Description and evaluation of a canine volunteer blood donor program.

Human volunteer blood donor programs are commonplace, but the concept of nonhuman animal blood banking is relatively new. Few studies exist regarding efficacy, donor screening, and safety for volunteer companion animals. This retrospective study evaluated a nonprofit, community-based canine volunteer donor program using community blood drives. Of 98 potential donors, 14 were ineligible to donate, including 4 who tested seropositive for blood-borne pathogens. Of 84 donors, 45 were Dog Erythrocyte Antigen (DEA) 1.1 positive and 39 were DEA1.1 negative. Donations totaling 143 included 29 repeat d…

Mosquito- and tick-borne orthoflaviviruses cross an in vitro endothelial-astrocyte barrier.

INTRODUCTION: The genus Orthoflavivirus of the Flaviviridae family includes several notable pathogens such as mosquito-borne West-Nile virus (Orthoflavivirus nilense, WNV) and Tick-borne encephalitis virus (Orthoflavivirus encephalitidis, TBEV) that are highly neurotropic and may cause severe neurological disease leading to lifelong disabilities, coma and death. These viruses have developed mechanisms to breach the compact blood-brain barrier (BBB) and establish infection within the central nervous system (CNS). Nevertheless, neuroinvasive mechanisms of orthoflaviviruses remain poorly understo

[The principal arthropod vectors of disease. What are the risks of travellers' to be bitten? To be infected?].

Many blood-sucking arthropods are potential vectors of disease. To become a vector, the arthropod must be susceptible to the infective agent and must survive the incubation period so as to transmit the pathogens to a host. While some arthropod associated diseases affect only man (e.g. malaria) most of these diseases are (anthropo-) zoonoses with man often an accidental host. The risk of contamination depends on the one hand on the biting behaviour of the vector, its biology and distribution, and on the other hand on the sites visited by the traveller, the length of his stay, his activities, th

Efforts Toward Elimination of Infectious Agents in Blood Products.

The US blood supply has never been safer. This level of safety depends on a multifaceted approach including blood donor screening, sensitive infectious disease testing, and good manufacturing practice. However, risks remain for transfusion-transmitted infections due to bacterial contamination of platelets and emerging diseases. Thus, ongoing improvements in screening and testing are required. Newer pathogen reduction technologies have shown promise in further ameliorating the safety of the blood supply.

First detection of Anaplasma phagocytophilum and enzootic focus of Theileria orientalis in cattle from Bosnia and Herzegovina.

Theileria orientalis is a tick-borne intraerythrocytic protozoan parasite of cattle causing oriental or benign theileriosis. It has a worldwide distribution and is not considered a zoonotic agent, while the disease symptoms range from subclinical to severe. Theileria spp. are very common co-infecting agents with other tick borne pathogens such as Babesia spp., Anaplasma spp. including zoonotic Anaplasma phagocytophilum with rising prominence. This study is an exploratory investigation of the occurrence of Anaplasma spp. and other blood parasites in cattle from Gacko and Bileća, Herzegovina. Bl

A new real-time PCR assay for improved detection of the parasite Babesia microti.

Babesiosis is an emerging zoonosis with important public health implications, as the incidence of the disease has risen dramatically over the past decade. Because the current gold standard for detection of Babesia is microscopic examination of blood smears, accurate identification requires trained personnel. Species in the genus cannot be distinguished microscopically, and Babesia can also be confused with the early trophozoite stage (ring forms) of Plasmodium parasites. To allow more accurate diagnosis in a format that is accessible to a wider variety of laboratories, we developed a real-time

An outbreak of bovine babesiosis in February, 2019, triggered by above average winter temperatures in southern England and co-infection with Babesia divergens and Anaplasma phagocytophilum.

BACKGROUND: Bovine babesiosis, commonly known as redwater fever, is a sporadic tick-borne disease in the United Kingdom. Outbreaks occur during the spring, summer and autumn months when ticks are active. This study reports the findings of an investigation of an outbreak of bovine babesiosis during the winter month of February, 2019. METHODS: DNA from blood, organ and tick samples taken from affected cattle were tested for the presence of piroplasm and Anaplasma phagocytophilum DNA using PCRs directed to the 18S rRNA gene and msp2 gene respectively. The species of piroplasm was confirmed by se

Pathogen surveillance in cave-dwelling phlebotomine sand flies from Chiang Rai, northern Thailand, with the first detection of Hepatocystis.

Sand flies are important vectors of several pathogens of medical and veterinary significance, including Leishmania spp., Trypanosoma spp., Bartonella spp., and various arboviruses. Cave ecosystems provide suitable habitats for sand flies and often harbor large bat populations that serve as reservoirs for diverse microorganisms, potentially facilitating pathogen circulation within cave environments. The present study aimed to investigate the presence of sand fly-borne pathogens in cave-dwelling sand flies from three caves in Chiang Rai Province, northern Thailand. A total of 616 sand fly specim

A Multi-Pathogen Screening of Captive Reindeer (Rangifer tarandus) in Germany Based on Serological and Molecular Assays.

Captive reindeer in German zoos and wildlife parks live outside their natural geographic range and are exposed to a variety of viral, bacterial and protozoan pathogens, some host-specific and some which they are not exposed to in their native habitat. Reindeer blood samples and ticks collected in 2013 from 123 reindeer at 16 different zoological facilities were available from a previous study. The aims of this study were to assess the serological status of these animals with regards to various microorganisms as well as to test ticks (Ixodes ricinus) and blood samples for the presence of Anapla

Na+/H+ antiport is essential for Yersinia pestis virulence.

Na(+)/H(+) antiporters are ubiquitous membrane proteins that play a central role in the ion homeostasis of cells. In this study, we examined the possible role of Na(+)/H(+) antiport in Yersinia pestis virulence and found that Y. pestis strains lacking the major Na(+)/H(+) antiporters, NhaA and NhaB, are completely attenuated in an in vivo model of plague. The Y. pestis derivative strain lacking the nhaA and nhaB genes showed markedly decreased survival in blood and blood serum ex vivo. Complementation of either nhaA or nhaB in trans restored the survival of the Y. pestis nhaA nhaB double delet

Assessing tick attachments to humans with citizen science data: spatio-temporal mapping in Switzerland from 2015 to 2021 using spatialMaxent.

BACKGROUND: Ticks are the primary vectors of numerous zoonotic pathogens, transmitting more pathogens than any other blood-feeding arthropod. In the northern hemisphere, tick-borne disease cases in humans, such as Lyme borreliosis and tick-borne encephalitis, have risen in recent years, and are a significant burden on public healthcare systems. The spread of these diseases is further reinforced by climate change, which leads to expanding tick habitats. Switzerland is among the countries in which tick-borne diseases are a major public health concern, with increasing incidence rates reported in

Contrasting pathogen prevalence between tick and dog populations at Chornobyl.

BACKGROUND: The 1986 disaster at the Chornobyl Nuclear Power Plant released massive amounts of radioactive material into the local environment. In addition to radiation, remediation efforts and abandonment of military-industrial complexes contributed to contamination with heavy metals, organics, pesticides and other toxic chemicals. Numerous studies have evaluated the effects of this contamination on the local ecology. However, few studies have reported the effect of this contamination on vector-borne pathogens and their hosts. In this manuscript, we characterize tick-borne pathogen presence a

Ehrlichia spp. close to Ehrlichia ruminantium, Ehrlichia canis, and "Candidatus Ehrlichia regneryi" linked to heartwater-like disease in Kenyan camels (Camelus dromedarius).

We present findings from an outbreak of a heartwater-like disease in camels that killed at least 2000 adult animals in Kenya in 2016. Clinical signs included excitability, head pressing, aimless wandering, recumbency, and fast breathing followed by death after about 4 days. The observed morbidity in one herd was 40% with an average mortality of 7.5% in animals that received early antibiotic treatments. In untreated adults, the case fatality rate reached 100%. Gross pathology showed pulmonary edema, pleural exudate, hydrothorax, hydropericardium, ascites, enlarged "cooked" liver, nephrosis, and

Arboviruses: How Saliva Impacts the Journey from Vector to Host.

Arthropod-borne viruses, referred to collectively as arboviruses, infect millions of people worldwide each year and have the potential to cause severe disease. They are predominately transmitted to humans through blood-feeding behavior of three main groups of biting arthropods: ticks, mosquitoes, and sandflies. The pathogens harbored by these blood-feeding arthropods (BFA) are transferred to animal hosts through deposition of virus-rich saliva into the skin. Sometimes these infections become systemic and can lead to neuro-invasion and life-threatening viral encephalitis. Factors intrinsic to t

Tick iron and heme metabolism - New target for an anti-tick intervention.

Ticks are blood-feeding parasites and vectors of serious human and animal diseases. Ixodes ricinus is a common tick in Europe, transmitting tick-borne encephalitis, Lyme borreliosis, anaplasmosis, or babesiosis. Immunization of hosts with recombinant tick proteins has, in theory, the potential to interfere with tick feeding and block transmission of pathogens from the tick to the host. However, the efficacy of tick antigens has, to date, not been fully sufficient to achieve this. We have focused on 11 in silico identified genes encoding proteins potentially involved in tick iron and heme metab

Haemoglobin-based blood substitutes: what's on the horizon?

Military and civilian medical organisations have long recognised that an oxygen-carrying volume expander which is easy to store, transport and administer could be life-saving for trauma victims. Concern over fatal blood-borne pathogens including hepatitis and human immunodeficiency virus (HIV), as well as a shrinking donor pool further highlight the need for such a product. A variety of substances that transport oxygen and augment intravascular volume in the absence of red cells are emerging as possible "blood substitutes" for use in humans. These include haemoglobin solutions, liposome encaps…

The Prevalence, Seroprevalence, and Risk Factors of Tick-Borne Encephalitis Virus in Dogs in Lithuania, a Highly Endemic State.

The rising awareness and increasing number of case reports of tick-borne encephalitis (TBE) in dogs indicate that the virus might be an important tick-borne pathogen in dogs, especially in endemic areas. Therefore, the aim of the present study was to investigate the prevalence rate of TBEV RNA and TBEV-specific antibodies in clinical samples of dogs living in a highly endemic region of Lithuania and to evaluate the main risk factors for severe disease course and death. The blood samples (n = 473) of dogs were collected in two veterinary clinics in central Lithuania. Tick-borne encephalitis vir
