C-terminus-outward orientation of SARS-CoV-2 envelope proteins on viral capsid enables a novel virus-cell interaction pathway.

Since the onset of COVID-19, emphasis has been largely placed on spike proteins, while other molecules of severe acute respiratory syndrome … roteins plus mass spectrometry revealed that the EC38-interacting proteins were enriched for "actin cytoskeleton organization," "cell-cell interaction," "cell-cell adhesion," "hemostasis," "viral infection pathways," "cytokine signaling in immune system," etc., among which CKAP4, vimentin, and ATP5B were further identified to bind EC38 peptides. Molecular dock …

Deciphering Membrane Protein Complexes in Plasmodium falciparum Gametocytes via Integrative Structural Systems Biology.

Malaria is caused by protozoan parasites of the genus Plasmodium that proliferate asexually in human erythrocytes. During each replication cycle, a small fraction of the parasites differentiates into gametocytes. These sexual gametocytes are the only stages of the parasite that can infect the mosquito vector and transmit malaria. Their progression and maturation depend on a profound remodeling of the erythrocyte. This is achieved by the export of parasite proteins into the erythrocyte, leading to structural and mechanistic changes in the cytoskeleton and membrane of the host cell, crucial for …

Cytoplasmic region of beta-dystroglycan is essential for postsynaptic maturation and neuromuscular function in mice.

The dystrophin-glycoprotein complex (DGC) provides structural integrity to the sarcolemma, and disruption of the DGC leads to muscular dystrophy. A core member of the DGC is dystroglycan (DG), which binds to extracellular ligands via α-DG and intracellular cytoskeleton via β-DG. Mutations in DAG1 or genes involved in the posttranslational processing of DG lead to a subset of neuromuscular diseases referred to as dystroglycanopathies. The importance of the α-DG extracellular interactions is well established; however, little is known about the significance of the β-DG intracellular interactions.…

HEV replication is promoted by blocking the NF-κB signaling pathway through inhibiting FLNa expression.

Hepatitis E virus (HEV) is the most common cause of acute viral hepatitis worldwide. Filamin A (FLNa), a cytoskeletal protein, is involved in cytoskeleton remodeling to construct a barrier to infection and participates in virus entry and release. However, how HEV enters host cells and how it is sensed by pattern recognition receptors are largely unexplored. In this study, the role of FLNa during HEV infection was evaluated in patients with HEV infection, animal models, and cell cultures. Notably, HEV interacted with FLNa at the early stage of infection and remarkably inhibited the expression o…

Avian herpesvirus-specific LORF5 is a late gene, interacts with 19 viral and 111 host proteins, critical for virulence of Duck plague virus.

Duck plague virus (DPV) poses a major threat to the waterfowl industry. Its genome encodes multiple avian herpesvirus-specific genes with un … nteractome analysis revealed extensive interactions between LORF5 and host proteins involved in the cytoskeleton (actin/myosin) and metabolic pathways, suggesting the potential roles of LORF5 in intracellular transport and metabolic reprogramming. Collectively, these findings identify LORF5 as a novel DPV late protein and demonstrate that it is a critical viru …

LINC01638 promotes epithelial-to-mesenchymal transition in endometriosis epithelial cells by up-regulating RHOB via HDAC1 suppression.

RESEARCH QUESTION: Is LINC01638 involved in regulation of epithelial-to-mesenchymal transition (EMT) in endometriosis? DESIGN: A prospectiv … of transcription factors associated with EMT are down-regulated in knockdown cells, along with the cytoskeleton regulatory gene RHOB, while HDAC1 was up-regulated. Chromatin immunoprecipitation analysis and HDAC1 inhibitory treatment combined with LINC01638 knockdown indicated that LINC01638 regulates RHOB expression via HDAC1-mediated promoter deacetylation. …

New molecular targets in acute leukemias: cytoskeletal regulatory proteins.

Acute leukemias are hematological malignancies characterized by the uncontrolled proliferation of immature bone marrow cells, disrupting normal hematopoiesis. These diseases, classified into acute lymphoblastic leukemia and acute myeloid leukemia (AML), often result from acquired genetic alterations that drive deregulated cell growth and inhibit differentiation. The cytoskeleton has emerged as a promising therapeutic target due to its pivotal role in cellular processes such as adhesion, motility, and division. Among its components, stathmin 1 (STMN1) and ezrin (EZR) stand out for their signifi…

Formation of an Amyloid-like Structure During In Vitro Interaction of Titin and Myosin-Binding Protein C.

Protein association and aggregation are fundamental processes that play critical roles in a variety of biological phenomena from cell signal … with each other, and this interaction plays a partial role in the formation of a strong sarcomeric cytoskeleton. We found that under physiological ionic-strength conditions titin and MyBP-C form complexes in which an amyloid-like structure is present. The possible functional significance of amyloid-like aggregation of these proteins in muscle cells in vivo is …

Angiopoietin-2 regulates the phenotypic switch of vascular smooth muscle cells.

During uterine spiral artery remodeling, vascular smooth muscle cells (VSMCs) become disorganized and undergo phenotypic switching from a co … d by decreased contractile marker expression, increased proliferation and migration, and an altered cytoskeleton. VSMC expressed integrin β6 interacted directly with Ang-2 and induced phosphorylation of FAK (S910 and Y397), AKT (S473), and mTOR (S2448). Knockdown of FAK recovered the calponin loss induced by Ang-2 and resulted in lower EZH2 abundance. Inhibiti …

Understanding the role of Shroom3 in the developing mouse myocardium.

Loss of actin cytoskeleton control can hinder integral developmental and physiological processes and can be the basis for a subset of developmental defects. SHROOM3 is an actin binding protein, best characterized as being essential for neural tube closure in vertebrates. Shroom3 expression has also been identified in the developing heart, with some associated congenital heart defects. Here we show that the expression pattern of Shroom3 in the developing and adult mouse heart is specific to the myocardium. Using a gene trap line, we show that embryos with homozygous full-body Shroom3 loss die a…
