Microbes in gynecologic cancers: Causes or consequences and therapeutic potential.

Gynecologic cancers, starting in the reproductive organs of females, include cancer of cervix, endometrium, ovary commonly and vagina and vu … le in blocking adhesions of incursive pathogens to vaginal epithelium by lowering the pH, producing bacteriocins and employing competitive exclusions. The optimum or personalized balance of the microbiota can be maintained using pre- and probiotics, and fecal microbiota transplantations loaded with specific bacteria. Current evidence strongly suggest that a he …

The role of the gut microbiota in the pathology and prevention of liver disease.

Several microorganisms belonging to the intestinal microbiota act in an ecosystem responsible for maintaining the homeostasis and vital func … une system. Also, the use of probiotics prevents harmful bacteria and their derived products (e.g., bacteriocins, endotoxins, hydrogen sulfide, etc.) to leak through the intestinal wall to the circulation that results in the activation of signaling pathways that may be implicated in liver disease. The liver receives a constant flow of noxious entities that pro …

Pervasive domestication of defective prophages by bacteria.

Integrated phages (prophages) are major contributors to the diversity of bacterial gene repertoires. Domestication of their components is th … gene repertoires compatible with described functions of adaptive prophage-derived elements such as bacteriocins, killer particles, gene transfer agents, or satellite prophages. We suggest that bacteria frequently domesticate their prophages. Most such domesticated elements end up deleted from the bacterial genome because they are replaced by analogous functio …

FhuA-SbmA-driven resistance pathway to microcin Y triggers transcriptomic reprogramming and virulence enhancement in Salmonella Typhimurium.

Salmonella Typhimurium poses critical safety risks owing to its multidrug resistance and persistence in food matrices. To address this threat, we focused on microcin Y (MccY) a lasso peptide with potent activity against multidrug-resistant S. typhimurium and elucidated how mutations in the FhuA/SbmA receptors mediate MccY resistance and differentially affect bacterial virulence. Molecular docking revealed that MccY tended to bind to the N-terminal segment of FhuA within its β-barrel cavity, with key interaction sites including FhuA564Leu-MccY5His (2.5 Å hydrogen bond, MIC = 200 μg/mL) and site

Two highly divergent lineages of exfoliative toxin B-encoding plasmids revealed in impetigo strains of Staphylococcus aureus.

Exfoliative toxin B (ETB) encoded by some large plasmids plays a crucial role in epidermolytic diseases caused by Staphylococcus aureus. We have found as yet unknown types of etb gene-positive plasmids isolated from a set of impetigo strains implicated in outbreaks of pemphigus neonatorum in Czech maternity hospitals. Plasmids from the strains of clonal complex CC121 were related to archetypal plasmid pETBTY4. Sharing a 33-kb core sequence including virulence genes for ETB, EDIN C, and lantibiotics, they were assigned to a stand-alone lineage, named pETBTY4-based plasmids. Differing from each

Novel antimicrobial peptide specifically active against Porphyromonas gingivalis.

Porphyromonas gingivalis, the major etiologic agent of chronic periodontitis, produces a broad spectrum of virulence factors, including outer membrane vesicles, lipopolysaccharides, hemolysins and proteinases. Antimicrobial peptides (AMPs) including bacteriocins have been found to inhibit the growth of P. gingivalis; however, these peptides are relatively large molecules. Hence, it is difficult to synthesize them by a scale-up production. Therefore, this study aimed to synthesize a shorter AMP that was still active against P. gingivalis. A peptide that contained three cationic amino acids (Arg…

[Therapeutic effectiveness of ristomycin in the visceral form of actinomycosis (experimental research)].

Therapeutic efficacy of ristomycin in visceral actinomycosis, a severe disease difficult for diagnosis requiring long-term complex therapy was studied on 24 rabbits. The animals were divided into 4 groups. A model of thoracal actinomycosis was developed. 7 rabbits were treated with ristomycin. The other 7 rabbits were treated with ristomycin in combination with actinolysate, a specific immune preparation. 5 rabbits were treated with actinolysate alone and the other 5 rabbits received no treatment. alone and especially in combination with actinolysate was an efficient therapeutic agent for trea

Genomic and pan-genomic analyses of Bacillus subtilis B13 provide insights into biosynthetic potential and genetic traits associated with environmental adaptation.

This study describes the genomic features of Bacillus subtilis B13 (= VTCC 910231) to elucidate the genetic basis underlying its reported antimicrobial activity, metabolic versatility, and environmental adaptability. The draft genome comprised 4,349,051 bp with a GC content of 43.5% and 4436 predicted coding sequences. Genome-based analyses assigned B13 to B. subtilis subsp. subtilis, supported by high average nucleotide identity (98.3%) and digital DNA-DNA hybridization (99.7-99.8%) values. Genome mining identified one gene cluster encoding an unidentified sactipeptide along with seven biosyn

Molecular Lung Imaging Following Exposure to Radiation Predicts Long-Term Survival in Rats.

Delayed effects of acute radiation exposure (DEARE), including radiation pneumonitis (lung-DEARE), develop weeks to months after radiation exposure. Pathway-targeted biomarkers that capture early oxidative stress and cell death could improve risk stratification and provide objective measures of mitigator efficacy. The objective was to test whether molecular lung imaging predicts long-term survival and mitigator response after irradiation. Rats received 13.5 Gy leg-out partial-body irradiation with a subset treated with the radiation-injury mitigator lisinopril. Rats underwent lung imaging at w

Microcins mediate competition among Enterobacteriaceae in the inflamed gut.

The Enterobacteriaceae are a family of Gram-negative bacteria that include commensal organisms as well as primary and opportunistic pathogens that are among the leading causes of morbidity and mortality worldwide. Although Enterobacteriaceae often comprise less than 1% of a healthy intestine's microbiota, some of these organisms can bloom in the inflamed gut; expansion of enterobacteria is a hallmark of microbial imbalance known as dysbiosis. Microcins are small secreted proteins that possess antimicrobial activity in vitro, but whose role in vivo has been unclear. Here we demonstrate that mic

[Origin of the plague microbe Yersinia pestis: structure of the process of speciation].

The origin and evolution of the plague microbe Yersinia pestis are considered in the context of propositions of modern Darwinism. It was shown that the plague pathogen diverged from the pseudotuberculous microbe Yersinia pseudotuberculosis O:1b in the mountain steppe landscapes of Central Asia in the Sartan: 22000-15000 years ago. Speciation occurred in the tarbagan (Marmota sibirica)--flea (Oropsylla silantiewi) parasitic system. The structure of the speciation process included six stages: isolation, genetic drift, enhancement of intrapopulational polymorphism, the beginning of pesticin synth

Sensitization of microcin J25-resistant strains by a membrane-permeabilizing peptide.

Microcin J25 (MccJ25) is a plasmid-encoded, 21-amino-acid, antibacterial peptide produced by Escherichia coli. MccJ25 inhibits RNA polymerase and the membrane respiratory chain. MccJ25 uptake into E. coli-sensitive strains is mediated by the outer membrane receptor FhuA and the inner membrane proteins TonB, ExbB, ExbD, and SbmA. This peptide is active on some E. coli, Salmonella, and Shigella species strains, while other Gram-negative bacteria, such as clinical isolates of Enterobacter cloacae, Citrobacter freundii, Klebsiella pneumoniae, Pseudomonas aeruginosa, Acinetobacter baumannii, Moraxe

Discovery of structurally diverse natural product antagonists of chemokine receptor CXCR3.

The chemokines (CXCL9, CXCL10 and CXCL11) and associated CXCR3 receptor are expressed during the inflammatory process from multiple sclerosis, atherosclerosis or organ transplantation resulting in the recruitment of lymphocytes leading to tissue damage. It is hypothesized that blocking of the ligand/CXCR3 receptor interaction has potential to provide opportunity for development of agents that would block tissue rejection. In this paper, four classes of natural product inhibitors (IC50 ranging 0.1-41 microM) have been described that block the CXCR3 receptor interaction of IP-10 ligand. These in

Control of ColE1 plasmid replication. Interaction of Rom protein with an unstable complex formed by RNA I and RNA II.

A transcript (RNA I) from ColE1 inhibits initiation of replication of the plasmid DNA by binding to the precursor of the primer RNA (RNA II). The ability of RNA I to inhibit replication is altered by the presence of a plasmid-specified small protein, Rom. In vitro, RNA I binds to RNA II to form a very unstable complex, C*. Binding of a single molecule of Rom converts C* to a more stable complex, Cm*. Each of these complexes, C* or Cm*, transforms to a more stable complex, C** or Cm**, respectively. While formation of complex C* or Cm* is inferred from the inhibition of binding caused by a seco
