Transforming growth factor-beta in calcium alginate beads for the treatment of articular cartilage defects in the rabbit.

PURPOSE: Articular cartilage has only limited capability for intrinsic repair. The use of growth factors has been suggested to improve the repair of cartilage after injury. Reliable delivery systems for these agents are needed. In this study we tested calcium alginate for the delivery of TGF-beta in the treatment of osteochondral defects in the rabbit knee. TYPE OF STUDY: Randomized trial animal study and basic science study. METHODS: In vitro, to establish the kinetics of TGF-beta release from the alginate, 125I- labeled TGF-beta was suspended in 1.2% sodium alginate at concentrations of 1

Reconstruction of rat peripheral nerve gap without sutures using freeze-dried alginate gel.

Many materials have been used for artificial tubular prostheses to assist peripheral nerve gap reconstruction. However, the clinical use of these devices has been restricted because a microsurgical procedure requires specialized techniques and expensive equipment, such as operating microscope systems. Therefore the authors developed a new gluing method, without sutures, that uses freeze-dried alginate gel. A 7-mm gap in the sciatic nerve of rats was bridged with freeze-dried alginate gel. Regeneration was evaluated by electrophysiologic testing and histologic study. Eighteen weeks after surger

Comparison of a dimethicone/antacid (Asilone gel) with an alginate/antacid (Gaviscon liquid) in the management of reflux oesophagitis.

Fifty-three patients with symptomatic reflux oesophagitis were entered into a single centre randomized study comparing the effects of a dimethicone/antacid (Asilone Gel) and an alginate/antacid (Gaviscon liquid) on symptoms and endoscopic changes over an 8-week period. Both treatments significantly improved heartburn, acid regurgitation and flatulence. Dimethicone/antacid but not alginate/antacid, produced a significant improvement in oesophagitis, oesophageal ulceration and histological grade of inflammation over the 8-week treatment period so that 14 patients treated with dimethicone/antacid

Mild-temperature conjugation of pea protein isolate with oxidized sodium alginate: structural, interfacial, and emulsion properties.

Pea protein isolate (PPI) has limited food applications because of poor aqueous solubility and emulsifying performance. Here, sodium alginate (SA) was oxidized to generate reactive aldehyde groups, enabling Schiff-base-mediated conjugation with PPI under mild conditions (40 °C, 90 min). By varying reaction parameters, conjugates with grafting degrees (GD) of 10.67-22.87% were obtained. Increasing GD was accompanied by increased β-sheet content (32.81-41.52%), tertiary structural reorganization, enhanced surface charge, a water contact angle of up to 79.35°, and a > 50% reduction in oil-water i

Bipartite Anchoring at the +3 Subsite Governs Trisaccharide Specificity in a PL7 Alginate Lyase and Is Transferable to a Homologous Enzyme.

Alginate lyases produce alginate oligosaccharides (AOS) whose bioactivities depend on the degree of polymerization (DP). Loop regions and individual residues influence product DP. However, how residues from loops cooperate at subsites to enforce a defined DP remains unresolved. Here, we identified ALG4, a PL7_5 alginate lyase specifically producing trisaccharides (ΔDP3), and ALG3, a same-subfamily homologue generating ΔDP2-ΔDP4 mixtures. Action-pattern analysis showed that ALG4 accommodates three sugar residues at the +1/+2/+3 subsites without extension toward +4. Structurally, the Orange Loop

Inhibition effects and binding behavior of alginate oligosaccharides on tyrosinase and elastase: multi-spectroscopic approaches and zebrafish evaluation.

Alginate oligosaccharides (AOSs) are natural marine oligosaccharides with diverse physiological functions, which have broad application prospects in both the cosmeceutical and food industries. This study systematically investigated the inhibitory mechanisms of AOSs against tyrosinase and elastase, two key enzymes associated with skin aging, by combining multi-spectroscopic approaches with zebrafish in vivo evaluation. Results demonstrated that AOSs acted as a reversible mixed-type inhibitor for both enzymes, exhibiting moderate inhibitory activities with IC50 values of 7.37 ± 0.15 mM for tyros

Turmeric essential oil infused pectin blended sodium alginate polymer as sustainable food packaging material.

Eco-friendly, renewable materials have been pushed in many applications to replace non-renewable resources in recent years. In the present work, a similar attempt was made to replace synthetic polymer-based food packaging films with films made out of waste-derived renewable materials. In this study, pectin (P) and sodium alginate (SA) polymers were blended with turmeric essential oil (O) to increase the functionality of the film. Turmeric essential oil was chosen over other plant based essential oils due to its potential antioxidant, antifungal and broad-spectrum antimicrobial activity. Calciu

This study aimed to evaluate the printability and biological performance of an alginate-carrageenan blend for applications in tissue bioengineering. A preliminary screening of different polymer proportions (1, 2, 3, 4, and 5% w/v) was conducted to assess diffusion rate, filament dispersion, and overall printability. The optimal concentration was then characterized rheologically and used to fabricate 3D constructs with an EnvisionTEC 3D Bioplotter. Scaffold morphology was examined using scanning electron microscopy (SEM). For biological evaluation, bovine ovaries (n = 2) were obtained from a lo

Colon-targeted alginate/chitosan microspheres for co-delivery of synbiotics and nanozymes ameliorate inflammatory bowel disease.

Inflammatory bowel disease (IBD) is closely associated with gut dysbiosis and immune imbalance. Synbiotics, combining probiotics and prebiotics, represent a promising dietary strategy; however, their therapeutic efficacy is often limited by poor gastrointestinal stability and insufficient colon delivery. In this study, we developed a colon-targeted delivery system based on alginate/chitosan microspheres for the co-delivery of L. acidophilus, prebiotic stachyose, and a redox-active component. The formulated system exhibited excellent stability under simulated gastrointestinal conditions and eff

Sodium Alginate Hydrogel with Zinc Ion Nanoparticles for Synergistic Neuroprotection and Functional Recovery in Spinal Cord Injury.

The current lack of effective treatments for traumatic spinal cord injury (SCI) presents a significant challenge in managing the complex microenvironmental alterations that follow the initial trauma. This study developed an injectable alginate hydrogel dynamically cross-linked by tannic acid-zinc nanoparticles (TA@Zn NPs), which exerts neuroprotective effects through the sustained release of zinc ions (Zn2+) and antioxidant TA@Zn NPs. TA@Zn NPs were cross-linked with phenylboronic acid-modified sodium alginate (SA) to form an injectable gel system. In response to the acidic and ROS-rich microe

Hierarchical alginate-bentonite beads enable instrument-free pre-analytic enrichment of liter-scale wastewater.

Wastewater-based surveillance (WBS) offers a test-independent window into public community health, but its fidelity depends critically on pre-analytic enrichment that must convert liter-scale, inhibitor-rich influent into microliter analytical inputs without compromising low-titer targets. Here we introduce hierarchical alginate-sulfuric acid-activated bentonite beads (Alg@SAB) that recast pre-analytic enrichment as an interfacial mass-transfer problem. A diffusion-optimized macro-mesoporous scaffold, amine-functionalized surfaces (-NH₃⁺), and Ba²⁺ crosslinks collectively enable dual-mode capt

Alginate-butterfly pea microbeads as anthocyanin-based colorimetric sensors for biogenic amines and chicken spoilage monitoring.

Alginate microbeads incorporating blue butterfly pea (Clitoria ternatea L.) extract were developed as natural, pH-responsive colorimetric indicators for monitoring food spoilage. The blue butterfly pea extract, rich in anthocyanins, exhibited distinct pH-dependent color transitions from red (pH 2) to green (pH 10), supported by UV-Vis bathochromic shifts. Structural analyses revealed the successful incorporation of blue butterfly pea extract into the alginate matrix, as demonstrated by SEM images that showed extract addition produced smoother bead morphologies compared to plain alginate beads.

Alginate-PVA interpenetrating network nanogels: a pH-responsive carrier for thermal stabilization and sustained release of phycocyanin.

Phycocyanin (PC) is a sensitive bioactive protein that suffers from poor thermal stability and premature release in conventional alginate (Alg) hydrogels. In this study, Alg-polyvinyl alcohol (PVA) interpenetrating polymer network (IPN) nanogels were developed as a pH-responsive carrier for improved PC stabilization and controlled oral delivery. Increasing PVA concentration produced nanogels with particle sizes ranging from 174.01 ± 2.66 to 255.02 ± 68.04 nm and significantly improved encapsulation efficiency from 69.59 ± 0.51% to 92.20 ± 0.84%, with a maximum loading capacity of 8.24 ± 0.06%.
