Salmonella-induced caspase-2 activation in macrophages: a novel mechanism in pathogen-mediated apoptosis.

The enterobacterial pathogen Salmonella induces phagocyte apoptosis in vitro and in vivo. These bacteria use a specialized type III secretion system to export a virulence factor, SipB, which directly activates the host's apoptotic machinery by targeting caspase-1. Caspase-1 is not involved in most apoptotic processes but plays a major role in cytokine maturation. We show that caspase-1-deficient macrophages undergo apoptosis within 4-6 h of infection with invasive bacteria. This process requires SipB, implying that this protein can initiate the apoptotic machinery by regulating components dist

Bcl-xL is a negative regulator of caspase-3 activation in immature neurons during development.

Caspases and Bcl-xL, the mammalian homologues of the Caenorhabditis elegans (C. elegans) ced-3 and ced-9 genes, respectively, regulate apoptosis of various cells. Caspase-3 is processed into an active form (p20 or p17 and p12) during apoptosis. We investigated the relation between caspase-3 and Bcl-xL during development by examining activation of caspase-3 and apoptotic cells in Bcl-x-deficient (bcl-x(-/-)) mice at embryonic (E) day 11.5. We used a double-staining technique with a cleavage site-directed antibody against caspase-3 (anti-p20/17) and terminal-deoxytransferase-mediated deoxyuridin…

Insulin-like growth factor-I prevents caspase-mediated apoptosis in Schwann cells.

Both neurons and glia succumb to programmed cell death (PCD) when deprived of growth factors at critical periods in development or following injury. Insulin-like growth factor-I (IGF-I) prevents apoptosis in neurons in vitro. To investigate whether IGF-I can protect Schwann cells (SC) from apoptosis, SC were harvested from postnatal day 3 rats and maintained in serum-containing media until confluency. When cells were switched to serum-free defined media (DM) for 12-72 h, they underwent PCD. Addition of insulin or IGF-I prevented apoptosis. Bisbenzamide staining revealed nuclear condensation an

Cuminaldehyde, a natural monocyclic terpenoid, attenuates isoproterenol-induced myocardial infarction in rats via modulation of Bax/Bcl-2/Bcl-xL/Cyt.c/caspase- 9/caspase-3, and MMP-2/MMP-9 pathways.

Myocardial infarction (MI) is a leading cause of death. In this study, we appraised the protective mechanisms of cuminaldehyde, a natural monocyclic terpenoid, in isoproterenol-induced myocardial infarcted rats. Our study included four groups: Group 1: normal control, Group 2: cuminaldehyde (20 mg/kg body weight) alone, Group 3: isoproterenol (100 mg/kg body weight) induced MI, Group 4: MI + cuminaldehyde (20 mg/kg body weight). Cardiac apoptosis and fibrosis were evaluated by biochemical analysis, transmission electron microscopy (TEM), ELISA, RT-PCR, and histopathology to explore the potenti

Structural insights into human caspase-9 CARD assembly modes driven by conserved arginine hotspots.

Caspase-9 (C9) is a key initiator of the intrinsic apoptotic pathway and contains an N-terminal caspase activation and recruitment domain (CARD) that mediates homotypic and heterotypic interactions. While previous studies have shown that C9CARD can form filamentous assemblies under specific conditions, its intrinsic structural properties and interaction modes below saturation concentration remain incompletely understood. Here, we present a 2.4 Å resolution crystal structure of the isolated human C9CARD, revealing a sulfate-mediated dimer-of-dimers assembly involving a conserved arginine-rich s

Berberine induces pyroptosis by promoting the NLRP3/Caspase-1/GSDMD pathway in papillary thyroid carcinoma.

BACKGROUND: PTC (Papillary thyroid carcinoma) is a complex disease. We previously found that BBR (Berberine), an effective component of Chinese herbal medicine Coptis, has an inhibitory effect on PTC. The exact process is not understood, possibly linked to pyroptosis controlled by NLRP3 (Nod-like receptor protein 3). The purpose of our research was to investigate the effects of pyroptosis in BBR treatment of PTC. METHODS: PTC cell lines with different genetic backgrounds, such as BRAF(V600E) mutant (K1, BCPAP) and wild-type (TPC-1) PTC cells, were utilized in in vitro experiments. Pyroptosis

Caspase-1-targeting siRNA nanoparticles suppress pyroptosis to prevent force-induced root resorption.

Mechanical force-induced hard tissue defects are clinically prevalent, particularly in dentistry. Excessive mechanical stimulation could induce pathological tooth root resorption (RR), which leads to tooth mobility and even tooth loss. This process is closely associated with dysregulated biological alterations in adjacent periodontal ligament stem cells (PDLSCs) and disrupted crosstalk between PDLSCs and osteoclast precursors, yet the specific molecular and cellular mechanisms remain insufficiently understood, severely limiting the development of targeted therapeutic strategies for such hard t

Selective cytotoxicity of solamargine via oxidative stress and caspase-independent mechanisms in human glioblastoma cells.

IDH-wild-type glioblastoma (IDH-wildtype GB) is an aggressive and genetically heterogeneous tumor characterized by intrinsic resistance to radiotherapy and chemotherapy, while surgical resection remains inherently limited by its diffuse infiltrative growth, leading to poor clinical outcomes. Natural products such as solamargine (SM), a steroidal glycoalkaloid with cytotoxic and antitumor properties, have emerged as potential adjuvant strategies. Here, we investigated the effects of SM on proliferation, clonogenic survival, morphology and migration of IDH-wildtype GB cell lines (U-87MG, U-251MG

Microvascular autophagy and caspase-3 activation are central regulators of renal fibrosis after ischemia-reperfusion.

Ischemia-reperfusion injury (IRI) is a common cause of acute kidney injury (AKI) leading to renal fibrosis. Here, we investigate the kinetics of autophagy, apoptosis, and necroptosis activation in tubular epithelial cells (TECs) and peritubular capillaries (PTCs) after renal IRI, and their relative contributions to renal fibrogenesis. IRI with renal artery clamping in GFP-LC3 transgenic mice induced a predominant and sustained necroptotic response in TECs, while apoptosis and autophagy played minor roles. PTCs showed early and persistent activation of apoptosis, brief necroptosis induction, an

Potent Caspase-1 Antagonists From Dihydropyridine Derivatives: In Vitro Fluorescence Assays and Cell Imaging.

Caspase-1 is a key marker in pyroptosis and inflammation, playing a central role in the pathogenesis of various acute and chronic inflammatory diseases. While some caspase-1 sensing and inhibition studies have been reported, the use of DHPs for these applications remains unexplored. To address this challenge, a novel fluorescent compound, zWEHD-NH-DHP, was synthesized and subsequently developed as a real-time caspase-1 detection in vitro and in cells. Furthermore, a new class of potential caspase-1 inhibitors was investigated through the screening of a series of nonfluorogenic DHPs, utilizing

Sublytic pyroptosis blocks Schwann cell remyelination through caspase-1-mediated Tyro3 cleavage.

BACKGROUND: Inflammasomes canonically trigger pyroptotic cell death through GSDMD-mediated membrane rupture. This study investigates a distinct sublytic pyroptosis in Schwann cells following peripheral nerve injury, where inflammasome activation causes functional impairment without extensive cell death. METHODS: Using a mouse sciatic nerve transection-repair model combined with in vitro and in vivo analyses, we examined NLRP3 inflammasome activation and cell fate. Primary Schwann cell cultures and RSC96 cells were used for biochemical assays, while ultrastructural analyses were performed on sc

Target degradation of CASPASE-1 for alleviation of inflammation in sepsis via optogenetically engineered extracellular vesicles.

Sepsis is a comprehensive ailment of systemic inflammatory response syndrome arising from infection. Activation of CASPASE-1 plays a central role in initiating the inflammatory cascade during sepsis. Herein, we construct optogenetically engineered extracellular vesicles (EVs) that achieve the specific degradation of CASPASE-1 and inhibit sepsis-associated inflammation. Specifically, blue light (460 nm)-induced CRY2/CIBN heterodimerization was applied during the EVs production stage to selectively load GCE-CTM fusion proteins into EVs by EXPLORs technology, yielding EVsGCE-CTM loading efficienc
