Chorioretinal lesions, sea-blue histiocytes and other manifestations in familial chronic granulomatous disease.

Several little-emphasized manifestations of familial chronic granulomatous disease are considered: destructive chorioretinal lesions may be as constant as the pigmented histiocytosis seen in reticuloendothelial organs and could be related to a defect in the phagocytic activity of the retinal pigment epithelium; pigmented histiocytes with the staining characteristics of sea-blue histiocytes may be present in the bone marrow; patients may present with lesions resembling eosinophilic granuloma. Also discussed are some observations related to the sequestration of bacteria within phagocytic cells a

WAARDENBURG'S SYNDROME AND HETEROCHROMIA IRIDUM IN A DEAF SCHOOL POPULATION.

Waardenburg's syndrome consists of lateral displacement of the inner canthi of the eyes (dystopia canthorum), a broad nasal root and confluent eyebrows, heterochromia iridum, a white forelock and congenital deafness. The syndrome is inherited as a dominant, but affected individuals do not necessarily have all of the characteristics cited.Five hundred and fourteen pupils at a school for the deaf were screened for features of this syndrome. Three cases were discovered. Eleven other deaf children were found to have heterochromia iridum and two more had white forelocks. The interocular dimensions

A protease-resistant D-amino acid αA-crystallin mini-chaperone with enhanced chaperone and antioxidant activities protects against amyloid-β proteotoxicity.

Protein misfolding, aggregation, and oxidative stress contribute to aging and numerous neurodegenerative diseases, including Alzheimer's disease. Although the αA-crystallin-derived mini-chaperone peptide suppresses protein aggregation, its therapeutic potential is limited by susceptibility to proteolytic degradation. Here, we synthesized a protease-resistant analog composed entirely of D-amino acids (D-MC-αA) and compared its biochemical and biological properties with those of the parent peptide (MC-αA). Chaperone activity was assessed using two aggregation-prone model proteins, antioxidant ac

Decoding the multidimensional landscape of skin aging: molecular pathways and next-generation diagnostics & therapeutics.

Skin aging is a multifactorial, dynamically regulated biological process modulated by the synergistic interplay of intrinsic genetic programming and extrinsic environmental stressors. Chronological skin aging is dominated by molecular signatures such as telomere attrition, mitochondrial dysfunction, epigenetic drift, and fibroblast senescence. Extrinsic factors such as UV-induced photooxidation, pollution, lifestyle toxins accelerate ROS surge, DNA photolesions, and extracellular matrix fragmentation. Extrinsic and intrinsic pathways converge to cause epidermal thinning, collagen degradation,

Nutritional supplements: current evidence for retinitis pigmentosa and Stargardt disease.

PURPOSE OF REVIEW: Inherited retinal diseases (IRDs) are genetically and phenotypically heterogeneous disorders that cause progressive vision loss and lack broadly effective disease-modifying therapies. Increasing evidence implicates metabolic stress, oxidative injury, and photoreceptor-retinal pigment epithelium (RPE) dysfunction in IRD pathophysiology. This review evaluates the evidence and limitations surrounding nutritional and metabolic interventions for retinitis pigmentosa and Stargardt disease.A narrative review of preclinical studies, randomized controlled trials, and contemporary gen

Are Müller glial cells gatekeepers of neuroprotection and regeneration in age-related macular degeneration? Unraveling their roles in pathophysiology and therapeutic innovation.

Age-related macular degeneration (AMD) is traditionally conceptualized as a disorder of the retinal pigment epithelium (RPE)-photoreceptor axis; however, this paradigm incompletely explains early disease dynamics and therapeutic failure in geographic atrophy (GA). This review aims to reframe AMD progression through a Müller glial cell-centered framework that integrates cellular homeostasis, structural transitions, and stage-dependent therapeutic implications. Emerging transcriptomic, histologic, and functional evidence demonstrates that Müller glial cells actively participate in AMD pathobiolo

Genome Wide Pleiotropic Analysis Reveals Shared Genetic Architecture and Pathological Basis Between Retinitis Pigmentosa and Relevant Ocular Comorbidities.

PURPOSE: Retinitis pigmentosa (RP) is a leading cause of irreversible blindness in working-age populations and frequently cooccurs with multiple ocular disorders. As their shared genetic basis remains poorly understood, this study aimed to elucidate the shared genetic architecture underlying RP and relevant ocular comorbidities. METHODS: Using large-scale genome-wide association data, we systematically investigated pleiotropy, genetic correlation, cell-type specificity, and genetically predicted causality between RP and major ophthalmic comorbidities. RESULTS: We observed significant genetic

Ophthalmic Manifestations of KIF11-Associated Microcephaly With or Without Chorioretinopathy, Lymphedema, or Intellectual Disability: A Case Report of a Novel Variant.

BACKGROUND Mutations in the kinesin family member 11 (KIF11) gene have been recently identified in several families worldwide. This gene plays a crucial role in cell division, chromosomal positioning, and separation. KIF11 mutations are associated clinically with microcephaly with or without chorioretinopathy, lymphedema, or intellectual disability (MCLMR). It is an autosomal dominant disorder with reduced penetrance, and few reports in the literature describe its phenotypic associations. CASE REPORT We present the case a 39-year-old man from the Kingdom of Saudi Arabia with a known history of

Interactions Between Epidermal Growth Factor-Containing Fibulin-Like Extracellular Matrix Protein 1 and Tissue Inhibitor of Metalloproteinases-3 and Relevance to Age-Related Macular Degeneration.

CLINICAL RELEVANCE: Age-related macular degeneration (AMD) remains the leading cause of irreversible blindness in Western populations, with no approved therapies for the dry form characterized by drusen accumulation and retinal pigment epithelium atrophy. This review examines extracellular matrix alterations in Bruch's membrane by comparing healthy aging, AMD pathogenesis, and Doyne Honeycomb Retinal Dystrophy-a monogenic disorder that serves as a surrogate model for AMD due to similar phenotypic manifestations. METHODS: We focus on the critical interactions between epidermal growth factor-co

Iron deficiency stress impairs iron absorption and accumulation, compromising photosynthetic performance and antioxidant defense to limit grapevine growth.

Iron (Fe) deficiency is a common nutritional disorder that causes iron chlorosis in crop production, affecting plant growth, development, yield, and quality. To elucidate the growth-physiological characteristics, iron accumulation dynamics, and allocation patterns of grapevines under long-term gradient iron deficiency stress, this experiment was conducted. Using 'Red Globe' grapevines as the experimental subject, four gradient Fe-deficiency treatments (1/5, 2/5, 3/5, and 4/5 of normal iron supply) were established under tissue culture conditions, with a control (CK) group maintained at normal

Efficacy and Safety Evaluation of Microneedling Combined with Tranexamic Acid-Arbutin Liquid Excipients in the Treatment of Melasma: A Retrospective Study.

BACKGROUND: The condition of melasma presents a complex pigmented skin disorder, characterized by increased production of melanin in the dermis. The main pathological change of melasma is photodamage to the dermis. The treatment of melasma is relatively complex, and tranexamic acid is a well-known drug, both topical and oral. While arbutin demonstrates notable skin brightening effects, it has not yet gained widespread clinical adoption. Microneedle therapy, however, not only enhances drug penetration and absorption but also stimulates tissue regeneration. Nevertheless, research on the combined
