During one week in 1985, 227 general practitioners in the County of Arhus, Denmark, recorded their prescriptions of benzodiazepines. In all 2,812 prescriptions were recorded, for 2,574 patients. The sex ratio (male/female) was 1.0/2.0. The incidence of first-time prescriptions was 26.0 prescriptions/1,000 inhabitants/year. The prescription rate, calculated as defined daily doses/1,000 inhabitants/day, was highest for minor tranquillizers in the age group 60-69, whereas for hypnotics/sedatives it was highest in the age group 70+. Various benzodiazepines (BZs) were investigated for their analgesic effect by the tail-flick method in mice. Central type BZ-receptor agonists such as clonazepam, chlordiazepoxide and diazepam showed analgesia while the peripheral BZ-receptor binding agent, Ro 5-4864, and the BZ micromolar binding agent phenytoin, failed to show any effect. The occurrence of thrombosis and phlebitis after intravenous injection of 10 mg diazepam, 4 mg lorazepam, or 1-2 mg flunitrazepam was studied on the second or third and the seventh to 10th days. A significantly higher incidence occurred with all drugs on days 7 to 10 than on days 2 and 3. Painless thrombosis occurred much more often with diazepam than with the other two benzodiazepines. Its incidence was greater in small hand or arm veins than in large antecubital vessels. Lorazepam and flunitrazepam therefore have clear advantages over diazepam.
