Genomic insights and molecular epidemiology through whole genome sequencing in clinical Serratia marcescens ST-367 isolated from the Kingdom of Bahrain.

BACKGROUND: Serratia marcescens may cause rare central nervous system infections. The growing antibiotic resistance in these isolates makes treatment challenging. S. marcescens (MIID-C14) was isolated from the cerebrospinal fluid of a 56-year-old female patient admitted to Salmaniya Medical Complex, Bahrain. Due to the virulence and multidrug resistance exhibited by this bacterium, we aimed to analyse the genetic makeup of this isolate. METHODS: The isolate was identified via MALDI-TOF mass spectrometry, antimicrobial susceptibility was performed by VITEK-2 system, and whole-genome sequencing

The relationship between insomnia and multiple gastrointestinal disorders: a Mendelian randomization study.

BACKGROUND: Some observational studies have suggested an association between insomnia and the risk of certain gastrointestinal disorders, such as gastroesophageal reflux, irritable bowel syndrome and peptic ulcers. However, the direction and magnitude of the causal relationship are difficult to determine due to confounding and reverse causation. The aim of this study is to explore the causal association between insomnia and a variety of gastrointestinal disorders from genetic level using Mendelian randomization. METHODS: We used a two-sample, two-way MR to analyze the link between insomnia an

Dietary advanced glycation end products may increase the incidence of allergic rhinitis depending on genetic susceptibility: a prospective cohort study.

Background: Limited evidence exists for the relationship between dietary advanced glycation end product (AGE) intake and allergic rhinitis (AR). Herein, the association between dietary AGEs and the risk of developing AR and whether genetic susceptibility influences the effects of dietary AGEs on AR were explored. Methods: In total, 125 276 participants without AR at baseline and having completed at least two 24-hour dietary recalls from the UK Biobank (2006-2010) were included. Dietary AGEs, specifically Nε-(1-carboxyethyl)-L-lysine (CEL), Nε-(carboxymethyl) lysine (CML), and Nδ-(5-hydro-5-met

CD36 and SR-B1 polymorphisms exhibit distinct association patterns in active and latent tuberculosis.

Introduction. Host genetics plays a pivotal role in determining disease susceptibility among individuals infected with Mycobacterium tuberculosis. Scavenger receptors (SRs) such as CD36 and SR-B1 mediate pathogen recognition and lipid uptake, both of which are central to mycobacterial entry and immune modulation.Gap Statement. Polymorphisms rs1761667 and rs3211938 in CD36 and rs4238001 in SR-B1 have not been investigated in any population in relation to both latent tuberculosis infection (LTBI) and active tuberculosis (TB).Aim. To genotype CD36 and SR-B1 polymorphisms and evaluate their associ

Whole transcriptome analysis to identify non-coding RNA regulators and hub genes in sperm of non-obstructive azoospermia by microarray, single-cell RNA sequencing, weighted gene co-expression network analysis, and mRNA-miRNA-lncRNA interaction analysis.

BACKGROUND: The issue of male fertility is becoming increasingly common due to genetic differences inherited over generations. Gene expression and evaluation of non-coding RNA (ncRNA), crucial for sperm development, are significant factors. This gene expression can affect sperm motility and, consequently, fertility. Understanding the intricate protein interactions that play essential roles in sperm differentiation and development is vital. This knowledge could lead to more effective treatments and interventions for male infertility. MATERIALS AND METHODS: Our research aim to identify new and

Gout-associated SNP at the IL1RN-IL1F10 region is associated with altered cytokine production in PBMCs of patients with gout and controls.

OBJECTIVES: Gout is caused by the response of the innate immune system to monosodium urate (MSU) crystals. A recent gout GWAS identified a signal of genetic association at a locus encompassing IL1RN-IL1F10. Colocalisation analysis using Genotype Tissue Expression Database (GTEx) eQTL data showed that the signal overlaps with genetic control of IL1RN/IL1F10 gene expression. We assess the functional implications of IL1RN rs9973741, the lead gout-associated variant. METHODS: We conducted functional validation of IL1RN rs9973741 in patients with gout and controls. The transcription level of IL1RN

Estimating the serological underrecognition of patients with weak or partial RHD variants.

BACKGROUND: For patients with weak or discrepant RhD RBC phenotypes, RHD genotyping is employed to determine need for RhD-negative management. However, many RHD variants are type D-negative or D-positive. Serological recognition rates (RRs) of weak and partial RHD variants are poorly characterized. STUDY DESIGN AND METHODS: Four US studies employing RHD genotyping for weak or discrepant RhD phenotypes provided data for race/ethnicity-specific serological recognition. Three studies used microplate, and 1 used gel and tube; 2 had anti-D data. We obtained White and Hispanic/Latino allele frequen

Causal effects and immune cell mediators between prescription analgesic use and risk of infectious diseases: a Mendelian randomization study.

INTRODUCTION: Clinical observations have found that prolonged use of analgesics increases the incidence of infection. However, the direct causal relationship between prescription analgesic use (PAU) and risk of infection (ROI) remains unclear. METHODS: This study used Mendelian randomization (MR) design to estimate the causal effect of PAU on ROI, as well as their mediating factors. Genetic data on prescription analgesics use and immune cells were obtained from published GWAS. Additionally, data on ROI were extracted from the FinnGen database. Two-sample MR analysis and multivariate MR (MVMR)

Exploring the mechanism of artificial selection signature in Chinese indigenous pigs by leveraging multiple bioinformatics database tools.

BACKGROUND: Chinese indigenous pigs in Yunnan exhibit considerable phenotypic diversity, but their population structure and the biological interpretation of signatures of artificial selection require further investigation. To uncover population genetic diversity, migration events, and artificial selection signatures in Chinese domestic pigs, we sampled 111 Yunnan pigs from four breeds in Yunnan which is considered to be one of the centres of livestock domestication in China, and genotyped them using Illumina Porcine SNP60K BeadChip. We then leveraged multiple bioinformatics database tools to f

Effects of Human Leukocyte Antigen DRB1 Genetic Polymorphism on Anti-Cyclic Citrullinated Peptide (ANTI-CCP) and Rheumatoid Factor (RF) Expression in Rheumatoid Arthritis (RA) Patients.

Rheumatoid arthritis (RA) is a systemic disease characterized by non-infectious inflammation of the joints and surrounding tissues, which can cause severe health problems, affect the patient's daily life, and even cause death. RA can be clinically diagnosed by the occurrence of blood serological markers, rheumatoid factor (RF) and anti-cyclic citrullinated peptide antibody (anti-CCP). However, about 20% of RA patients exhibit negative results for both markers, which makes RA diagnosis difficult and, therefore, may delay the effective treatment. Previous studies found some evidence that human l
