Motoneurons Inhibitory Synapses Homeostatically Respond to Neuronal Activity and Modulate Amyotrophic Lateral Sclerosis Pathogenesis.

Alterations in excitation/inhibition (E/I) balance and changes in motor neurons (MN) activity may contribute to MN vulnerability in ALS. The balance of pathogenic versus adaptive changes occurring in inhibitory synapses and affecting E/I balance remain unclear. Confocal microscopy of MN from P45 male SOD1G93A mice reveal downregulated GlyR but upregulated GABAR clusters at inhibitory synapses. GlyR and GABAR respond to PSAM and DREADD chemogenetic alterations of MN excitability, with increased activity driving increase in inhibitory clusters. An E3 ligase-conjugated intrabody (GFE3) degrades G

Dentate gyrus network regulation by somatostatin- and parvalbumin-expressing interneurons differentially impacts spatial memory processing.

GABAergic interneurons regulate circuit dynamics of memory-processing brain structures. However, the role of the dentate gyrus (DG) and its predominant interneuron subtypes - somatostatin-expressing (SST+) and parvalbumin-expressing (PV+) - in different stages of memory processing is unknown. We tested how the chemogenetic manipulation of DG SST+ and PV+ interneurons in mice influences encoding, consolidation, and retrieval of hippocampus-dependent object-location memory (OLM). The activation of DG SST+ interneurons impairs OLM encoding and retrieval, dramatically suppresses DG granule cell ac

Disrupted temperature-sleep coupling mechanism in a Dravet syndrome mouse model.

Dravet syndrome (DS) is associated with epilepsy, developmental delays, thermal dysregulation, and sleep disturbances. While seizures have been linked to hippocampal dysfunction, what drives sleep disturbances and thermal dysregulation is poorly understood. Using DS mice (Scn1aA1783V), we identified a link between sleep and thermoregulation. We found that DS mice exhibited lower core body temperature. Next, using electrocorticography, local field potential recordings, and core temperature monitoring, we showed that DS mice exhibited a lack of core temperature change during the transition from

The glutamatergic NTS-PAG/PVN pathway underlies myocardial injury and anxiety-like behavior induced by myocardial ischemia-reperfusion.

For patients with myocardial ischemia-reperfusion injury (MIRI), cardiac dysfunction accompanied by anxiety is an extremely common comorbidity. Although the association between these two conditions has received some attention, the underlying neural mechanisms remain unclear. Here, we functionally characterized two distinct glutamatergic (Glu) neural pathways in the nucleus tractus solitarius (NTS) of male mice, a pathway from the NTS to the periaqueductal gray (PAG) that mediates anxiety-like behaviors in MIRI mice, and a pathway from the NTS to the paraventricular nucleus of the hypothalamus

The role of supraoptic hypothalamic arginine vasopressin neurons in aging-associated water balance and thermoregulatory deficits in male mice.

Aging disrupts physiological homeostasis, impairing thermoregulation, metabolism, and water balance, but the underlying neural mechanisms remain unclear. Here, we identify arginine vasopressin (AVP) neurons in the supraoptic nucleus (SON) of the hypothalamus as a critical driver of these changes in male mice. Single-nucleus RNA-sequencing revealed Avp among the most upregulated neuronal transcripts with age. Aged SONAVP neurons displayed enlarged size and heightened excitability, features consistent with hyperactivity. Chemogenetic activation of SONAVP neurons in young mice reproduced aging-as

Early-Stage Corticostriatal Hyperactivity Impairs Cognitive Flexibility Alongside Striatal Cholinergic Dysfunction in an Alzheimer's Disease Model.

Cognitive flexibility declines early in Alzheimer's disease, yet the underlying circuit mechanisms remain unknown. Here, we report that young 5xFAD mice exhibit deficits in instrumental reversal learning prior to spatial memory impairment. This behavioral inflexibility is associated with abnormal neuronal reactivation in the medial prefrontal cortex and dorsomedial striatum. Electrophysiological recordings reveal that medial prefrontal cortex neurons are hyperexcitable and receive increased excitatory input. Furthermore, glutamatergic transmission from the medial prefrontal cortex to striatal

Vagal subcircuits differentially regulate sepsis-induced sickness and anxiety-related behaviors.

The brain monitors peripheral immune activity via vagus nerve sensory neurons, whose cell bodies lie in the nodose ganglia and project to the brainstem dorsal vagal complex. This relay activates neural circuits mediating infection-related behavioral changes, such as sickness behavior. In severe inflammation like sepsis, this signaling can contribute to brain dysfunction, including persistent anxiety. Using the rodent sepsis model of cecal ligation and puncture, we investigated how vagal subcircuits contribute to these effects, by integrating surgical and chemogenetic gain- and loss-of-function

Human retinal organoids functionally bridge a transected optic nerve in a rat model.

Functional restoration of vision in adult mammals remains unattainable following complete transection of the optic nerve. In this study, we address this challenge by heterotopically transplanting human retinal organoids into the transection cavity of the optic nerve in adult rats. One month after transplantation, treated animals exhibited partial recovery of visual function, including a direct pupillary light reflex, visually evoked electrophysiological responses, and visually guided behaviors, which were sustained for at least 3 months. Neuroanatomical tracing and three-dimensional imaging re

Auricular vagus nerve stimulation drives analgesia via an auricle-brain axis in a mouse model of neuropathic pain.

Despite its long-recognized effects in relieving pain, the neural substrates of auricular stimulation remain elusive. Here, we show that trans-auricular vagus nerve stimulation (taVNS), i.e., electrical stimulation of the auricular concha, effectively induces analgesia in a mouse model of neuropathic pain. Viral tracing, microendoscopic calcium imaging, and multi-electrode recordings reveal that auricular vagal signals travel to the jugular-nodose ganglia (JNG), which in turn connect to pro-opiomelanocortinergic neurons in nucleus tractus solitarius (NTS), subsequently activating glutamatergic

Impaired brain-gut circuit for gut motility dysfunction in mouse models of Parkinson's disease.

Gut motility dysfunction is a common symptom of Parkinson's disease (PD), but its central mechanisms remain unclear. Here, we utilize multiple mouse models, which mimic both genetic and sporadic PD, to reproduce PD-related gut motility dysfunction. We find that α-synuclein pathology significantly impairs the excitability of cholinergic neurons in the dorsal motor nucleus of the vagus (DMV). Chemogenetic activation of these neurons effectively restores gut motility in PD models, revealing a causal link between DMV neuronal hypoexcitability and gut motility dysfunction. Notably, electroacupunctu

RNA-mediated condensation of TFE3 oncofusions facilitates transcriptional hub formation to promote translocation renal cell carcinoma.

Transcription factor E3 (TFE3) oncofusions are frequently detected in the Microphthalmia transcription factor (MiT) family translocation renal cell carcinoma (tRCC), a rare pediatric renal cancer with limited treatment options. The mechanisms by which TFE3 oncofusions promote tRCC malignancy remain inadequately defined. Here, we demonstrate that the RNA-binding capability conferred by TFE3 fusion partners drives the formation of TFE3 condensates. This further enables TFE3 oncofusions to co-condensate with RNA polymerase II (RNAPII) and other RNA-binding proteins, such as paraspeckle component

Moderate UVB exposure ameliorate chronic stress-induced anxiety and social impairment by activating mPFC to basal lateral amygdala pathway.

Ultraviolet radiation B (UVB), the most biologically active ultraviolet ray in sunlight, exert broad effects on physiological and behavioral functions, including circadian rhythm, mood, and cognition. However, its underlying mechanisms are still unknown. In this study, in order to verify effects of UVB on anxiety and social behaviors, C57BL/6 mice receiving 2 h UVB exposure after chronic restraint stress were used. UVB exposure improved anxiety-like behaviors and social activities in normal and restraint stressed mice. Meanwhile, UVB exposure increased the neural excitability in mPFC according

nNOS-expressing neurons in the mPFC mediate depression-related behaviors in mice through pPVT-mPFC projections.

Depression is one of the most common mental disorders, but its etiology remains poorly understood. Neuronal nitric oxide synthase (nNOS) has been implicated in depression, but the role of nNOS-expressing neurons is still unknown. We used chemogenetic strategies to show that nNOS-expressing neurons in the medial prefrontal cortex (mPFC) are essential for depression-related behaviors. Using electrophysiology, we determined that mPFC nNOS-expressing neurons receive projections from the posterior subregion of the paraventricular thalamic nucleus (pPVT). We show that excitatory projections from the

Neuropathic Pain-Like Responses in a Chronic CNS Injury Model Are Mediated by Corticospinal-Targeted Spinal Interneurons.

Chronic neuropathic pain is a persistent and debilitating outcome of traumatic central nervous system injury, affecting up to 80% of individuals. Postinjury pain is refractory to treatments due to the limited understanding of the brain-spinal cord circuits that underlie pain signal processing. The corticospinal tract (CST) plays critical roles in sensory modulation during skilled movements and tactile sensation; however, a direct role for the CST in injury-associated neuropathic pain is unclear. Here we show that complete, selective CST transection at the medullary pyramids leads to hyperexcit

Functional synaptic connectivity of engrafted spinal cord neurons with hindlimb motor circuitry in the injured spinal cord.

Spinal cord injury (SCI) results in significant neurological deficits, and curative therapies are lacking. Neural progenitor cell (NPC) transplantation shows promise, as graft-derived neurons (GDNs) can integrate into host spinal cord and support axon regeneration. Here, we examined the synaptic integration of GDNs into hindlimb motor circuits in a mouse thoracic contusion SCI model. Transsynaptic tracing revealed that GDNs form synaptic connections with host motor circuits. Axon mapping showed distinct termination patterns of cholinergic and V2a interneurons within host spinal cord. Chemogene

Brainstem noradrenergic modulation of the kisspeptin neuron GnRH pulse generator in mice.

Brainstem noradrenaline (NA) neurons modulate the activity of many neural networks including those responsible for the control of fertility. Using brain slice electrophysiology, we demonstrate that the arcuate nucleus kisspeptin (ARNKISS) neurons, recently identified to be the gonadotropin-releasing hormone (GnRH) pulse generator, are directly hyperpolarized by NA through both alpha 2- and beta-adrenergic receptors. Retrograde viral tracing shows that NA innervation of the ARN is primarily from the dorsal subdivision of the locus coeruleus (LC)-NA cell group and is substantially greater in fem

Brainstem circuit for sickness-induced sleep.

Increased sleep induced by immune activation plays a crucial role in facilitating recovery from illness. However, the neural mechanisms underlying sickness-induced sleep remain poorly understood. Here, we identify a brainstem circuit originating in the nucleus of the solitary tract (NST) that mediates sickness-induced nonrapid eye movement (NREM) sleep. Using activity-dependent genetic labeling, we tagged NST neurons activated by lipopolysaccharide (LPS) injection and showed that their chemogenetic activation strongly promotes NREM sleep. These NST neurons project extensively to the parabrachi

A brain-to-small intestine circuit mediates morphine-induced constipation in male mice.

Opioid-induced constipation is one of the most common and persistent side effect of opioid analgesics, yet the underlying neural mechanism(s) remain unclear. Here we show morphine-induced constipation is mediated by a neural circuit from glutamatergic neurons in the paraventricular nucleus of hypothalamus (PVNGlu) to acetylcholinergic neurons in the dorsal motor nucleus of the vagus (DMVAch), and subsequently to the small intestine in mice. Microendoscopic calcium imaging revealed morphine inhibits the PVNGlu→DMVAch→small intestine circuit, and this is accompanied by decreased small intestinal

Modeling craniopharyngioma for drug screening reveals a neuronal mechanism for tumor growth.

Tumors occurring along the hypothalamus-pituitary axis receive axonal projection from neuroendocrine neurons, but it remains unclear whether neuroendocrine neuronal activity drives tumor expansion. Craniopharyngioma is a common suprasellar tumor with a propensity for invading the hypothalamus, leading to devastating endocrine and metabolic disorders. Here, we developed two autochthonous animal models that faithfully recapitulate the molecular pathology, clinical manifestations, and transcriptomic profiles of papillary craniopharyngioma. Using high-throughput drug screening, we identified 74 co

Days-old zebrafish rapidly learn to recognize threatening agents through noradrenergic and forebrain circuits.

Animals need to rapidly learn to recognize and avoid predators. This ability may be especially important for young animals due to their increased vulnerability. It is unknown whether, and how, nascent vertebrates are capable of such rapid learning. Here, we used a robotic predator-prey interaction assay to show that 1 week after fertilization-a developmental stage where they have approximately 1% the number of neurons of adults-zebrafish larvae rapidly and robustly learn to recognize a stationary object as a threat after the object pursues the fish for ∼1 min.
