Mesenchymal Neoplasms of the Kidney and Perinephric Soft Tissue.

Mesenchymal neoplasms of the kidney present challenges because they are uncommon, and because perinephric soft tissue biopsies are sometimes submitted as "kidney" masses, causing diagnostic confusion. Here, the author thoroughly reviews mesenchymal neoplasms of the kidney, including metanephric stromal tumor, classic and cellular congenital mesoblastic nephroma, anaplastic sarcoma and clear cell sarcoma of the kidney, malignant rhabdoid tumor, PEComa/angiomyolipoma, and anastomosing hemangioma. The author also discusses perinephric myxoid pseudotumor of fat, as well as diagnostic pitfalls pres

Retardation of finger growth after injury to the flexor tendons.

The absence of any part of the total mechanical force (i.e., flexor tendons) may be responsible for retardation of bone growth during a child's growth phase. The cause of retardation may be dedifferentiation of cartilage cells at the epiphyseal plate. Four cases in which unrepaired flexor tendons in digits caused retardation of growth illustrate this possible phenomenon. We feel that this is not a well recognized problem and thus deserves mention to stimulate further study. We believe that meticulous primary repair in children is desirable not only to preserve function, but also to prevent gro

Multifunctional Bismuth Nanoplatforms Augment Radioactive Iodine Therapy in Anaplastic Thyroid Cancer.

INTRODUCTION: Radioactive iodine (RAI) therapy is a highly specific targeted treatment for thyroid cancer. However, the intrinsic low energy of 131I limits its efficacy in tumor eradication. Additionally, certain thyroid cancers exhibit a loss of sodium/iodine symporter (NIS) function due to severe dedifferentiation, compromising the therapeutic effectiveness of RAI. METHODS: Our work was based on two distinct RAI-sensitizing strategies: (1) the generation of secondary electrons by irradiated metallic nanomaterials to promote hydrolysis and enhance reactive oxygen species (ROS) production, an

SOX9 keeps growth plates and articular cartilage healthy by inhibiting chondrocyte dedifferentiation/osteoblastic redifferentiation.

Cartilage is essential throughout vertebrate life. It starts developing in embryos when osteochondroprogenitor cells commit to chondrogenesis, activate a pancartilaginous program to form cartilaginous skeletal primordia, and also embrace a growth-plate program to drive skeletal growth or an articular program to build permanent joint cartilage. Various forms of cartilage malformation and degeneration diseases afflict humans, but underlying mechanisms are still incompletely understood and treatment options suboptimal. The transcription factor SOX9 is required for embryonic chondrogenesis, but it

Time course of pathogenic and adaptation mechanisms in cystinotic mouse kidneys.

Cystinosis, a main cause of Fanconi syndrome, is reproduced in congenic C57BL/6 cystinosin knockout (KO) mice. To identify the sequence of pathogenic and adaptation mechanisms of nephropathic cystinosis, we defined the onset of Fanconi syndrome in KO mice between 3 and 6 months of age and analyzed the correlation with structural and functional changes in proximal tubular cells (PTCs), with focus on endocytosis of ultrafiltrated disulfide-rich proteins as a key source of cystine. Despite considerable variation between mice at the same age, typical event sequences were delineated. At the cellula

Lrig1 marks a population of gastric epithelial cells capable of long-term tissue maintenance and growth in vitro.

The processes involved in renewal of the epithelium that lines the mouse stomach remain unclear. Apart from the cells in the isthmus, several other populations located deeper in the gastric glands have been suggested to contribute to the maintenance of the gastric epithelium. Here, we reveal that Lrig1 is expressed in the basal layer of the forestomach and the lower part of glands in the corpus and pylorus. In the glandular epithelium of the stomach, Lrig1 marks a heterogeneous population comprising mainly non-proliferative cells. Yet, fate-mapping experiments using a knock-in mouse line expre

Simulating the Dedifferentiation Process of Thyroid Cancer: Insights from Mouse Models and Ultrasound Imaging.

RATIONALE AND OBJECTIVE: Anaplastic thyroid carcinoma (ATC) may arise from the progression of differentiated thyroid cancers, but the mechanisms are not well understood. Establishing appropriate animal models and conducting ultrasound imaging monitoring can facilitate the study of this dedifferentiation process. MATERIALS AND METHODS: We constructed mouse models of papillary thyroid carcinoma (PTC) and ATC by editing the Braf mutation and Trp53 deletion, and monitored the process using small-animal ultrasound. The you only look once computer model was employed to process ultrasound images, an

SMAD3/Stat3 Signaling Mediates β-Cell Epithelial-Mesenchymal Transition in Chronic Pancreatitis-Related Diabetes.

Many patients with chronic pancreatitis develop diabetes (chronic pancreatitis-related diabetes [CPRD]) through an undetermined mechanism. Here we used long-term partial pancreatic duct ligation (PDL) as a model to study CPRD. We found that long-term PDL induced significant β-cell dedifferentiation, followed by a time-dependent decrease in functional β-cell mass-all specifically in the ligated tail portion of the pancreas (PDL-tail). High levels of transforming growth factor β1 (TGFβ1) were detected in the PDL-tail and were mainly produced by M2 macrophages at the early stage and by activated …

Adipogenesis induces growth inhibition of dedifferentiated liposarcoma.

Well-differentiated liposarcoma (WDLPS) and dedifferentiated liposarcoma (DDLPS) are the most common types of liposarcoma. Although WDLPS and DDLPS patients receive intensive treatment including radical surgery and systemic therapy, their overall 5-year survival rates are 90% and 30%, respectively, indicating that DDLPS is clinically more aggressive. We examined whether adipogenic stimulation induces adipogenesis in human WDLPS/DDLPS cells by using dexamethasone, indomethacin, insulin, and 3-isobutyl-1-methylxanthine (IBMX), all putative medications or drugs. Functional in vitro experiments sh

Taurine Promotes the Cartilaginous Differentiation of Human Umbilical Cord-Derived Mesenchymal Stem Cells in Vitro.

Taurine has been reported to influence osteogenic differentiation, but the role of taurine on cartilaginous differentiation using human umbilical cord-derived mesenchymal stem cells (hUC-MSCs) remains unclear. In this study, we investigated the effect of taurine (0, 1, 5 and 10 mM) on the proliferation and chondrogenesis of hUC-MSCs by analyzing cell proliferation, accumulation of glycosaminoglycans and expression of cartilage specific mRNA. The results show though taurine did not affected the proliferation of hUC-MSCs, 5 mM of taurine is sufficient to enhanced the accumulation of glycosaminog

Malignant Melanoma With Transdifferentiation Into a Ganglioneuroblastic Phenotype After Anti-PD-1 Therapy.

The clinicopathologic spectrum of dedifferentiated, undifferentiated, and transdifferentiated melanoma occurring de novo or following systemic therapy is gaining recognition. Here, we present a patient with locally advanced cutaneous melanoma who received neoadjuvant anti-PD-1 therapy and whose post-treatment excisional pathology revealed transdifferentiation into a ganglioneuroblastic phenotype. This rare phenotype is hypothesized to be connected to melanoma's shared origin from pluripotent neural crest cells. This case report highlights the proposed pathogenic mechanisms underlying this tran

A protoplast-based method to visualize early cell biological events in plant cellular reprogramming and regeneration.

Plant developmental biology lacks cell-based experimental systems comparable to the organoids and live-imaging platforms that have transformed mechanistic discovery in animal research. To address this gap, we present a robust, trackable protoplast regeneration platform in Arabidopsis thaliana that enables high-resolution, time-resolved analysis from the single-cell stage through microcolony formation and early regenerative development. Protoplasts are embedded in thin alginate matrices containing fluorescent fiducial beads, maintaining physical separation and allowing repeated return to the sa

FoxO1 Deacetylation Decreases Fatty Acid Oxidation in β-Cells and Sustains Insulin Secretion in Diabetes.

Pancreatic β-cell dysfunction contributes to onset and progression of type 2 diabetes. In this state β-cells become metabolically inflexible, losing the ability to select between carbohydrates and lipids as substrates for mitochondrial oxidation. These changes lead to β-cell dedifferentiation. We have proposed that FoxO proteins are activated through deacetylation-dependent nuclear translocation to forestall the progression of these abnormalities. However, how deacetylated FoxO exert their actions remains unclear. To address this question, we analyzed islet function in mice homozygous for knoc…

Mycobacterium leprae-induced nerve damage: direct and indirect mechanisms.

Leprosy is a chronic infectious disease caused by Mycobacterium leprae. This disease is characterized by skin and peripheral nerve trunk damage. The mechanisms responsible for the observed nerve damage in leprosy could be directly related to the ability of M. leprae to infect Schwann cells, leading to triggering of signaling events. Therefore, we hypothesize that in response to M. leprae infection, activation of the Notch signaling pathway in Schwann cells could play a crucial role in glial cell dedifferentiation. On the other hand, nerve damage evidenced in this disease may be additionally ex…
