Salt sensitivity of volume and blood pressure in a mouse with globally reduced ENaC γ-subunit expression.

The epithelial Na+ channel (ENaC) promotes the absorption of Na+ in the aldosterone-sensitive distal nephron, colon, and respiratory epithelia. Deletion of genes encoding subunits of ENaC results in early postnatal mortality. Here, we present the initial characterization of a mouse with dramatically suppressed expression of the ENaC γ-subunit. We used this hypomorphic (γmt) allele to explore the importance of this subunit in homeostasis of electrolytes and body fluid volume. At baseline, γ-subunit expression in γmt/mt mice was markedly suppressed in the kidney and lung, whereas electrolytes re

Erythrocytes labeled with [(18) F]SFB as an alternative to radioactive CO for quantification of blood volume with PET.

Inhaled radioactive CO is currently the tracer of choice for blood volume quantification by positron emission tomography (PET). This measurement is of great interest for several clinical and research applications. However, owing to the short half-life of the radiolabeled CO, it can only be used in centers equipped with a cyclotron. In the present work, we propose an alternative method to label the red blood cells with [(18) F] in order to obtain blood volume measurements by PET. The use of the radioactive synthon [(18) F] N-succinimidyl 4-[(18) F]fluorobenzoate ([(18) F]SFB) was evaluated for …

Blood volume measurements in patients with heart failure and a preserved ejection fraction: implications for diagnosing anemia.

Racial differences in the prevalence of anemia in patients with heart failure have been noted. The diagnosis of anemia in heart failure patients can be confounded by many factors. Plasma volume expansion is one of the most prominent confounders. The authors investigated the difference of anemia prevalence using two different diagnostic techniques: peripheral hemoglobin recommended by the World Health Organization criteria and blood volume (BV) analysis. Racial disparities in the prevalence of anemia using both measures were compared. Sixty patients with heart failure and preserved ejection fra…

The effects of low-pressure carbon dioxide pneumoperitoneum on cerebral oxygenation and cerebral blood volume in children.

UNLABELLED: We examined the effects of low-pressure carbon dioxide pneumoperitoneum on regional cerebral oxygen saturation (ScO(2)) and cerebral blood volume (CBV) in children. Fifteen children, ASA I--III, scheduled for laparoscopic fundoplication, were investigated in the head-up position (10) and ventilated to a baseline end-tidal CO(2) (PETCO(2)) between 25 and 33 mm Hg. Ventilatory settings remained unchanged during the operation. ScO(2) and CBV were assessed with near-infrared spectroscopy and recorded together with end-tidal and arterial carbon dioxide (PaCO(2)) at 5 time points: before…

The effect of urine volume and nitric oxide on basal bladder blood flow: response to catheterization and drainage.

Preliminary studies demonstrated that catheterization and drainage of the urinary bladder resulted in a significant increase in blood flow to the bladder. The objectives of this study were to determine 1) the relationship between urine volume and basal blood flow to the bladder smooth muscle and mucosa, 2) the effect of acute catheterization and drainage on bladder mucosal and smooth muscle blood flow, and 3) whether nitric oxide was involved in regulation of basal blood flow or the increase in blood flow observed after catheterization and bladder drainage. Twenty-four rabbits were separated i

Brain volumes and regional cerebral blood flow in carriers of the Swedish Alzheimer amyloid protein mutation.

As a preliminary part of a longitudinal clinical study of carriers of the Swedish amyloid precursor protein (APP) 670/671 mutation, 13 members of a family were investigated with magnetic resonance imaging (MRI) brain volumetry and single photon emission computed tomography (SPECT) cerebral blood flow (CBF) measurements. Five of the family members were mutation carriers; eight were not carriers. Two carriers were younger than 40 years of age and had no evidence of cognitive dysfunction or structural or functional brain changes. One carrier with 4 years to expected disease onset showed poor perf

Radioimmunoassay of atrial natriuretic peptide in human plasma: application to studies of volume and blood pressure homeostasis.

Sensitive radioimmunoassay for determination of immunoreactive atrial natriuretic peptide (ANP) in human plasma was developed and employed for the study of plasma ANP concentrations in healthy controls under basal conditions (2.4 +/- 0.1 pmol/l) and during volume expansion by saline infusion (9.6 +/- 2.0 pmol/l and 14.2 +/- 1.8 pmol/l, respectively). Plasma renin activity and plasma aldosterone concentration exhibited opposite changes during saline infusion. In pathological states associated with extracellular fluid volume (ECFV) expansion, ANP concentration were significantly higher than in t

Assessment of blood substitutes: I. Efficacy studies in anesthetized and conscious rats with loss of 1/3, 1/2 and 2/3 blood volume.

Acute and long-term recovery (14 days) studies were conducted in conscious rats bled 1/3, 1/2, and 2/3 blood volume, and in anesthetized rats bled 1/3 volume. Study I: bled 1/3 blood volume. Study Ia: Anesthetized rats bled 1/3 blood volume were in shock and most died within 1 hour; the group which received infusion of 3x volume Ringer's lactate regained blood pressure with 100% long-term recovery. Study Ib: In conscious rats bled 1/3 blood volume, blood pressure did not fall to shock levels, and the long-term recovery in both the control group and the group which received 3x volume Ringer's l…

[Analysis of regional blood flow, blood volume, oxygen and glucose metabolism in heterogeneous parts of untreated high grade gliomas using positron emission tomography (PET)].

A high resolution positron emission tomography (PET), HEADTOME III, has enabled us to visualize heterogeneous parts, i.e. viable, necrotic, and edematous portions in malignant gliomas, and to quantify regional hemocirculation and metabolism of the tumors using 15O and 18F-fluorodeoxyglucose tracers. Hemocirculatory and metabolic indices of regional cerebral blood flow (rCBF), blood volume (rCBV), oxygen extraction fraction (rOEF), oxygen consumption (rCMRO2) and glucose consumption (rCMRGl) were studied in eight patients with untreated malignant gliomas. Regions of interest (ROIs) in PET image…

Bioimpedance-assessed volume overload predicts interdialytic hypertension and disrupted circadian blood pressure rhythm in maintenance hemodialysis patients.

OBJECTIVE: Hypertension and abnormal circadian blood pressure (BP) rhythm are prevalent in maintenance hemodialysis (MHD) patients, and are closely associated with cardiovascular diseases (CVD) and increased all-cause mortality. Volume overload represents a critical factor in effectively controlling hypertension. Bioelectrical Impedance Analysis (BIA) has been validated as an accurate method for assessing volume status. This study aimed to investigate the predictive value of BIA-derived volume indicators for hypertension and circadian BP rhythm abnormalities in MHD patients, providing a theore

Predictive value of blood urea nitrogen to creatinine ratio and estimated plasma volume status in heart failure.

BACKGROUND: The blood urea nitrogen to creatinine ratio (BCR) and estimated plasma volume status (ePVS) may be prognostic markers in heart failure (HF), but their combined efficacy is unclear. This research aims to determine the prognostic utility of BCR and ePVS in critically ill HF patients in the intensive care unit. METHODS: Data from the MIMIC-IV database were analyzed. ePVS was calculated using hemoglobin and hematocrit levels with Strauss-derived Duarte method. The primary outcome was 1-year all-cause mortality (ACM). Receiver operating characteristic (ROC) curves identified cutoff val
