In order to clarify the hypotensive mechanisms of alpha-human atrial natriuretic polypeptide (alpha-hANP), hemodynamic responses were investigated in anesthetized rats. Mean blood pressure (MBP), cardiac index (CI) and total peripheral resistance index (TPRI) were measured before and after infusion of alpha-hANP (30 ng/min per 100 g body weight) in intact, nephrectomized, atropinized or chemically sympathectomized rats. Local and systemic capsaicin pretreatment as well cryosurgery induced a long-lasting loss of sensory substance P-immunoreactive nerves in the guinea-pig nasal mucosa. In addition, cryosurgery caused a loss of noradrenergic sympathetic nerves, sclerosis of blood vessels, epithelial damage and fibrosis of the mucosa. The sneezing response to local application of capsaicin--but not that to nicotine--was reduced or abolished by capsaicin pretreatment and cryosurgery, while the response to tactile stimulation was unaffected. Nineteen patients undergoing coronary revascularization (14 patients) or valve replacement (five patients) were studied to monitor the catecholamine levels following operation and for each of three consecutive postoperative days. We determined the number, distribution size, and morphology of paraganglia near the glossopharyngeal, vagus, and sympathetic nerves of rats. The location of paraganglia was revealed by a method that takes advantage of the comparatively high permeability of their blood vessels to Evans blue dye. The effect of N-(2-chloroethyl)-N-ethyl-2-bromobenzylamine (DSP4) on monoamine neurons was studied in mice and rats. The haemodynamic effects of the antiarrhythmic compound, QX 572, have been studied in anaesthetized cats. It was found that QX 572 increased the heart rate and decreased the blood pressure of the cats during the infusion of the drug. A slight increase in blood pressure was seen after the infusion was terminated. The same results have been obtained in patients. The effects of repeated high doses of the adrenergic neuron blocking drug guanethidine or a hexahydropyrazinoindole compound (2-guanyl-1,2,3,10,10a, hexahydro-1,2,a-pyrazinoindole, EMD 21192) (30 mg/kg i.p., 21.5 mg/kg i.p. respectively, equimolar doses) on sympathoadrenal activity were investigated in normotensive adult rats.
