To evaluate the role of oxidative stress and aflatoxin exposure on risk of hepatocellular carcinoma (HCC), a case-control study nested within a large community-based cohort was conducted in Taiwan. Baseline urine samples, collected from a total of 74 incident HCC cases and 290 matched controls, were used to determine by enzyme-linked immunosorbent assays the level of urinary 15-F(2t)-isoprostane (15-F(2t)-IsoP), a biomarker of lipid peroxidation. These samples had been previously analyzed for urinary aflatoxin B(1) (AFB(1)) metabolites and 8-oxo-7,8-dihydro-2'-deoxyguanosine (8-oxodG). Pearson

OBJECTIVES: The prevalence of hepatitis B virus (HBV) is reportedly the main cause of hepatocellular carcinoma (HCC) in India, where hepatitis C virus (HCV)-associated HCC is believed to be relatively less prevalent. We verified the usefulness of alpha-fetoprotein (AFP) as a tumor marker and analyzed the influence of viral etiology on AFP levels in HCC.
METHODS: Of a total of 1012 cases with liver disease, 202 were investigated for the presence of AFP (142 HCC cases, 30 cirrhosis cases, and 30 chronic liver disease (CLD) cases). In addition, serum samples from 30 healthy patients, 30 hepatiti

Aflatoxin B(1) (AFB(1)) is a fungal toxin that causes both acute hepatotoxicity and hepatocellular carcinoma in humans and experimental animals. Previous studies demonstrated that a small, noninjurious dose of bacterial lipopolysaccharide (LPS) augments the hepatotoxicity of AFB(1) through activation of inflammatory cells and production of soluble inflammatory mediators (Barton et al., 2000b, 2001). This study was conducted to examine the effect of LPS on the dose-response relationship for AFB(1)-induced liver injury. Male Sprague-Dawley rats (250-350g) were treated with AFB(1) (0.1 mg/kg-6.3

Weanling rats were kept on a synthetic vitamin A free diet for 9 weeks, and subsequently on the same diet but with oral supplementation of vitamin A palmitate at different doses for 2-3 weeks. The ability of liver microsomes to catalyze reactions of aflatoxin B1 (AFB1) leading to its activation and DNA adduct formation was measured after each period of experimental feeding. An increased molecular reactivity of AFB1 was evident during vitamin A deficiency such that the degree of its conversion to its reactive metabolite and subsequent DNA adduct formation was higher with microsomes from vitamin

Aflatoxins are potent carcinogenic and genotoxic secondary metabolites from fungi, posing significant health risks. This study aimed to detect the presence of aflatoxins (AFB1, AFB2, AFG1, AFG2, and AFT-total aflatoxins) in rice and rice-derived products, including puffed and flattened rice, from different local markets of Dhaka using HPLC with fluorescence detection, and to estimate health risks. AFT was detected in two periods, with a higher prevalence in period I (Rice: 29.55%, Flattened-rice: 60%) than in period II (Rice: 25%, Flattened-rice: 50%), whereas no aflatoxin-positive samples wer…

Aflatoxin M1 (AFM1), the 4-hydroxylated metabolite of aflatoxin B1, is excreted in the milk of lactating animals fed AFB1-contaminated feed and is of public-health concern because of its well-documented thermal stability during dairy processing. In this study, a matrix-specific extraction, immunoaffinity clean-up, and HPLC-fluorescence detection method were developed and validated for AFM1 in khoa, a traditional heat-desiccated Indian dairy product. The method was linear (R2 ≥ 0.999) over 0.25-20 µg kg- 1, with satisfactory recoveries (83%-102%) and precision (RSD ≤ 6.4 %). Experimentally veri

A dual-emission, ratiometric fluorescence sensor is reported based on a europium(III)-doped metal-organic framework (Eu/DUT-52) that exhibits blue emission from DUT-52 and red emission from Eu³⁺. Notably, we fond that aflatoxin B1 (AFB1) enhances the fluorescence intensity of DUT-52, whereas the Eu³⁺ emission remains essentially unchanged, enabling ratiometric detection with a concentration-dependent color change from pink to purple. We further investigated the origin of this fluorescence enhancement. The ratiometric response provides a detection limit of 3.9 ng mL⁻¹. For field deployment, Eu/

BACKGROUND: Aflatoxin M1 (AFM1), a highly toxic hydroxylated metabolite of aflatoxin B1, is a major contaminant of concern in milk and dairy products, posing significant risks to food safety and public health. Since AFM1 contamination is closely associated with feed quality, storage conditions, climatic factors, and farm management, reliable AFM1 detection is important for supporting quality control and safety assessment of dairy products. However, the determination of trace AFM1 in dairy matrices remains challenging, as complex matrix components may compromise analyte recognition specificity

Peanut oil is prone to contamination with aflatoxin B1 (AFB1). Traditional ultraviolet (UV) irradiation methods demonstrate limited effectiveness in detoxification and may compromise the oil quality. To address this, this study developed a novel UV LED cold light source temperature control system, offering a new solution for AFB1 detoxification in peanut oil production. On the basis of this system, peanut oil samples spiked with varying concentrations of AFB1 were prepared and treated under different operational conditions. Then, the AFB1 content, acid value, and peroxide value of the decontam

Porcine gastric epithelial cells (PGECs) serve as a valuable model for studying the molecular and pathogenic mechanisms of the stomach. However, PGECs face limitations such as isolation challenges, short lifespan, and restricted proliferation. To address this, we established an immortalized PGECs (i-PGECs) to enable in vitro investigation of pathogen infection mechanisms. Primary PGECs were isolated from the acid-secreting glands using stepwise digestion with multiple enzymes (dispase II/collagenase I/hyaluronidase). Immortalization was achieved via lentiviral vectors expressing simian virus 4

Aflatoxin B1 (AFB1) contamination and micronutrient deficiencies pose a major challenge to food safety and nutritional security. This study elucidated the synergistic potential of boron (B) and zinc (Zn) co-application to fortify nutritional quality and mitigate AFB1 accumulation in turnip (Brassica rapa L) roots. Seeds were cultivated in soils amended with individual or combined B and Zn at concentrations of 10–25 mg kg− 1. In results, individual B supplementation at 20 mg kg− 1 optimized protein content (10.9%), and the B-Zn interactome provided superior overall metabolic performance. B-Zn s

Aspergillus ear rot (AER) and aflatoxin contamination in maize threaten global food security and public health, with developing countries such as Bangladesh-where maize cultivation is rapidly expanding-being particularly vulnerable. This study evaluated the resistance of 25 commercially available maize hybrids to AER infection and aflatoxin accumulation under natural infection (field exposure without artificial inoculation) and artificial inoculation conditions. Hybrids were artificially inoculated using a toxigenic Aspergillus flavus strain (ES103) via the pin bar inoculation method to ensure

Aflatoxin B1 (AFB1) and patulin (PAT) are prevalent foodborne mycotoxins with hepatotoxic potential, but the hepatic effects of combined exposure remain largely unclear. This study investigated the hepatotoxic consequences of co-exposure to AFB1 and PAT using no-observed adverse effect levels (NOAELs) in C57BL/6 mice and low-cytotoxic concentrations in HepG2 cells selected by viability screening. Mice and cells were assigned to four groups: control, AFB1, PAT and AFB1 + PAT. Exposure to either toxin individually did not cause evident liver injury, whereas co-exposure significantly elevated ala

BACKGROUND: Aflatoxin B1 (AFB1) is a potent group 1 carcinogen closely associated with hepatocellular carcinoma (HCC), particularly in regions with high dietary exposure and concurrent hepatitis B virus infection. However, the molecular mechanisms by which AFB1 promotes HCC progression remain incompletely understood, and there is a lack of prognostic models specifically tailored to AFB1-associated HCC. Integrating bioinformatics and network toxicology approaches may help identify key genes and construct reliable predictive tools for this unique subtype of liver cancer. This study aims to ident

BACKGROUND: Peanut meal, a high-protein agricultural by-product, faces challenges as animal feed due to anti-nutritional factors, poor protein digestibility, aflatoxin contamination, and imbalanced amino acids. Microbial fermentation is one of the most effective methods to reduce anti-nutritional factors and enhance the nutritional value of peanut meal. Compared to single-strain fermentation, microbial-enzyme co-fermentation exhibits enhanced degradation efficiency, accelerates nutrient release, improves product safety by reducing mycotoxins and anti-nutritional factors, enhances sensory prope

BACKGROUND: Of the mycotoxins, aflatoxin is the most significant. The detection of aflatoxin B1 (AFB1) is crucial for ensuring food safety, as this highly carcinogenic toxin readily contaminates crops such as grains and nuts, and timely detection can effectively prevent associated health risks. The selection of luminophores is of paramount importance in the detection of ECL (electrochemiluminescence). Tetraphenylethylene (TPE) and its derivatives are the most representative molecules in aggregation-induced electrochemiluminescence. The energy resonance transfer phenomenon in ECL-RET (electroch
