Benchmarking In Silico Metabolite Prediction Tools against Human Radiolabeled ADME Data for Small-Molecule Drugs.

Artificial intelligence-based in silico metabolite prediction tools are increasingly used in drug development, but their performance against high-quality human data remains uncertain. We evaluated four open-access models, including SyGMa, GLORYx, BioTransformer 3.0, MetaPredictor, and MetaTrans, using 11 small-molecule drugs with published human radiolabeled ADME data. Predicted metabolites were compared with experimentally identified human metabolites, and model performance was assessed using recall, precision, balanced accuracy, F1, and Jaccard scores. Results indicated a distinct trade-off

Phytodegradation and phytotoxicity of N-ethyl perfluorooctane sulfonamidoethanol (N-EtFOSE) in pakchoi (Brassica chinensis L.) via root and foliar uptake from physio-biochemical and transcriptomic perspectives.

N-ethyl perfluorooctane sulfonamidoethanol (N-EtFOSE), a known volatile perfluorooctane sulfonate (PFOS) precursor (PreFOS), has been widely detected in the environment. However, the mechanisms underlying the behaviors and phytotoxicity of N-EtFOSE in plants are unknown. Integrative physio-biochemical and transcriptomic analyses were used to reveal the accumulation, biotransformation, and toxicity of N-EtFOSE in pakchoi (Brassica chinensis L.) through root and foliar exposure experiments. Root uptake from exposure medium and efficient translocation to shoot, and foliar uptake from air and inef…

Utilization of naproxen by Amycolatopsis sp. Poz 14 and detection of the enzymes involved in the degradation metabolic pathway.

The pollution of aquatic environments by drugs is a problem for which scarce research has been conducted in regards of their removal. Amycolatopsis sp. Poz 14 presents the ability to biotransformation naphthalene at high efficiency, therefore, in this work this bacterium was proposed as an assimilator of naproxen and carbamazepine. Growth curves at different concentrations of naproxen and carbamazepine showed that Amycolatopsis sp. Poz 14 is able to utilize these drugs at a concentration of 50 mg L-1 as a source of carbon and energy. At higher concentrations, the bacterial growth was inhibited…

Co-occurrence of Photochemical and Microbiological Transformation Processes in Open-Water Unit Process Wetlands.

The fate of anthropogenic trace organic contaminants in surface waters can be complex due to the occurrence of multiple parallel and consecu … o moieties, respectively. Despite substantial differences in kinetics of different antiviral drugs, biotransformation reactions mainly involved oxidation of hydroxyl groups to the corresponding carboxylic acids. Phototransformation rates of parent antiviral drugs and their biotransformation products were similar, indicating that prior exposure to microorganisms (e. …

Scale-up and intensification of (S)-1-(2-chlorophenyl)ethanol bioproduction: economic evaluation of whole cell-catalyzed reduction of o-chloroacetophenone.

Escherichia coli cells co-expressing genes coding for Candida tenuis xylose reductase and Candida boidinii formate dehydrogenase were used for the bioreduction of o-chloroacetophenone with in situ coenzyme recycling. The product, (S)-1-(2-chlorophenyl)ethanol, is a key chiral intermediate in the synthesis of polo-like kinase 1 inhibitors, a new class of chemotherapeutic drugs. Production of the alcohol in multi-gram scale requires intensification and scale-up of the biocatalyst production, biotransformation, and downstream processing. Cell cultivation in a 6.9-L bioreactor led to a more than t…

Identification of New Hepatic Metabolites of Miconazole by Biological and Electrochemical Methods Using Ultra-High-Performance Liquid Chromatography Combined with High-Resolution Mass Spectrometry.

The main objective of this study was to investigate the metabolism of miconazole, an azole antifungal drug. Miconazole was subjected to incubation with human liver microsomes (HLM) to mimic phase I metabolism reactions for the first time. Employing a combination of an HLM assay and UHPLC-HRMS analysis enabled the identification of seven metabolites of miconazole, undescribed so far. Throughout the incubation with HLM, miconazole underwent biotransformation reactions including hydroxylation of the benzene ring and oxidation of the imidazole moiety, along with its subsequent degradation. Additio…

Antibiotic application may raise the potential of methylmercury accumulation in fish.

Mercury (Hg) biotransformation can significantly affect the Hg speciation and bioaccumulation in fish, where gut microbiota play an important role in this process. Antibiotics have been extensively used in aquaculture and can affect gut microbial structure. However, the influence of antibiotics on Hg biotransformation in fish has not been thoroughly understood. The present study investigated the effects of antibiotic (florfenicol) application on gut microbiota and subsequent impacts on Hg biotransformation and bioaccumulation in tilapia (Oreochromis mossambicus). The results showed that the fl…

Evaluation of drug uptake and deactivation in plant: Fate of albendazole in ribwort plantain (Plantago laceolata) cells and regenerants.

Albendazole (ABZ) is a benzimidazole anthelmintic widely used especially in veterinary medicine. Along with other drugs, anthelmintics have become one of a new class of micro-pollutants that disturb the environment but the information about their fate in plants remains limited. The present study was designed to test the uptake and biotransformation of ABZ in the ribwort plantain (Plantago lancelota), a common meadow plant, which can come into contact with this anthelmintic through the excrements of treated animals in pastures. Two model systems were used and compared: cell suspensions and whol…

Pharmacokinetics of diuretics and methylxanthines in the neonate.

The elimination of diuretics and methylxanthines is considerably slower in the neonate than in the adult. Dose guidelines, especially during long term maintenance, must be adjusted to account for this slower drug elimination. Pharmacokinetic studies and the requisite pharmacologic evaluation on diuretics such as hydrochlorothiazide, spironolactone, ethacrynic acid and others should be done. Furosemide undergoes biotransformation in the newborn producing an acid metabolite and a glucuronide conjugate. Methylxanthines are effective in the treatment of neonatal apnea. Plasma elimination of theoph…

Saccharomycopsis fibuligera solid-state fermentation of wheat bran for glutathione enrichment and functional improvement.

Wheat bran (WB) is an abundant and low-cost cereal by-product, but its compact cell-wall structure and high levels of anti-nutritional factors limit its high-value utilization. In this study, a GSH-producing Saccharomycopsis fibuligera strain K2 with broad extracellular hydrolytic activities was selected for wheat bran biotransformation, and its glutathione (GSH)-producing capacity was further enhanced by atmospheric and room-temperature plasma (ARTP) mutagenesis. The resulting mutant 1-7 produced 29.57% more GSH than the parental strain K2 and was subsequently applied to WB solid-state fermen…

Transformation of Chlorofluorocarbons Investigated via Stable Carbon Compound-Specific Isotope Analysis.

Compound-specific isotope analysis (CSIA) is a valuable tool in contaminant remediation studies. Chlorofluorocarbons (CFCs) are ozone-depleting substances previously thought to be persistent in groundwater under most geochemical conditions but more recently have been found to (bio)transform in some laboratory experiments. To date, limited applications of CSIA to CFCs have been undertaken. Here, biotransformation-associated carbon isotope enrichment factors, εC,bulk for CFC-113 (εC,bulk = -8.5 ± 0.4‰) and CFC-11 (εC,bulk = -14.5 ± 1.9‰), were determined. δ13C signatures of pure-phase CFCs and h…

Arsenic urinary speciation in Mthfr deficient mice injected with sodium arsenate.

In most mammalian species, arsenic biotransformation occurs primarily by biomethylation and reduction reactions, with dimethylarsinic acid being the predominant metabolite excreted in the urine. Methylenetetrahydrofolate reductase (Mthfr) plays a key role in folate metabolism by channeling one-carbon units between nucleotide synthesis and methylation reactions. In the study on transgenic Mtfhr knockout mice we investigated: (1) whether Mthfr is an important determinant in arsenic biotransformation by performing urinary arsenic speciation, and (2) whether dietary folate deficiency alters arseni…

Isoniazid potentiation of a guinea pig model of halothane-associated hepatotoxicity.

Isoniazid (INH) is a selective inducer of cytochrome P-450 isozymes that are involved in the biotransformation of organohalogen anesthetics. It has been used to produce a rat model of halothane-associated hepatotoxicity that was linked to enhanced oxidative biotransformation of the anesthetic. Guinea pigs were pretreated with INH in order to potentiate halothane-induced hepatic necrosis and to study the oxidative pathway as a hepatotoxic mechanism in this species. The animals received either 12.5, 25.0 or 50.0 mg kg-1 INH i.p. for 7 days. Following halothane exposure, there were dose-dependent…

Clinical consequences of the lipophilicity and plasma protein binding of antiarrhythmic drugs and active metabolites in man.

In two series of antiarrhythmic drugs tested, as the octanol/water partition coefficient increases so do the following: elimination from the body by biotransformation, first-pass biotransformation in the liver and gastrointestinal tract after oral administration, protein binding to some extent, and penetration into brain tissue. Patients receiving lipophilic beta-adrenoreceptor blocking drugs may experience more central nervous system side effects than those receiving hydrophilic beta blockers. Structural modification of a drug, guided by the concept of bioisosterism, may allow the disassociat…

Production of iron enriched Saccharomyces boulardii: impact of process variables.

About half of the 1.62 billion cases of anemia are because of poor diet and iron deficiency. Currently, the use of iron-enriched yeasts can be used as the most effective and possible way to prevent and treat anemia due to the ability of biotransformation of mineral compounds into the organic form. In this research, for the first time, Saccharomyces (S.) boulardii was used for iron enrichment with the aim that the probiotic properties of yeast provide a potential iron supplement besides improving the bioavailability of iron. Also, due to its higher resistance than other Saccharomyces strains ag…
