Prevention of amitriptyline-induced avoidance impairment by tacrine in mice.

The effects of two cognition enhancers on avoidance impairment induced by the tricyclic antidepressant amitriptyline were assessed during shuttle-box avoidance acquisition and in previously trained mice of the DBA/2 strain. The nootropic agent piracetam (50, 100 or 200 mg/kg, i.p.) had slight or no effect in mice receiving amitriptyline (5 or 10 mg/kg, i.p.). Conversely, the acetylcholinesterase inhibitor tacrine (0.5, 1, 2 or 3 mg/kg, i.p.) prevented the avoidance impairment induced by 5 mg/kg amitriptyline on shuttle-box avoidance acquisition as well as on a previously learned avoidance resp

Graph Neural Network Learning on the Pediatric Structural Connectome.

PURPOSE: Sex classification is a major benchmark of previous work in learning on the structural connectome, a naturally occurring brain graph that has proven useful for studying cognitive function and impairment. While graph neural networks (GNNs), specifically graph convolutional networks (GCNs), have gained popularity lately for their effectiveness in learning on graph data, achieving strong performance in adult sex classification tasks, their application to pediatric populations remains unexplored. We seek to characterize the capacity for GNN models to learn connectomic patterns on pediatri

Optimizing Existing Mental Health Screening Methods in a Dementia Screening and Risk Factor App: Observational Machine Learning Study.

BACKGROUND: Mindstep is an app that aims to improve dementia screening by assessing cognition and risk factors. It considers important clinical risk factors, including prodromal symptoms, mental health disorders, and differential diagnoses of dementia. The 9-item Patient Health Questionnaire for depression (PHQ-9) and the 7-item Generalized Anxiety Disorder Scale (GAD-7) are widely validated and commonly used scales used in screening for depression and anxiety disorders, respectively. Shortened versions of both (PHQ-2/GAD-2) have been produced. OBJECTIVE: We sought to develop a method that ma

Targeting the lateral hypothalamus with short hairpin RNAs reduces habitual behaviour following extended instrumental training in rats.

Instrumental actions are initially goal-directed but with repeated performance can become habitual. Habitual actions are adaptive, learned behaviours that are automated in order to reduce cognitive load and to allow for efficient interaction with the environment. Goal-directed and habitual actions are mediated by distinct neurocircuits which centre on the dorsal striatum and involve different cortical and limbic inputs. The lateral hypothalamus (LH) has yet to be considered in this neurocircuitry despite its anatomical connections with these neurocircuits and its established role in motivated

Multimodal intervention in 8- to 13-year-old French dyslexic readers: Study protocol for a randomized multicenter controlled crossover trial.

BACKGROUND: Developmental dyslexia, a specific and long-lasting learning disorder that prevents children from becoming efficient and fluent readers, has a severe impact on academic learning and behavior and may compromise professional and social development. Most remediation studies are based on the explicit or implicit assumption that dyslexia results from a single cause related to either impaired phonological or visual-attentional processing or impaired cross-modal integration. Yet, recent studies show that dyslexia is multifactorial and that many dyslexics have underlying deficits in severa

Quantifying deep grey matter atrophy using automated segmentation approaches: A systematic review of structural MRI studies.

The deep grey matter (DGM) nuclei of the brain play a crucial role in learning, behaviour, cognition, movement and memory. Although automated segmentation strategies can provide insight into the impact of multiple neurological conditions affecting these structures, such as Multiple Sclerosis (MS), Huntington's disease (HD), Alzheimer's disease (AD), Parkinson's disease (PD) and Cerebral Palsy (CP), there are a number of technical challenges limiting an accurate automated segmentation of the DGM. Namely, the insufficient contrast of T1 sequences to completely identify the boundaries of these st

Stricter national standards are required for credentialing of endoscopic-retrograde-cholangiopancreatography in the United States.

Endoscopic-retrograde-cholangiopancreatography (ERCP) is now a vital modality with primarily therapeutic and occasionally solely diagnostic utility for numerous biliary/pancreatic disorders. It has a significantly steeper learning curve than that for other standard gastrointestinal (GI) endoscopies, such as esophagogastroduodenoscopy or colonoscopy, due to greater technical difficulty and higher risk of complications. Yet, GI fellows have limited exposure to ERCP during standard-three-year-GI-fellowships because ERCP is much less frequently performed than esophagogastroduodenoscopy/colonoscopy

A machine learning framework for classifying dementia risk in mild cognitive impairment: evidence from a Korean genome-wide association study cohort.

BACKGROUND: Alzheimer's disease (AD) is a progressive neurodegenerative disorder that presents challenges for early detection and intervention. Mild cognitive impairment (MCI), a critical precursor of AD, progresses to dementia in a substantial proportion of individuals annually. Genetic factors, particularly single nucleotide polymorphisms (SNPs), play a key role in the pathogenesis of AD, as identified by genome-wide association studies (GWAS). Therefore, we aimed to develop and evaluate predictive models for classifying patients with MCI into high- and low-risk groups for dementia using SNP

Digital Marker for Early Screening of Mild Cognitive Impairment Through Hand and Eye Movement Analysis in Virtual Reality Using Machine Learning: First Validation Study.

BACKGROUND: With the global rise in Alzheimer disease (AD), early screening for mild cognitive impairment (MCI), which is a preclinical stage of AD, is of paramount importance. Although biomarkers such as cerebrospinal fluid amyloid level and magnetic resonance imaging have been studied, they have limitations, such as high cost and invasiveness. Digital markers to assess cognitive impairment by analyzing behavioral data collected from digital devices in daily life can be a new alternative. In this context, we developed a "virtual kiosk test" for early screening of MCI by analyzing behavioral d

Intratumoral and Peritumoral Functional Connectivity Reveals Network-Level Signatures of Glioma.

Gliomas are recognized as network-level diseases that remodel functional connectivity across the brain. Using resting-state functional MRI (rs-fMRI) data from 306 patients with pathologically confirmed glioma at two hospitals, we delineated individualized intra- and peritumoral functional networks to quantify tumor-related alterations. Two novel indices-the ratio of intratumoral functional regions (RIFR) and ratio of peritumoral tumor-related regions (RPTR)-were proposed to describe network remodeling. Both indices were related to pathological, structural, functional, cognitive, language, and

Why can't they "Go back"? - A multi-group comparative analysis of pre-service teachers' hometown return intention in local universities based on latent profile analysis.

The high-quality development of rural education urgently requires strengthening the supply of outstanding rural teachers. Based on Social Cognitive Career Theory (SCCT), a survey of 6,546 teacher education students from local universities in Heilongjiang Province revealed that the willingness to teach in rural areas among three identified profiles-the Weak Foundation Profile, the Development-Oriented Profile, and the Excellence-Prepared Profile-is significantly influenced by academic year and family capital. This influence manifests differentiated patterns via the "environment-person-behaviour

A proposed anxiety-hypertonic pelvic floor axis in postpartum women: a narrative review of mechanistic links and rehabilitation implications.

Pelvic floor dysfunction (PFD) and postpartum anxiety are commonly assessed and treated separately, although accumulating evidence suggests that psychological distress and pelvic floor muscle over activity may interact. This narrative review synthesizes epidemiological, neurobiological, and rehabilitation evidence and proposes the Anxiety-Hypertonic Pelvic Floor Axis as a framework for this bidirectional association. Postpartum anxiety and chronic stress may contribute to hypothalamic-pituitary-adrenal axis dysregulation, sympathetic predominance, inflammatory signaling, and central sensitizat

Religious leaders as mental health gatekeepers in Nigeria: Belief-attitude typologies and implications for faith-health system integration: Findings from the CLERIC study.

In Nigeria, religious leaders are de facto mental health gatekeepers, yet their explanatory models and attitudes remain poorly characterised at the population level. This cross-sectional study maps causal beliefs and attitudes among 207 Christian and Muslim religious leaders in Lagos using validated, culturally adapted instruments, forming the attitudinal and typological component of the CLERIC research programme. An integrated conceptual model posits that explanatory models structure causal attributions, which constitute the cognitive foundation of stigma-related attitudes clustering into typ

Measuring student perceptions of motivation and engagement with Kahoot!: a case study in primary education in Portugal.

The integration of digital tools in teaching remains a challenge for many educators, often due to resistance to change and limited training. Drawing on Self-Determination Theory, as proposed by Deci and Ryan, and Cognitive Load Theory, developed by Sweller, this exploratory study examines student motivation and engagement in a gamified learning environment. Specifically, it explores students' perceptions of the use of Kahoot! in the classroom and the extent to which the tool may add value to the teaching and learning process. The study involved 20 fourth-grade students and their class teacher

Deep learning for early detection of cerebral small vessel disease using self-supervised graph embeddings and retinal image analysis.

The primary driver or cause of cognitive decline and stroke is Cerebral Small Vessel Disease (CSVD), which currently requires neuroimaging tests, which are expensive to obtain and inaccessible in standard clinical settings. Low-cost retinal imaging techniques offer non-invasive assessments that mirror the condition of the brain’s small blood vessels (cerebral microvasculature). State-of-the-art diagnostic methods currently have no accessible, non-invasive, or cost-effective solution to identify CSVD at its earliest stages through visual assessment of retinal biomarkers. This study presents the
