Changes in Methionine Metabolism and Histone H3 Trimethylation Are Linked to Mitochondrial Defects in Multiple Sclerosis.

UNLABELLED: Mitochondrial changes, including decreased expression of electron transport chain subunit genes and impaired energetic, have been reported in multiple sclerosis (MS), but the mechanisms involved in these changes are not clear. To determine whether epigenetic mechanisms are involved, we measured the concentrations of methionine metabolites by liquid chromatography tandem mass spectrometry, histone H3 methylation patterns, and markers of mitochondrial respiration in gray matter from postmortem MS and control cortical samples. We found decreases in respiratory markers as well as decre

Fe(III) Mineral Encrustation Uncouples Respiration from Reproduction of the Nitrate-Reducing Fe(II)-Oxidizing Bacterium Acidovorax sp. BoFeN1.

Fe(II) can be oxidized by mixotrophic nitrate-reducing Fe(II)-oxidizing (NRFeOx) bacteria to Fe(III), leading to mineral precipitation. However, the effects of this process on the metabolism and reproductive ability of Fe(II)-oxidizing bacterial cells have not yet been quantified. In this study, we investigated the effects of Fe(II) on the mixotrophic NRFeOx bacteria Acidovorax sp. BoFeN1 by performing cell plate counts, chemical analyses, scanning electron microscopy (SEM) imaging, live/dead fluorescence staining, and proteomic analysis. The results showed that nitrate-reducing Fe(II)-oxidizi

An Integrated Methodology to Quantify the Glycolytic Stress in Plasma Cell Myeloma in Response to Cytotoxic Drugs.

Multiple myeloma (MM) is an incurable plasma cell malignancy primarily localized within the bone marrow (BM). Myeloma plasma cells, like many other cancer cells, change their metabolism in response to internal and external stimuli. The main metabolic alterations of MM cells include deregulated glycolysis (commonly associated with enhanced uptake and utilization of glucose), lipid metabolism dysregulation, as well as deregulated mitochondrial respiration (commonly associated with the deregulated formation of reactive oxygen species). Over the past decade, the discovery of novel methodologies an

Childhood trauma in women with HIV is associated with increased mitochondrial respiration despite reduced mitochondrial DNA copy number.

Early life stress (ELS) is highly prevalent among women with HIV (WWH) and contributes to risk for brain health disorders. Mitochondria represent key regulators of cellular stress processes and may represent a key mechanism linking ELS to biological vulnerability. While mitochondrial dysfunction has been associated with ELS in other populations, it remains unknown whether ELS is related to multidimensional mitochondrial health in WWH. Thus, we examined associations between ELS and mitochondrial health metrics in WWH. Sixty-one WWH completed the Childhood Trauma Questionnaire (CTQ) and provided

The Contribution of Phospholipase A2 and Metalloproteinases to the Synergistic Action of Viper Venom on the Bioenergetic Profile of Vero Cells.

Increasing concern about the use of animal models has stimulated the development of in vitro cell culture models for analysis of the biological effects of snake venoms. However, the complexity of animal venoms and the extreme synergy of the venom components during envenomation calls for critical review and analysis. The epithelium is a primary target for injected viper venom's toxic substances, and therefore, is a focus in modern toxinology. We used the Vero epithelial cell line as a model to compare the actions of a crude Macrovipera lebetina obtusa (Levantine viper) venom with the actions of

Effect of Chicken Bone Extracts on Metabolic and Mitochondrial Functions of K562 Cell Line.

BACKGROUND: Tetracyclines' use in intensive animal farming has raised some concerns regarding the biosafety for humans. Increasing evidences have revealed the presence of these drugs in processed animal by-products, such as bone, throughout the food chain. A potential off-target of tetracyclines is the bacterial-like mitochondrial translational machinery, thereby causing proteostatic alterations in mitochondrial DNA-encoded components of the oxidative phosphorylation system. METHODS: The Seahorse methodology, confocal microscopy imaging of mitochondrial potential and reactive oxygen species,

Hypothermia Alleviates Reductive Stress, a Root Cause of Ischemia Reperfusion Injury.

Ischemia reperfusion injury is common in transplantation. Previous studies have shown that cooling can protect against hypoxic injury. To date, the protective effects of hypothermia have been largely associated with metabolic suppression. Since kidney transplantation is one of the most common organ transplant surgeries, we used human-derived renal proximal tubular cells (HKC8 cell line) as a model of normal renal cells. We performed a temperature titration curve from 37 °C to 22 °C and evaluated cellular respiration and molecular mechanisms that can counteract the build-up of reducing equivale

Piceatannol alleviate ROS-mediated PC-12 cells damage and mitochondrial dysfunction through SIRT3/FOXO3a signaling pathway.

Oxidative stress-associated mitochondrial dysfunction has been identified as a major mechanism in multiple neurodegenerative diseases. This study aims to investigate the cytoprotective effects of piceatannol on ROS-mediated PC-12 cells damage and related mitochondrial dysfunction. Piceatannol treatment could significantly attenuate PC-12 cells oxidative damage and ROS-mediated cells apoptosis. Moreover, pretreatment with piceatannol effectively decreased mitochondrial membrane depolarization, cleaved-caspase 3, and increased Bcl-2 and Bcl-2/Bax compared with control H2 O2 group. Meanwhile, pic

Isolation of a carbon disulfide utilizing Thiomonas sp. and its application in a biotrickling filter.

The carbon disulfide (CS2)-oxidizing bacterium Thiomonas sp. WZW was enriched and isolated using activated sewage sludge as inoculum. Growth of Thiomonas sp. WZW was observed on CS2, thiosulfate, dimethylsulfide (DMS), dimethyldisulfide (DMDS), and H2S. No growth occurred on dimethylsulfoxide, methanol, acetate, and on complex media with glucose, yeast extract, or tryptone. DMDS-grown cells respired CS2, DMS, and DMDS, while thiosulfate-grown cells did not respire CS2. Chemostat cultures growing on thiosulfate could be rapidly adapted to growth on CS2. Growth was observed between pH 6 and 8. T

Peripheral chemoreceptors in congenital central hypoventilation syndrome.

Congenital central hypoventilation syndrome (CCHS) is a rare disorder of unknown etiology, characterized by failure of the autonomic control of respiration. The primary defect is believed to involve central respiratory control; however, no specific lesion has been identified. We report two cases of CCHS (one female, 3 mo of age and one male 2 yr of age) in which there was detailed examination of the neural, muscular, and chemoreceptor components of respiratory control. Although no specific abnormalities were identified in the central nervous system (CNS) or muscles of respiration, striking cha
