Interleukin-17 receptor A drives cancer stem-like properties in colorectal cancer through STAT3 activation.

Cancer stem cells (CSCs) play pivotal roles in tumor relapse, metastasis, and therapy resistance. Interleukin 17 receptor A (IL-17RA) is a key mediator in colorectal cancer (CRC) pathogenesis and progression. Our recent study demonstrated that reduced IL-17RA expression correlates with favorable prognosis in CRC patients and suppresses tumor growth in murine models. This study aimed to investigate the role of IL-17RA in promoting cancer stem-like properties and its impact on colorectal cancer prognosis and chemoresistance. Expression levels of IL-17RA and CSC markers in CRC cells were evaluate

Interleukin-6 Limits Host Susceptibility to Urinary Tract Infection by Promoting Urothelial Expulsion of Intracellular Bacteria.

INTRODUCTION: Interleukin (IL)-6 has an important role in limiting urinary tract infection (UTI). Mice lacking IL-6 are more susceptible to uropathogenic Escherichia coli (UPEC), including increased formation of intracellular bacterial communities (IBCs). How IL-6 promotes UPEC clearance is unknown. We hypothesize IL-6 reduces UTI susceptibility by limiting IBC formation through an early mechanism of infection. METHODS: Female mice were treated with vehicle or neutralizing antibodies to inhibit IL-6 or the IL-6 receptor (IL-6R) prior to transurethral UPEC infection. In rescue experiments, mur

Interleukin 27 (IL-27) in gastrointestinal cancers: from dual roles in tumor immunity to immunotherapeutic opportunities.

Gastrointestinal (GI) cancers continue to account for a major global cancer burden, with immune dysregulation recognized as a key determinant of their initiation and progression. Interleukin-27 (IL-27), a heterodimeric cytokine of the IL-12 family, has emerged as a pivotal but paradoxical regulator of tumor immunity in the GI cancers. IL-27 signals through the WSX-1/gp130 receptor complex, activating signal transducer and activator of transcription (STAT1) - and STAT3-dependent transcriptional pathways, yielding opposing outcomes. STAT1-dominant signaling promotes cytotoxic T-cell proliferatio

Interferon-γ and interleukin-17A associations with vascular dysfunction following paediatric cardiac surgery with cardiopulmonary bypass.

BACKGROUND: Cardiopulmonary bypass-induced inflammation is associated with poor postoperative outcomes. Bypass exposure has been associated with shifts in lymphocyte populations. This study aimed to describe two cytokine profiles associated with T and NK cells and their effects on clinical markers of postoperative cardiovascular dysfunction in children undergoing cardiac surgery with cardiopulmonary bypass. METHODS: Children from two major children's hospitals undergoing corrective cardiac surgery with cardiopulmonary bypass were included. Plasma was collected pre-, 0 to 4 hours post- and 24

NF-κB p65 modulates the promoter activity of large yellow croaker (Larimichthys crocea) interleukin-2.

Interleukin-2 (IL-2) plays pivotal roles in immune regulation, particularly in T cell-mediated adaptive immunity. Precise control of IL-2 transcription is therefore essential for proper immune function. Although the regulation of mammalian IL-2 has been well characterized, the molecular mechanisms governing IL-2 expression in teleost remain poorly understood. Here, we identified and characterized the 5' flanking region of the large yellow croaker (Larimichthys crocea, Lc) IL-2, which contains many putative conserved binding sites of transcription factors critical in regulating IL-2 expression,

Association of interleukin-6 rs1800796 polymorphism with keloid susceptibility and severity in a Vietnamese population: a case-control study.

Keloids are fibroproliferative skin tumors characterized by excessive collagen deposition and persistent inflammation. Interleukin (IL)-6 plays a key role in wound healing and fibrosis, and promoter polymorphisms of the IL-6 gene have been implicated in keloid susceptibility in several populations. However, data on Vietnamese patients remain limited. We conducted a case-control study including 47 patients with clinically diagnosed keloids and 43 age- and sex-matched healthy controls to investigate the association between the IL-6 rs1800796 (-572 C>G) polymorphism and both keloid susceptibility

Exploring the association between interleukin-27 and Dermatophagoides-specific IgE responses in children with allergic rhinitis and asthma.

PURPOSE: House dust mite (HDM) allergy is a common cause of allergic rhinitis (AR) and allergic asthma (AA). Interleukin-27 (IL-27) is known to suppress Th2-mediated inflammation, a key driver of these diseases. This study aimed to assess regional sensitization to Dermatophagoides subspecies and to investigate the association between HDM-specific IgE responses and serum IL-27 levels. METHODS: Fifty-eight children with HDM allergy were evaluated, of whom 53 were sensitized to D. Pteronyssinus. Serum Der p 1/Der p 2 specific IgE (sIgE) (FEIA) and IL-27, IL-5, and IL-13 levels (ELISA) were measu

Receptor-tuned Interleukin-2 Mutein Decouples Treg Expansion from ILC2-driven Cutaneous Inflammation.

Low-dose interleukin-2 (IL-2) and IL-2 muteins are being developed to expand regulatory T cells (Tregs) for autoimmune disease therapy, but injection site reactions (ISRs) remain frequent and poorly understood. Here we show IL2Mut24, a murine surrogate of IL-2 receptor α (IL-2Rα, CD25)-biased IL-2 mutein, enhances Treg expansion yet paradoxically exacerbates cutaneous inflammation compared to wild-type IL-2. Using immunodeficient mice, antibody blockade and adoptive transfer, we identify group 2 innate lymphoid cells (ILC2s) as key drivers of IL-2-induced skin inflammation, defining a CD25-dep

Systemic interleukin signaling and Alzheimer's disease risk: A two-sample cross-replication Mendelian randomization study.

BackgroundAlzheimer's disease (AD) is the leading cause of dementia worldwide, yet no disease-modifying therapy exists. Systemic inflammation has been proposed as a causal factor, supported by observational studies reporting elevated plasma interleukins in AD, though such studies cannot establish causality. Recent evidence instead highlights local neuroinflammation and microglial dysfunction as key mechanisms.ObjectiveThis study aimed to determine whether circulating interleukins and their receptors causally influence AD risk.MethodsWe conducted a two-sample Mendelian randomization (MR) analys…

Zwitterionic polymer-modified silicon nanowire field-effect transistor biosensor for interleukin-6 detection in saliva.

As a key inflammatory cytokine biomarker, interleukin-6 (IL-6) plays an essential role in the early diagnosis of inflammatory diseases. Nevertheless, long-term, continuous biosensing monitoring of ultralow-level IL-6 remains great challenges owing to severe nonspecific adsorption effects in complex biological media such as saliva. In this study, a silicon nanowire (SiNW) field-effect transistor (FET) functionalized with the zwitterionic polymer sulfobetaine methacrylate (SBMA) on the SiNW surface has been developed for IL-6 detection in saliva. After aptamer functionalization, the SBMA-modifie

Basophil-specific deletion of interleukin 15 impairs memory CD8+ T cell homeostasis in the bone marrow.

Basophils develop in the bone marrow and are associated with allergic inflammation but also contribute to protective immune responses against pathogens. Upon activation, they produce large amounts of cytokines, including interleukin (IL)-4 and IL-13. However, their physiological functions during homeostasis remain poorly defined. Here, using single-cell RNA sequencing and cytokine knock-in reporter mice, we identify basophils as the immune cell population with the highest IL-15 expression among immature and mature bone marrow leukocytes. Conditional deletion of IL-15 in basophils using Mcpt8-C

Clazosentan, Endothelin-A Receptor Antagonist, Suppresses Interleukin-6 Expression in Cerebrospinal Fluid after Subarachnoid Hemorrhage.

Clazosentan (CLA), an endothelin-A receptor antagonist, is used to prevent cerebral vasospasm (CV) following subarachnoid hemorrhage (SAH). Although its efficacy has been demonstrated in large-scale clinical trials, the effects of CLA on cytokine and chemokine profiles in cerebrospinal fluid (CSF) after SAH remain unclear. Twenty-two patients with SAH were enrolled. Ten patients received fasudil hydrochloride (FH), a Rho kinase inhibitor, and 12 received CLA. CSF samples were collected on days 1, 3, 5, 7, 10, and 14. Concentrations of interleukin-6 (IL-6), monocyte chemoattractant protein-1 (M

Discordant Interleukin-27 (IL-27) Subunit Transcription in Human Sepsis Exposes a Molecular-Attribution Gap.

Background/Objectives: Interleukin-27 (IL-27) is an obligate EBI3-p28 heterodimer, yet sepsis studies commonly measure and target the axis as a single entity. We tested whether both subunit transcripts are coordinately induced in septic monocytes and whether the axis has functional consequences. Methods: We integrated 272,993 whole-blood single-cell transcriptomes from 39 individuals, six additional single-cell cohorts and eight bulk cohorts, with two human macrophage datasets, a tolerant-like THP-1 model and caecal ligation and puncture in male C57BL/6 mice. Results: Across three cohorts (166

Interleukin-6 responses to acute stress are not altered in alcohol use disorder despite elevated baseline inflammation.

Acute stress activates the immune system, leading to the release of pro-inflammatory cytokines, such as interleukin-6 (IL-6). Chronic alcohol consumption alters the physiological stress systems and is associated with increased chronic inflammation. However, it remains unclear how IL-6 responds to acute stress in individuals with alcohol use disorder (AUD). Forty patients with AUD during early abstinence and 37 healthy controls (HC) completed two study visits. On one day, an acute stress induction task was performed, and on the other, a non-stressful control task, with the order of tasks being

Interleukin-1 Receptor Antagonist Levels in Patients With Heart Failure and Reduced Ejection Fraction Treated With Anakinra.

Patients with heart failure and reduced ejection fraction (HFrEF) commonly show signs of systemic inflammation. Interleukin-1 (IL-1) is a proinflammatory cytokine, known to modulate cardiac function. We aimed to determine the effects of treatment with anakinra, a recombinant IL-1 receptor antagonist (IL-1Ra), on plasma IL-1Ra levels. We measured IL-1Ra levels at baseline and longest available follow-up to 24 weeks in 63 patients (44 men, 40 self-identified Black Americans) with recent hospitalization for HFrEF and systemic inflammation [C-reactive protein (CRP) levels >2 mg/L] who were assigne
