Immunohistochemistry of aldosterone synthase leads the way to the pathogenesis of primary aldosteronism.

Our group previously purified human and rat aldosterone synthase (CYP11B2 and Cyp11b2, respectively) from their adrenals and verified that it is distinct from steroid 11β-hydroxylase (CYP11B1 or Cyp11b1), the cortisol- or corticosterone-synthesizing enzyme. We now describe their distributions immunohistochemically with specific antibodies. In rats, there is layered functional zonation with the Cyp11b2-positive zona glomerulosa (ZG), Cyp11b1-positive zona fasciculata (ZF), and Cyp11b2/Cyp11b1-negative undifferentiated zone between the ZG and ZF. In human infants and children (<12 years old), th

Abiological catalysis by artificial haem proteins containing noble metals in place of iron.

Enzymes that contain metal ions--that is, metalloenzymes--possess the reactivity of a transition metal centre and the potential of molecular evolution to modulate the reactivity and substrate-selectivity of the system. By exploiting substrate promiscuity and protein engineering, the scope of reactions catalysed by native metalloenzymes has been expanded recently to include abiological transformations. However, this strategy is limited by the inherent reactivity of metal centres in native metalloenzymes. To overcome this limitation, artificial metalloproteins have been created by incorporating

Biosynthesis of versipelostatin: identification of an enzyme-catalyzed [4+2]-cycloaddition required for macrocyclization of spirotetronate-containing polyketides.

Versipelostatin (VST) is an unusual 17-membered macrocyclic polyketide product that contains a spirotetronate skeleton. In this study, the entire VST biosynthetic gene cluster (vst) spanning 108 kb from Streptomyces versipellis 4083-SVS6 was identified by heterologous expression using a bacterial artificial chromosome vector. Here, we demonstrate that an enzyme, VstJ, catalyzes the stereoselective [4+2]-cycloaddition between the conjugated diene and the exocyclic olefin of a newly identified tetronate-containing intermediate to form the spirotetronate skeleton during VST biosynthesis.

Structure of a modular polyketide synthase.

Polyketide natural products constitute a broad class of compounds with diverse structural features and biological activities. Their biosynthetic machinery, represented by type I polyketide synthases (PKSs), has an architecture in which successive modules catalyse two-carbon linear extensions and keto-group processing reactions on intermediates covalently tethered to carrier domains. Here we used electron cryo-microscopy to determine sub-nanometre-resolution three-dimensional reconstructions of a full-length PKS module from the bacterium Streptomyces venezuelae that revealed an unexpectedly dif

Rubber oxygenase and latex clearing protein cleave rubber to different products and use different cleavage mechanisms.

Two types of enzyme for oxidative cleavage of poly(cis-1,4-isoprene) are known. One is rubber oxygenase (RoxA) that is secreted by Xanthomonas sp. strain 35Y and a few other Gram-negative rubber-degrading bacteria during growth on polyisoprene. RoxA was studied in the past, and the recently solved structure showed a structural relationship to bacterial cytochrome c peroxidases (J. Seidel et al., Proc. Natl. Acad. Sci. U. S. A. 110:13833-13838, 2013, http://dx.doi.org/10.1073/pnas.1305560110). The other enzyme is latex-clearing protein (Lcp) that is secreted by rubber-degrading actinomycetes, b

The interaction of isopenicillin N synthase with homologated substrate analogues δ-(L-α-aminoadipoyl)-L-homocysteinyl-D-Xaa characterised by protein crystallography.

Isopenicillin N synthase (IPNS) converts the linear tripeptide δ-(L-α-aminoadipoyl)-L-cysteinyl-D-valine (ACV) into bicyclic isopenicillin N (IPN) in the central step in the biosynthesis of penicillin and cephalosporin antibiotics. Solution-phase incubation experiments have shown that IPNS turns over analogues with a diverse range of side chains in the third (valinyl) position of the substrate, but copes less well with changes in the second (cysteinyl) residue. IPNS thus converts the homologated tripeptides δ-(L-α-aminoadipoyl)-L-homocysteinyl-D-valine (AhCV) and δ-(L-α-aminoadipoyl)-L-homocys

Optimization of lipase-mediated synthesis of 1-nonene oxide using phenylacetic acid and hydrogen peroxide.

Herein, an efficient epoxidation of 1-nonene is described. In a simple epoxidation system, commercially available Novozym 435, an immobilized Candida antarctica lipase B, and hydrogen peroxide (H(2)O(2)) were utilized to facilitate the in situ oxidation of phenylacetic acid to the corresponding peroxy acid which then reacted with 1-nonene to give 1-nonene oxide with high yield and selectivity. The aliphatic terminal alkene was epoxidised efficiently in chloroform to give an excellent yield (97%-99%) under the optimum reaction conditions, including temperature (35 °C), initial H(2)O(2) concentr

Ultrasound irradiation promoted enzymatic transesterification of (R/S)-1-chloro-3-(1-naphthyloxy)-2-propanol.

(R)-1-Chloro-3-(1-naphthyloxy)-2-propanol, which is the key intermediate of (S)-propranolol, was successfully prepared via enantioselective transesterification catalyzed by lipase under ultrasound irradiation. Compared with conventional shaking, the enzyme activity and enantioselectivity were dramatically increased under ultrasound exposure. Effects of various reaction conditions on the synthetic activity of enzyme as well as enantioselectivity, including the type of enzyme, ultrasound power, solvent, acyl donor, temperature and substrate molar ratio, were investigated. One-step enzymatic synthesis of nucleosides from low water-soluble purine bases in non-conventional media.

The effect of several water-miscible cosolvents on activity and stability of soluble and immobilized 2'-deoxyribosyltransferase from Lactobacillus reuteri on Sepabeads® has been studied in order to establish optimal conditions for enzymatic synthesis of nucleosides using purine bases with low solubility in aqueous buffer. As a rule of thumb, there was a general reduction of soluble enzyme activity when cosolvent content was gradually increased in reaction medium. In contrast, immobilized enzyme activity was enhanced 1.2-1.4-fold at 20% of methanol, ethanol, 2-propanol, diethylene glycol, and a

A highly stable plastidic-type ferredoxin-NADP(H) reductase in the pathogenic bacterium Leptospira interrogans.

Leptospira interrogans is a bacterium that is capable of infecting animals and humans, and its infection causes leptospirosis with a range of symptoms from flu-like to severe illness and death. Despite being a bacteria, Leptospira interrogans contains a plastidic class ferredoxin-NADP(H) reductase (FNR) with high catalytic efficiency, at difference from the bacterial class FNRs. These flavoenzymes catalyze the electron transfer between NADP(H) and ferredoxins or flavodoxins. The inclusion of a plastidic FNR in Leptospira metabolism and in its parasitic life cycle is not currently understood. B

An exceptionally DMSO-tolerant alcohol dehydrogenase for the stereoselective reduction of ketones.

A novel short-chain alcohol dehydrogenase from Paracoccus pantotrophus DSM 11072, which is applicable for hydrogen transfer, has been identified, cloned, and overexpressed in E. coli. The enzyme stereoselectively reduces several ketones in a sustainable substrate-coupled approach using 2-propanol (5% v/v) as hydrogen donor. The enzyme maintained its activity in organic co-solvents in biphasic as well as monophasic systems and was even active in micro-aqueous media (1% v/v aqueous buffer). In general, a higher conversion was observed at higher log P values of the solvent, however, DMSO, which e

Deciphering the Enzyme-Mediated Oxidation of Dehydroabietic Acid and Other Diterpenoid Resin Acids.

Cytochrome P450 enzymes are versatile monooxygenase biocatalysts. Members of the bacterial CYP226 family are hypothesized to catalyze the oxidation of diterpenoid resin acids as the first step in their catabolism. Two CYP226 enzymes (CYP226A3 and CYP226A31) from the thermophilic bacterium Zestomonas thermotolerans were characterized. Both could bind the diterpenoids in gum rosin. Oxidation metabolites were formed using peroxide-driven reactions with the wild-type enzymes and variants with modifications at the dioxygen-binding groove. The CYP226A31 enzyme was used to enantioselectively hydroxyl

Improvement of Activity, Stability, and Soluble Expression of Nitrilase through Stepwise Sequence Optimization for Highly Efficient Synthesis of Nicotinic Acid.

Nitrilases are important industrial enzymes that catalyze the conversion of nitrile compounds into carboxylic acids. This study employed a stepwise sequence optimization strategy to enhance nitrilase performance, combining PROSS for combinatorial mutation design with SPIRED-Fitness for fitness-guided optimization. Beneficial distal mutations were identified and iteratively combined. Ultimately, optimal mutant PD1F4 showed over 2-fold increase in specific activity compared to the starting variant C57E10, and its half-life at 50 °C improved from 10.53 ± 1.13 to 123.40 ± 5.94 min. Adaptive Pore Deformation in Metal-Organic Framework Nanosheets for Enhanced Enzymatic Catalysis.

The nanoconfinement of enzymes within porous scaffolds is a pivotal strategy for constructing robust and industrially relevant biocatalysts. However, a molecular-level understanding of the host-guest interactions that dictate catalytic efficiency remains a significant challenge, largely due to the inherent heterogeneity of enzyme guests and the scarcity of synthetic material hosts that permit atomic-level investigation of their pores upon enzyme loading. Here, we report the design of two redox-active biocatalysts via the encapsulation of cytochrome c within the mesopores of isostructural metal

Beyond Branching: Unlocking the Catalytic Versatility of Branching Enzymes for the Design of Diverse α-Glucan Structures.

Starch-modifying glycoside hydrolases (GHs) typically operate via a retaining double-displacement mechanism, involving formation of a covalent glycosyl-enzyme intermediate. This intermediate can be resolved either by water, resulting in hydrolysis, or by a glucan acceptor, leading to transglucosylation. Many GHs exhibit both catalytic activities, although they are classified according to their predominant reaction. For example, branching enzymes (BEs) catalyse α-(1→4) bond cleavage and α-(1→6) branch formation via transglucosylation, while also exhibiting minor hydrolytic and disproportionatio

Regulating relative nitrogen locations of bipyridine covalent organic frameworks with enhanced peroxidase-like activity for biosensing.

Since metal-based enzyme mimics can raise biosafety concerns due to potential toxicity accumulation, it is necessary to develop metal-free alternatives that exhibit high catalytic efficiency and enhanced selectivity. Herein, we report the development of bipyridine-bridged covalent organic frameworks (COFs) exhibiting peroxidase-like (POD-like) activity as intelligent artificial enzymes for biomarker detection. Their catalytic function is enabled by a facile strategy that modulates charge transfer within the framework through precise regulation of the nitrogen site in the bipyridine units. The

Domain-Swapping of a GT-B Glycosyltransferase Broadens Its Sugar Donor Range for Glycodiversification of Natural Products.

Glycosylation of natural compounds modulates their bioactivity as well as their physicochemical properties, making glycodiversification a valuable strategy for identifying glycosidic derivatives with improved functional profiles. However, the use of glycosyltransferases (GTs) for their synthesis is constrained by stringent sugar nucleotide donor selectivity, which limits the structural diversity of accessible glycosides. The chimeric NB1//CC1, constituted of 2 self-assembled domains originating from plant glycosyltransferases, was previously demonstrated to transfer glucose onto a wider range
