsy (PSP) and Corticobasal Syndrome (CBS) patients. Twenty-seven participants with CBS and 31 with PSP were evaluated using Interpersonal Reactivity Index scales in correlation with gray matter atrophy using a voxel-based morphometry approach. Lower levels of empathy were associated with an increased atrophy in fronto-temporal cortical structures. At subc

Neonatal immune challenge poses a sex-specific risk for epigenetic microglial reprogramming and behavioral impairment.

While the precise processes underlying a sex bias in the development of central nervous system (CNS) disorders are unknown, there is growing evidence that an early life immune activation can contribute to the disease pathogenesis. When we mimicked an early systemic viral infection or applied murine cytomegalovirus (MCMV) systemically in neonatal female and male mice, only male adolescent mice presented behavioral deficits, including reduced social behavior and cognition. This was paralleled by an increased amount of infiltrating T cells in the brain parenchyma, enhanced interferon-γ (IFNγ) sig

Experimental and computational analyses of calcium dynamics in 22q11.2 deletion model astrocytes.

Methods for deriving mechanistic information from intracellular calcium dynamics have largely been applied to neuronal data despite the knowledge of roles of glial cells in behavior, cognition, and psychiatric disorders. Using calcium imaging, computer vision, and Bayesian kinetic inference (BKI), we analyzed calcium dynamics in primary astrocytes derived from control or Df1/+ mice, a model of 22q11.2 deletion (DiGeorge syndrome). Inference of the highest-likelihood molecular kinetic characteristics of intracellular calcium dynamics identified changes in the activity of the sarcoendoplasmic re

Epigenetic Regulation of the Hippocampus, with Special Reference to Radiation Exposure.

The hippocampus is crucial in learning, memory and emotion processing, and is involved in the development of different neurological and neuropsychological disorders. Several epigenetic factors, including DNA methylation, histone modifications and non-coding RNAs, have been shown to regulate the development and function of the hippocampus, and the alteration of epigenetic regulation may play important roles in the development of neurocognitive and neurodegenerative diseases. This review summarizes the epigenetic modifications of various cell types and processes within the hippocampus and their

Co-Occurrence of Autism Spectrum Disorder and Achondroplasia.

Autism spectrum disorder (ASD) and achondroplasia are common disorders on their own. However, this case of co-occurrence in the same patient has not yet been reported in literature except for a hypothesized statistical probability based on prevalence studies stating that two to five in 10 million children could have the probability of having both conditions occurring simultaneously. Achondroplasia typically presents with motor delays and difficulties that are related to musculoskeletal impairments that can affect self-care, mobility, and social cognition; however, the presence of delays in oth

Hallucinogens and Serotonin 5-HT2A Receptor-Mediated Signaling Pathways.

The neuropsychological effects of naturally occurring psychoactive chemicals have been recognized for millennia. Hallucinogens, which include naturally occurring chemicals such as mescaline and psilocybin, as well as synthetic compounds, such as lysergic acid diethylamide (LSD), induce profound alterations of human consciousness, emotion, and cognition. The discovery of the hallucinogenic effects of LSD and the observations that LSD and the endogenous ligand serotonin share chemical and pharmacological profiles led to the suggestion that biogenic amines like serotonin were involved in the psyc

Atrial fibrillation, cognition and dementia: A review.

Atrial fibrillation (AF) is one of the most common types of cardiac arrhythmia, particularly among older adults. AF confers a 5-fold risk for thromboembolic stroke as well as a 2-fold higher risk for congestive heart failure, morbidity, and mortality. Although stroke remains an important and impactful complication of AF, recent studies have shown that AF is independently associated with other neurological disorders, including cognitive impairment and dementia, even after adjusting for prior ischemic stroke. We performed a review of the published literature on the association between AF and cog

HIV-associated neurocognitive disorders and the impact of combination antiretroviral therapies.

HIV-associated neurocognitive disorders (HAND) are the most common preventable and treatable cause of dementia. While the incidence of the most severe form of HAND, HIV-associated dementia, has decreased since the introduction of combination antiretroviral therapy (cART), the prevalence of less severe forms of HAND has continued to rise. HAND leads to a subcortical dementia consisting of a triad of cognitive, behavior, and motor dysfunction. No single laboratory test can establish HAND, but ancillary studies including neuropsychological testing, neuroimaging studies, and cerebrospinal fluid (C
