Spinal muscular atrophy: untangling the knot?

Spinal muscular atrophy (SMA), a clinically and genetically heterogeneous group of neuromuscular diseases, is a disorder of motor neurones characterised by degeneration of spinal cord anterior horn cells and muscular atrophy. SMA is an autosomal recessive disorder with a carrier frequency of about 1150. Three candidate genes, the survival motor neurone (SMN) gene, the neuronal inhibitory protein (NAIP) gene, and the p44 (subunit of basal transcription factor TFIIH) gene, have been considered as genes involved in this condition. The region spanning these genes has a complex organisation includi

Genes regulating hypothalamic and pituitary development.

Several pituitary transcription factors have been identified in the last 3 years. They offer new insights into the processes that direct organogenesis, cell commitment, proliferation and differentiated function. All are DNA-binding proteins, but they have ties to different families of homeodomain proteins. They differ in their distribution and in the timing of their appearance and extinction. The Rathke's pouch homeobox protein (Rpx) has a paired-like homeodomain. In mice, it appears on embryonic day 8.5 (day e8.5) and is gone by day e14.5. Its targets for activation are unknown. Pituitary OTX

Molecular basis of spectrin deficiency in beta spectrin Durham. A deletion within beta spectrin adjacent to the ankyrin-binding site precludes spectrin attachment to the membrane in hereditary spherocytosis.

We describe a spectrin variant characterized by a truncated beta chain and associated with hereditary spherocytosis. The clinical phenotype consists of a moderate hemolytic anemia with striking spherocytosis and mild spiculation of the red cells. We describe the biochemical characteristics of this truncated protein which constitutes only 10% of the total beta spectrin present on the membrane, resulting in spectrin deficiency. Analysis of reticulocyte cDNA revealed the deletion of exons 22 and 23. We show, using Southern blot analysis, that this truncation results from a 4.6-kb genomic deletion

Modulation of allosteric interactions in neurophysin induced by succinylation of serine-56 or cleavage of residues 1-8.

Neurophysin is an allosteric protein in which peptide binding and self-association are positively linked. Reaction of neurophysin with succinic anhydride led to a large decrease in peptide affinity assignable to succinylation of a serine or threonine hydroxyl group. To identify the residue involved, acetimidated protein was reacted with [14C]succinic anhydride and the active and inactive components were separated by affinity chromatography. Performic acid oxidation and tryptic and Asp-N mapping of the two components, followed by automated Edman degradation, allowed identification of the critic

Photosensory transduction in ciliates. II. Possible role of G-protein and cGMP in Stentor coeruleus.

The heterotrichous ciliate, Stentor coeruleus, exhibits a well-defined photophobic response to a sudden increase in the intensity of visible light. The phobic reactions usually appear with a latency period (i.e. a time delay between the onset of the stimulus and the stop response). This latency of phobic response was significantly increased when the cells were incubated with 8-bromo-guanosine 3',5'-cyclic monophosphate. In the presence of this nucleotide, a reduction of cell responsiveness (i.e. the number of photophobically responding cells) was also observed. Similar effects were observed wh

Integrated plasma proteomic and metabolomic profiling demonstrated specific proteins and bile acids remodeling after liver transplantation of children with biliary atresia.

This study aimed to characterize plasma protein and bile acid remodeling after liver transplantation (LT) in children with biliary atresia (BA). Paired plasma samples were collected from seven BA patients before and after LT following unsuccessful Kasai portoenterostomy, and profiled using label-free quantitative proteomics and targeted metabolomics of 15 bile acids. A total of 113 proteins were significantly altered after LT, enriched in pathways governing inflammation, coagulopathy, and extracellular matrix reorganization; eight proteins were selected as signature biomarkers, including upreg

NADPH oxidase subunit p22phox: A marker of oxidase-dependent oxidative stress and target for stress suppression in nonphagocytic cells.

Reactive oxygen species (ROS)-producing NADPH oxidase (Nox) family proteins are involved in host defense. The overproduction of ROS leads to oxidative stress, which is associated with a myriad of diseases. The Nox subunit p22phox is essential for Nox1-4 activity, and p22phox and Nox2 proteins are mutually stabilized in phagocytic cells. This study investigated the suitability of p22phox protein as a marker of Nox activity. To avoid contamination by phagocytic p22phox, we developed global Cybb (encoding Nox2)-knockout mice and analyzed p22phox stability and the expression profiles of Nox protei

Differential Expression of MARK4 Protein and Related Perturbations in Females with Ovulatory PCOS.

BACKGROUND: Ovulatory PCOS (OPCOS) is the mildest form of the polycystic ovarian syndrome among all four determined phenotypes. Though the females with OPCOS are ovulating, hyperandrogenism and polycystic ovarian morphology increase the susceptibility of cardiovascular diseases, insulin resistance, hyperlipidemia and metabolic syndrome in these females. OBJECTIVES: The aim of the study was to identify the significance associated with OPCOS phenotype through serum proteomic profiling of OPCOS females and normal age-matched healthy ovulating females. METHODS: One and two-dimensional gel-based

Comparison of an expanded ataxia interactome with patient medical records reveals a relationship between macular degeneration and ataxia.

Spinocerebellar ataxias 6 and 7 (SCA6 and SCA7) are neurodegenerative disorders caused by expansion of CAG repeats encoding polyglutamine (polyQ) tracts in CACNA1A, the alpha1A subunit of the P/Q-type calcium channel, and ataxin-7 (ATXN7), a component of a chromatin-remodeling complex, respectively. We hypothesized that finding new protein partners for ATXN7 and CACNA1A would provide insight into the biology of their respective diseases and their relationship to other ataxia-causing proteins. We identified 118 protein interactions for CACNA1A and ATXN7 linking them to other ataxia-causing prot

Stable production of peptide antigens in transgenic tobacco chloroplasts by fusion to the p53 tetramerisation domain.

The production of short peptides as single molecules in recombinant systems is often limited by the low stability of the foreign peptide. In the plant expression system this problem has been solved by translational fusions to recombinant proteins that are highly stable or are able to form complex structures. Previously, we demonstrated that the highly immunogenic 21 amino acid peptide 2L21, which is derived from the canine parvovirus (CPV) VP2 protein, did not accumulate in transgenic tobacco chloroplasts. In this report, we translationally fused the 2L21 peptide to the 42 amino acid tetrameri

G proteins as a biochemical tool for diagnosis and monitoring treatments of mental disorders.

There is a significant gap between advances in medication for mental disorders and the present static situation of diagnosis and monitoring treatments of these disorders. Heterotrimeric G proteins play a pivotal role in postreceptor information transduction. These proteins were previously implicated by us in the biochemical mechanism underlying lithium action, and in the pathophysiology of mood disorders. We aimed at quantitatively and functionally evaluating G proteins in patients with major mental disorders in an attempt to unravel a differential pattern of G protein measures characterizing

Immune response to natural and recombinant antigens of Helicobacter pylori in patients with dyspeptic complaints.

Sera of 223 dyspeptic patients with endoscopic findings of nonulcer dyspepsia (72%), gastric ulcer (15%) and duodenal ulcer (13%) were tested for antibodies against Helicobacter pylori with an enzyme immunoassay and an immunoblot technique using lysates of Helicobacter pylori cells as antigen source. One hundred and fifty-one (68%) sera were found to be positive for Helicobacter pylori IgG with both methods; 5% of the positive results in the enzyme immunoassay were false-positive due to cross-reactions mainly of proteins with a molecular mass of 43-66 kDa. Since cross-reactivity not only reduc

CCT8 associates with the MYO10 motor domain and regulates filopodia and breast cancer cell invasion.

Cancer cells utilize filopodia to explore, adhere to and invade their surrounding microenvironment, yet the protein networks that organize these protrusions remain incompletely defined. To uncover the molecular machinery underlying MYO10-positive filopodia, we targeted the fast biotin ligase TurboID to the motor protein MYO10. Proximity biotinylation in two cell types revealed hundreds of potential MYO10 interactors. A targeted microscopy and siRNA screen identified MINK1, SCRIB, CSNK1A1 and CCT8 as new regulators of MYO10-mediated filopodia formation. Focusing on one of the interactors shared

Integrins and tetraspanins mediate CD36-actin coupling and regulate CD36 organization and signaling.

Submembrane cortical actin (CA) plays a large role in regulating the dynamic organization of cell surface receptors, which in turn regulates receptor signaling. Many receptors have short intracellular domains and no known link to actin. Here, we identified the β1-integrin subunit and several tetraspanins (CD9, CD81, and CD151) as part of the hitherto unknown molecular link between the receptor CD36 and CA. Our data indicate that CD36 in vascular endothelial cells interacts with these proteins, with stronger interactions near the cell edge. Compromising these interactions via the point mutation

The role of the follicle-stimulating hormone receptor in supporting male reproduction: A narrative review.

The follicle-stimulating hormone receptor (FSHR) in the testis is expressed exclusively in Sertoli cells. Follicle-stimulating hormone (FSH) is mediated by FSHR via the cyclic adenosine monophosphate (cAMP)/protein kinase A (PKA), mammalian target of rapamycin complex 1 (mTORC1), and AMP-activated protein kinase (AMPK) signaling pathways to induce the proliferation and maturation of immature Sertoli cells. In addition, FSH and androgens regulate spermatogenesis in an overlapping and synergistic manner. Based on these physiological mechanisms, mutations in the FSHβ subunit often lead to azoospe

Isomeranzin activates Gnas-AMPK signaling to drive white adipose browning and curb obesity in mice.

Obesity is a major global health challenge, and promoting the browning of white adipose tissue (WAT) represents a promising therapeutic strategy. However, pharmacological approaches to induce adipose thermogenesis remain limited. Through a Connectivity Map-based screen, we identified isomeranzin (ISM) as a potent small-molecule activator of WAT browning. ISM enhances thermogenesis in adipocytes by activating the AMP-activated protein kinase (AMPK) pathway. Integrated limited proteolysis-mass spectrometry, cellular thermal shift assays, and molecular docking identified guanine nucleotide-bindin

Biological and structural characterization of the Type 3 fimbrial subunit MrkA from Klebsiella pneumoniae.

Klebsiella pneumoniae is a Gram-negative opportunistic pathogen responsible for a wide range of community-associated and hospital-acquired infections and a major cause of neonatal sepsis in low- and middle-income countries. The pathogen's surface fimbriae, particularly the Type 3 fimbriae, are critical for bacterial adhesion, biofilm formation, and host defense evasion. MrkA, the pathogen's major Type 3 fimbrial subunit, has a structural function in fimbrial assembly, but its three-dimensional structure remains to be fully elucidated. In this study, we utilized solution-state nuclear magnetic

A multiomics analysis identifies retinol metabolism in fibroblasts as a key pathway in wound healing.

Impaired wound healing poses a major and increasingly frequent health problem. Among the key players in the healing process are fibroblasts, but their metabolic profile in healing wounds is largely unknown. Using a combination of transcriptomics, targeted proteomics, and metabolomics, we identified retinol metabolism as a top regulated pathway in wound fibroblasts. This is functionally relevant, since even a mild retinol deficiency caused a delay in wound closure and reepithelialization, which mainly resulted from misdirected keratinocyte migration on the new granulation tissue. Quantitative p

Secreted Glycoproteins That Regulate Synaptic Function: the Dispatchers in the Central Nervous System.

Glycoproteins are proteins that contain oligosaccharide chains. As widely distributed functional proteins in the body, glycoproteins are essential for cellular development, cellular function maintenance, and intercellular communication. Glycoproteins not only play a role in the cell and the membrane, but they are also secreted in the intercell. These secreted glycoproteins are critical to the central nervous system for neurodevelopment and synaptic transmission. More specifically, secreted glycoproteins play indispensable roles in neurite growth mediation, axon guiding, synaptogenesis, neurona

Cognitive impairments in type 1 diabetes mellitus model mice are associated with synaptic protein disorders.

An association between type 1 diabetes mellitus (T1DM) and cognitive impairment was recently reported. However, the mechanisms by which T1DM induces cognitive impairment are still unknown. Here, we confirmed that T1DM mice induced by streptozotocin (STZ) injection had impaired working memory and spatial memory. We observed long-term potentiation (LTP) induction defects and synaptic loss in mice 20 weeks after STZ injection. We also found decreased levels of synaptic proteins, including the N-methyl-D-aspartic acid receptor (NMDAR) subunit NR2A, synaptophysin (SYP), and postsynaptic density 95
