Decisional factors supporting the use of direct or indirect techniques and related esthetic biomaterials.

There are many options for restoring or improving a smile, including various tooth-colored biomaterials and clinical protocols. Some simple and limited tooth form or color corrections in young patients with healthy teeth logically call for a direct approach using composite resins, whereas more extensive smile rehabilitation in older patients with high esthetic demands should rationally be performed with ceramics. The various patient conditions that comprise these 'simple' examples, for which all practitioners may apply the same decision-making rationale, do not apply in increasingly complicate

Enhancement of Copper Antiviral Activity with Glutathione Treatment.

Copper and its alloys have antimicrobial activity effective for various pathogens. Their application to touch surfaces successfully reduced bioburden in hospital intensive care units but failed to completely prevent the transmission of pathogens derived from the hospital room. Discoloration of pure copper with surface oxide growth is another issue to discourage its application to touch surfaces. Common copper alloys have relatively high resistance to discoloration, but their antimicrobial activity is lower than that of pure copper. Therefore, enhancement of the antimicrobial activity of copper

Investigating the Impact of Fused Filament Fabrication Process Parameters on the Compressive Properties of Porous PEEK and PEKK Biomaterials.

Additive manufacturing (AM) can create orthopedic devices with integrated porosity that enables bone fixation post-implantation. While porosity is key in promoting bone ingrowth and long-term fixation, the device must provide adequate mechanical strength and functionality. Since AM process parameters dictate the final mechanical performance of printed parts, identifying key process parameter levels that preserve or improve such behavior in load-bearing devices with integrated porosity is essential. Using a Taguchi design of experiments, gyroid-structured polyether-ether-ketone (PEEK) and polye

Corrosion and Wear Properties of Low-Modulus TiZrNb-Based Complex Concentrated Alloys under Simulated Physiological Conditions.

Titanium (Ti) and its alloys have evolved through multiple generations to possess enhanced biomedical performance. However, challenges such as stress shielding, low yield strength, and inadequate corrosion resistance persist in conventional Ti-based alloys. In this study, we explore the complex concentrated alloy (CCA) approach to develop two novel Ti-Zr-Nb alloys: Ti50Zr35Nb15 and Zr50Ti35Nb15. These compositions were designed using CALPHAD-based phase stability predictions based on nontoxic lightweight approach and fabricated via vacuum arc melting, yielding a single-phase body-centered cubi

Biomechanical analysis of diamond-like carbon coated growth-guidance spinal system under cyclic axial compression.

PURPOSE: Growth-guidance spinal systems are used in children with progressive spinal deformity to reduce repeated surgical lengthening while allowing continued spinal growth. Their long-term biomechanical behaviour under cyclic loading remains insufficiently characterised. This study assessed whether diamond-like carbon (DLC) coating of rods and sliding screws affects axial stiffness and local kinematics in a growth-guidance spinal system. METHODS: Twelve porcine Th11-L7 spine specimens were assigned to three groups: non-instrumented control, non-coated titanium alloy instrumentation, and ins

A Facile Fabrication Process for Handmade Fully Polymeric Neural Interfaces.

Conventional neural interfaces are typically manufactured by photolithographic micromachining using thermoplastic insulators and noble-metal conductors. Although effective, these approaches require costly, time-intensive infrastructure, restrict material selection, and often produce devices with substantial mechanical and interfacial mismatch relative to soft neural tissue, limiting long-term performance. Here, we introduce CASPER (CAsted and Screen-Printed polymeric ElectRodes), a cleanroom-free and low-cost benchtop strategy for the fully manual fabrication of implantable neural interfaces f

Evaluating Cytocompatibility of Corynebacterium glutamicum-poly Vinyl Alcohol Living Biomaterials for Ocular Use.

In ophthalmology, living biomaterials have appeared as promising drug delivery and biosensor devices to tackle dynamic sensing and delivery of compounds. Their living character complicates their assessment with the also dynamic ocular tissues. The use of animal experiments increases complexity, and most animal ocular models are anatomically different from humans. Thus, in vitro ocular systems applied specifically to living biomaterials are required to assess their safety, compatibility and efficacy. Here, we report on an in vitro cornea model for co-cultures with Corynebacterium glutamicum-pol

Biomimetic tubular materials: from native tissues to a unifying view of new vascular, tracheal, gastrointestinal, oesophageal, and urinary grafts.

Repairing tubular tissues-the trachea, the esophagus, urinary and gastrointestinal tracts, and the circulatory system-from trauma or severe pathologies that require resection, calls for new, more effective graft materials. Currently, the relatively narrow family of materials available for these applications relies on synthetic polymers that fail to reproduce the biological and physical cues found in native tissues. Mimicking the structure and the composition of native tubular tissues to elaborate functional grafts is expected to outperform the materials currently in use, but remains one of the

Fiber-reinforced zwitterionic elastomer composites for artificial heart valves.

Valvular heart disease (VHD) is a leading cause of cardiovascular morbidity and mortality. Polymeric heart valves (PHVs) offer potential solutions for treating VHDs but are limited by issues like thrombosis, calcification, and inflammation. Surface modification with antifouling coatings has been explored to mitigate those complications, but these coatings often exhibit poor stability and mechanical mismatch with elastomer substrates. Here, we report a fiber-reinforced zwitterionic elastomer composite for PHVs that simultaneously achieves antifouling surfaces and robust mechanical properties. T

Mimicking techniques in small diameter vascular grafts: enhancing design through multi-layered electrospinning techniques.

The increasing prevalence of occlusive arterial diseases, such as peripheral arterial disease and coronary heart disease, underscores the critical need for effective vascular interventions. Traditional methods, including autologous vessel grafting, often fall short in the case of small-diameter vascular grafts (SDVGs), which present challenges such as thrombosis and limited long-term patency. This study reviews recent advancements in electrospinning techniques aimed at generating biomimetic SDVGs that closely replicate the structure and mechanical properties of natural blood vessels. By employ

Macroheterogeneities Induced by Disulfide Bond Reduction in Native Mucus.

Disulfide bond reducing agents have long been used as therapeutic drugs (mucolytics) for mucus hypersecretions. Breakage of disulfide bridges is known to cause a reduction in the degree of cross-linking, making the mucus more fluid. In addition to the drop in viscoelastic properties, the disulfide breakage is also known to affect the structure of the mucus on both micro- and mesoscale. Despite this knowledge, little is known about the reorganization of the mucus at the macroscopic scale. This contribution explores the effect of the reducing agent, tris(2-carboxyethyl)phosphine hydrochloride (T

Histidine Tag-Specific PEGylation Improves the Circulating Half-Life of TIMP2.

An overarching limitation of therapeutic biologics is the limited half-life these proteins often exhibit once in circulation. PEGylation, the chemical conjugation of proteins to poly(ethylene glycol) (PEG), is a common strategy to improve protein pharmacokinetics (PK) by enhancing stability, reducing immunogenicity, and decreasing renal clearance. Tissue Inhibitor of Metalloproteinases 2 (TIMP2) is a 22 kDa matrisome protein that exhibits therapeutic potential across a range of human disease models yet possesses a short serum half-life. To advance the therapeutic development of recombinant His

Biological, Biomechanical, and Histopathological Evaluation of Polyetherketoneketone Bioactive Composite as Implant Material.

While polyetherketoneketone is a high-performance thermoplastic polymer, its hydrophobicity and inertness limit bone adhesion. This study aimed to evaluate a novel PEKK/CaSiO3/TeO2 nanocomposite, comparing it to PEKK/15 wt.% CaSiO3 and PEKK groups. The in vitro study, involving 90 discs (n = 30), assessed the cytotoxicity of all groups after 24, 72, and 168 h. The in vivo animal study, using cylinder-type implants (n = 30), evaluated osseointegration through biomechanical push-out tests, descriptive histopathological examinations of decalcified sections stained with hematoxylin and eosin, and

Osteointegration and remodeling of a novel mineral magnesium phosphate-based bone glue in-vivo.

BackgroundBone adhesives represent a major challenge in the field of materials science. Magnesium phosphate-based mineral bone cements exhibit high biocompatibility, particularly for the use as bone substitutes. In combination with calcium ions and phytic acid as liquid components, these cements demonstrate adhesive properties and potentially represent a promising new type of bone glue. Two of these first cement-adhesives were further investigated for their in vivo properties on distal femoral defects in rabbits in this study.Materials and methodsIn 4.5-month-old New Zealand White rabbits, dis

Multifunctional Characteristics of Cu/Zn Co-Doped Hydroxyapatite: Enhanced Electrical, Surface, and Biocompatibility.

Developing multifunctional biomaterials with both electrical and biological properties is crucial for next-generation biomedical platforms. This study looks into how Cu/Zn co-doping affects the structural, electrical, and biological performance of hydroxyapatite (Ca10-x-yZnxCuy(PO4)6(OH)2; x = y = 0.2-1.2), which was synthesized through a solid-state reaction. Among the samples, the CZ6 composition (x = y = 0.6) showed the best properties. It had a single-phase hexagonal structure, a nanoscale crystallite size of about 32 nm, a d-spacing of 0.27 nm along the (112) plane, and a grain size that

Biodegradation study in FBS media of polycaprolactone patch as a potential prenatal treatment for myelomeningocele.

Myelomeningocele (MMC) is a congenital defect of the spine characterized by meningeal and spinal cord protrusion through open vertebral archs, and its exposure to the amniotic fluid. Given that the progression of neuronal loss begins early in fetal life, an early coverage of the defect is required to improve the neurological outcomes. Several studies have proposed patches as an alternative to full surgical repair, to achieve an early protection of the spine and possibly reduce the rate of complications of current prenatal surgical procedures. In our previous work, we developed a biocompatible,

Hydrogel Encapsulation of a Designed Fluorescent Protein Biosensor for Continuous Measurements of Sub-100 nM Nicotine.

The reinforcing, addictive, and therapeutic properties of nicotine depend strongly on the concentration and time dependence of nicotine in human biofluids. This pattern, termed [nicotine]t, varies markedly among methods of nicotine consumption and, for a given method, among individual users. Measuring [nicotine]t presents challenges: current approaches are expensive, invasive, tedious, and discontinuous. We report the entrapment of a purified, previously developed fluorescent biosensor protein, iNicSnFR12, in hydrogels. We optimized poly(ethylene glycol) diacrylate hydrogels for optical clarit

Bilayer Electrospun Polyurethane Membrane With Tunable Elastomeric Properties for the Treatment of Congenital Diaphragmatic Hernia.

Congenital diaphragmatic hernia is a rare condition characterized by the development of a defect in the diaphragm during early embryogenesis. For the most severe cases, when the diaphragmatic defect is large, the gap is currently closed by a prosthetic patch made of e-PTFE (Gore-Tex) materials, which lack sufficient elasticity, causing early rupture of stitches and subsequent hernia recurrence. In this study, we introduce a novel thermoplastic polyurethane membrane designed to accommodate the child's growth. This film/fiber bilayer membrane, produced in a single continuous electrospinning proc

Evaluation Strategies for Tissue-engineered Tracheas: From In Vitro Characterization to In Vivo Assessment.

The trachea plays a critical role in maintaining airway patency, ventilation, and mucociliary clearance, supported by its unique anatomical and structural features. Tracheal defects resulting from congenital anomalies, malignancies, trauma, or prolonged intubation present significant clinical challenges. Traditional reconstruction methods, such as end-to-end anastomosis and patch grafts, are often limited by technical feasibility and suboptimal outcomes. Recently, tissue-engineered tracheal scaffolds (TETs), particularly those fabricated using 3D bioprinting technologies, have emerged as promi
