Iron overload involved in the enhancement of unloading-induced bone loss by hypomagnetic field.

During deep-space exploration missions, astronauts will be exposed to abnormal space environments including microgravity and hypomagnetic field (HyMF) that is 10,000 times weaker than geomagnetic field (GMF). It is well known that microgravity in space can induce bone loss; however, it is ill-defined whether HyMF involved in this process. Herein, we aimed to investigate the combined effects of HyMF and microgravity on bone loss. A mouse model of hindlimb suspension (HLU) was adopted to simulate microgravity-induced bone loss, that was exposed to a hypomagnetic field of <300 nanotesla (nT) gene…

Eye movements and motion perception during off-vertical axis rotation after spaceflight.

Constant velocity off-vertical axis rotation (OVAR) provides dynamic linear acceleration stimuli that can be used to assess otolith function. Eight astronauts were rotated in darkness about their longitudinal axis 20° off vertical at low (0.125 Hz) and high (0.5 Hz) frequencies and their responses were compared before and after spaceflight. Eye movements were recorded using infrared videography and perceived motion was evaluated using a joystick with four degrees of freedom - pitch and roll tilt, front-back and lateral translation. Low-frequency OVAR generates tilt otolith-induced responses - …

Hyperbaric oxygen pre-breathe modifies the outcome of decompression sickness.

Deep sea divers, aviators and astronauts are at risk of decompression sickness when the ambient pressure reductions exceed a critical threshold. Venous bubbles associated with decompression sickness have the potential to react with the vascular membrane and adjacent blood products, eliciting an inflammatory cascade. Preventive measures usually involve careful decompression procedures to avoid or reduce bubble formation. De-nitrogenation with 100% oxygen pre-breathing as a preventive measure has been well established at least in altitude decompression exposures. The objective of this study was …

Hearing and performance during a 70-h exposure to noise simulating the space station environment.

INTRODUCTION: Elevated hearing thresholds have been documented in some astronauts after long-term spaceflights although noise levels were lower than those normally associated with noise-induced hearing loss in ground-based operations. The present study was conducted to determine whether prolonged exposure (70 h) to levels (72 dBA) recorded on the International Space Station (ISS) service module would impact diverse measures of auditory function, as well as cognition and memory, motivation, and cardiovascular function. METHOD: Five mixed gender subgroups of five normal-hearing subjects, aged 2…

Developing protocols for recombinant adeno-associated virus-mediated gene therapy in space.

With the advent of the era of International Space Station (ISS) and Mars exploration, it is important more than ever to develop means to cur … ply to other disorders as well. For example, the procedure may become directly relevant to treating astronauts for space-anemia, immune suppression and bone marrow derived tumors, e.g. leukemia. The adeno-associated virus serotype 2 (AAV2) is a non-pathogenic human parvovirus with broad host-range and tissue specificity. Exploiting these characteristics we h …

AstroOphthalmology: Basic and Pathophysiological Concepts of Space-Induced Ocular Changes.

Long-duration space missions pose a considerable risk to eye health, and visual dysfunction is recognized as a primary health concern for astronauts. Hence, we have developed a series of review reports to study the effects of space on the eye, called AstroOphthalmology (including Basics, Ocular Anterior Segment, Ocular Posterior Segment). This report serves as the first report of our series, and integrates the current insights into the pathophysiological consequences of the space environment on the eye. The two key space-specific stressors include microgravity and cosmic radiation. Both stress…

Space Physiology and Technology: Adaptations, Countermeasures, and Opportunities for Wearable Systems.

Space poses substantial challenges for humans, leading to physiological adaptations in response to an environment vastly different from Earth. A comprehensive understanding of these physiological adaptations is necessary to develop effective countermeasures that support human life in space. This narrative review first focuses on the impact of the space environment on the musculoskeletal system. It highlights the complex interplay between bone and muscle adaptations and their implications on astronaut health. Despite advances in current countermeasures, such as resistive exercise and pharmacolo

CB001 bioactive compounds mitigate intestinal immune dysregulation induced by simulated microgravity and radiation via B cells modulation.

BACKGROUND: Gastrointestinal discomfort is a prevalent issue during spaceflight, yet targeted therapeutic strategies remain elusive. Existing research largely relies on single-factor modeling, limiting a comprehensive understanding of the underlying pathologies. METHODS: We established a composite mouse model combining simulated microgravity (hindlimb unloading) and low-dose radiation (Cobalt-60) to evaluate intestinal injury and the therapeutic potential of CB001. Mechanisms were investigated through histological analysis, in vitro co-culture assays, and flow cytometry assessment of B-cell d

Effects of long-term simulated microgravity on liver metabolism in rhesus macaques.

The liver is an essential multifunctional organ and constantly communicates with nearly all the tissues in the body. Spaceflight or simulated microgravity has a significant impact on the livers of rodent models, including lipid accumulation and inflammatory cell infiltration. Whether similar liver lipotoxicity could occur in humans is not known, even though altered circulating cholesterol profile has been reported in astronauts. Using a 42-day head-down bed rest (HDBR) model in rhesus macaques, the present study investigated whether simulated microgravity alters the liver of non-human primates…

Protection against neurodegenerative disease on Earth and in space.

All living organisms have evolutionarily adapted themselves to the Earth's gravity, and failure to adapt to gravity changes may lead to pathological conditions. This perspective may also apply to abnormal aging observed in bedridden elderly patients with aging-associated diseases such as osteoporosis and sarcopenia. Given that bedridden elderly patients are partially analogous to astronauts in that both cannot experience the beneficial effects of gravity on the skeletal system and may suffer from bone loss and muscle weakness, one may wonder whether there are gravity-related mechanisms underly…

Characterization of metabolic changes and adaptive remodeling during a hypobaric hypoxic exploration atmosphere environment.

INTRODUCTION: The International Space Station maintains normoxic atmospheric conditions of 14.7 psia (101 kPa) and 21% oxygen. Astronauts don spacesuits to conduct extravehicular activities (EVAs), which operate at a reduced pressure of 4.3 psia (29.6 kPa), and a hyperoxic atmosphere of 100% oxygen. The reduction in pressure presents a risk of decompression sickness and is mitigated by a several hour denitrogenation (aka, prebreathe) protocol. To reduce the time required for prebreathe, future lunar and planetary exploration habitats and vehicles are expected to maintain a hypobaric, mildly hy…
