Telomere shortening in alcohol dependence: Roles of alcohol and acetaldehyde.

Heavy drinking leads to premature aging and precipitates the onset of age-related diseases. Acetaldehyde (AcH), a toxic metabolite of ethanol, has been implicated in various types of cancer. However, whether alcohol accelerates biological aging at a cellular level is controversial and the mechanism involved is unclear. We addressed these questions by measuring telomere length (TL) in peripheral blood leukocytes of Japanese patients with alcohol dependence (AD) and examined the association between TL, genetic variants of alcohol dehydrogenase (ADH)1B and aldehyde dehydrogenase (ALDH)2, and othe

Fetal Alcohol Syndrome and Fetal Alcohol Spectrum Disorders.

Fetal alcohol syndrome (FAS) and fetal alcohol spectrum disorders (FASD) result from intrauterine exposure to alcohol and are the most common nonheritable causes of intellectual disability. The percentage of women who drink or binge drink during pregnancy has increased since 2012. FAS is commonly missed or misdiagnosed, preventing affected children from receiving needed services in a timely fashion. Diagnosis is based on the presence of the following clinical features, all of which must be present: prenatal and/or postnatal growth retardation, facial dysmorphology, central nervous system dysfu

Contribution of liver alcohol dehydrogenase to metabolism of alcohols in rats.

The kinetics of oxidation of various alcohols by purified rat liver alcohol dehydrogenase (ADH) were compared with the kinetics of elimination of the alcohols in rats in order to investigate the roles of ADH and other factors that contribute to the rates of metabolism of alcohols. Primary alcohols (ethanol, 1-propanol, 1-butanol, 2-methyl-1-propanol, 3-methyl-1-butanol) and diols (1,3-propanediol, 1,3-butanediol, 1,4-butanediol, 1,5-pentanediol) were eliminated in rats with zero-order kinetics at doses of 5-20 mmol/kg. Ethanol was eliminated most rapidly, at 7.9 mmol/kgh. Secondary alcohols (2

Colchicine for alcoholic and non-alcoholic liver fibrosis and cirrhosis.

BACKGROUND: Alcohol and hepatotropic viruses cause the majority of liver cirrhosis cases in the Western World. Colchicine is an anti-inflammatory and anti-fibrotic medication. Several randomised clinical trials have addressed the question whether colchicine has any efficacy in patients with alcoholic or non-alcoholic fibrosis and cirrhosis. OBJECTIVES: To assess the beneficial and harmful effects of colchicine in patients with alcoholic or non-alcoholic fibrosis or cirrhosis, excluding primary biliary cirrhosis. SEARCH STRATEGY: The Cochrane Hepato-Biliary Group Controlled Trials Register, T

A comparison of alcohol-positive and alcohol-negative trauma patients.

OBJECTIVE: Hospital admission for an alcohol-related traumatic injury may offer a "teachable moment" to address a patient's alcohol problem. Although trauma teams provide a number of other health-related services, there may be characteristics of alcohol-positive victims that act as barriers toward providing alcohol counseling. The purpose of this study was to compare the characteristics and hospital outcomes of trauma patients who tested positive for alcohol at the time of hospital admission with those who did not. This information is useful for planning interventions and referrals for treatme

Acamprosate and alcohol: II. Effects on alcohol withdrawal in the rat.

The suppressing effect of acamprosate (calcium-acetyl homotaurinate) on alcohol drinking is well established; however, little is known about its effects upon the alcohol-induced withdrawal syndrome. Male Wistar rats received as a sole drinking fluid a 20% (v/v) alcohol solution for one week. Animals consumed on average 5.3 +/- 0.3 g/kg per day alcohol, which resulted in blood alcohol levels of 38 +/- 14 mg/dl. For the quantification of alcohol withdrawal we used a new radio-telemetric system which enabled us to monitor body temperature, locomotor activity, food and water intake patterns consta

Acamprosate and alcohol: III. Effects on alcohol discrimination in the rat.

It has been shown that acamprosate (calcium-acetyl homotaurinate) decreases voluntary ethanol drinking in laboratory animals as well as relapse behaviour in human alcoholics. Although glutamatergic mechanisms have been implicated in several recent studies in the action of acamprosate, the basic mechanism remains unknown. In order to gain more insight into possible mechanisms underlying the ethanol intake-suppressing effects of acamprosate, we investigated this compound in an ethanol discrimination test. Male Wistar rats were trained in a two-lever operant drug discrimination paradigm to make d…

[A new view of the evaluation of alcohol withdrawal and alcohol dependence].

This study was designed to evaluate 21 alcohol dependent male individuals during withdrawal syndrome. The intensity of withdrawal symptoms was measured using Sandowal-Wang scale. The level of blood serum prolactin was measured twice a day (8 a.m. and 8 p.m.) on the lst, 3rd, 7th, 14th, 21st day of the study. Also the depth and the time of the development of dependence were evaluated using DSM-III-R criteria, alcohol dependence (WGU) and presence of dependence symptoms scale (WWO). It was establish that WGU, WWO and DSM-III-R criteria, separated similar groups of patients with the same depth of

Assessment of family history of alcoholism in sons of alcoholic fathers.

Results of studies of sons of alcoholics (SOAs) may vary depending on the density of the family history of alcoholism of the SOA subjects selected. To assess how reliably SOA subjects report a history of alcoholism in first- and second-degree relatives, we compared family histories obtained from 20 SOAs with those provided by their fathers. In all 20 cases, SOAs and their fathers agreed that the father met criteria for alcohol dependence but for no other primary Axis I psychiatric disorder. However, agreement was not as good for whether other paternal relatives in addition to the father were a…

Low level of response to alcohol as a predictor of future alcoholism.

OBJECTIVE: As part of a search for measurable attributes of an individual that might be related to the risk of alcoholism, the author's group previously compared 227 sons of alcoholics and 227 matched comparison subjects at the age of about 20 years. Forty percent of the men at high risk for alcoholism and less than 10% of the comparison subjects demonstrated a low intensity of response to alcohol challenge. This article reports the results of the follow-up of the first half of this study group almost a decade later. METHOD: Of the men who had been tested at about age 20, 223 were about age 3…

Chronic alcoholic gastritis. Roles of alcohol and Helicobacter pylori.

We assessed the relative roles of alcohol and infection with Helicobacter pylori in the pathogenesis of chronic gastritis in alcoholic patients. Helicobacter pylori was found in 14 of 18 alcoholics with dyspepsia and was associated with chronic antral gastritis. Gastric biopsy specimens were normal in four H pylori-negative alcoholics. Studies were repeated 3 to 4 weeks after controlled abstinence. There was no change in histologic findings during this period, indicating that alcohol itself was not the major causative agent. We then eliminated H pylori in 10 subjects by giving triple therapy (…

The nature of alcoholism in patients with different family histories for alcoholism.

1. The course and consequences of alcohol abuse were examined in male and female patients being treated for alcoholism. 2. Demographic characteristics, family history for alcoholism, psychopathology, drinking history and social and physical consequences of alcohol abuse were assessed. 3. Probands with alcoholism on both the maternal and paternal sides of their pedigree experience greater psychosocial and physical consequences of alcohol abuse than other alcoholic patients. 4. Sex of the proband and type of family pedigree for alcoholism were found not to be interactive but to contribute separa

[Liver damage due to alcohol and acute alcoholic hepatitis].

In 15 patients suffering from slight forms of alcohol-induced liver damage, the clinical picture was so varied that correct diagnosis was only possible on the basis of all anamnestic, clinical, biochemical and histological data. From the histopathological viewpoint, the forms presented as simple hepatic steatosis or acute inflammatory lesion (alcoholic hepatitis). Clinical and histological signs are reversible if the patient stops drinking, whereas if he continues to consume alcohol the prognosis is much more serious. This is why it is important to make correct diagnosis even in slight forms o

Severe alcoholic cardiomyopathy reversed with abstention from alcohol.

In a chronic alcoholic with a severe congestive cardiomyopathy the angiographic and hemodynamic evidence of left ventricular dysfunction was completely reversed after 1 year of abstinence. At the time of initial presentation two of the traditional hallmarks of a poor prognosis were present, namely, a long exposure to alcohol before the onset of symptoms and severe diffuse left ventricular dilation and hypokinesis (ejection fraction 14.9 percent). Yet, today, 18 months later he is asymptomatic, receiving no medications and has normal left ventricular function. A reexamination of prognostic fact
