BACKGROUND: Currently, transvitreal fine-needle aspiration biopsy is the most widely used tissue biopsy technique in cases of suspected intraocular lymphoma due to its relative simplicity and low trauma. The small sample produced, however, may be inadequate for diagnostic and prognostic analyses due to mechanical artefacts, insufficient material, or sampling errors. Small case series have demonstrated choroidal biopsy via vitrectomy to be safe and effective. With smaller-gauge vitrectomy instruments, visual recovery is rapid, and post-operative inflammation and conjunctival scarring is minimis…

PURPOSE: To describe the histopathologic features of an early case of presumably bilateral polypoidal choroidal vasculopathy (PCV) in two eyes obtained at autopsy from a patient with no prior ocular therapy.
OBSERVATIONS: The choroid of both eyes at the macular and peripapillary regions was greatly thickened with dilated, thin walled choroidal venules intertwining with arteriosclerotic arterioles in the Sattler's layer of the choroidal vasculature. At the temporal and nasal equatorial regions of both eyes many of these congested venular channels abruptly disappeared and were replaced by loose…

We report a case of enucleation for painful blind eye secondary to recurrent bleeding from choroidal neovascularisation in an eye that was irradiated following presumed metastatic breast carcinoma to the choroid. A 58-year-old woman with a history of treated breast malignancy presented with haemophthalmus and intractable glaucoma in the right eye. One year previously she had presented with right vitreous haemorrhage with subretinal mass that had been irradiated given her history of breast carcinoma. Following irradiation, vitrectomy was performed to clear the blood. Intraoperative and postoper…

AIMS: To determine whether choroidal thickness influences the risk of clinically significant pseudophakic cystoid macular oedema (CME).
METHODS: We performed a retrospective case-control study of non-diabetic adults who underwent uncomplicated cataract surgery at the Eye & ENT Hospital of Fudan University between June 2023 and December 2024. Electronic records were reviewed to estimate pseudophakic CME incidence. For each CME case diagnosed, age-matched and sex-matched patients without CME within 3 months postoperatively were randomly selected as controls. Potential ocular risk factors, inclu…

Brain barriers, cerebrospinal fluid (CSF) dynamics, and peripheral factors are implicated as significant contributors to Alzheimer's disease (AD). The choroid plexus (ChP) is a blood-brain interface that produces CSF and forms the blood-CSF barrier. However, how ChP pathology develops across the lifespan and contributes to AD has not been systematically characterized. Here, we report a multi-modal ChP atlas integrating single-nucleus transcriptomics from 49 individuals, AI-assisted quantitative histopathology across >500 postmortem samples age 16 to 105, spatial transcriptomics, and functional…

Functioning both as the site of cerebrospinal fluid production and as an interface between the peripheral circulation and the central nervous system, the choroid plexus (ChP) modulates the gating of immune cells and signals from the periphery as they pass from the blood into cerebrospinal fluid. We generated a single-nucleus and spatial transcriptomic atlas of the adult human ChP, in donors with and without Alzheimer's disease (AD), revealing transcriptionally and spatially distinct macrophage states. We identified a previously unrecognized population of titin-expressing (TTN+) tissue-resident…

Choroid plexus carcinoma (CPC) is a class of rare malignant life-threatening pediatric brain tumors, with a median survival of ~2.5-3 years [1, 2]. Recent advances in human genomics, cellular studies, and animal models have improved our understanding of the pathophysiology and molecular characteristics of CPC. However, because this cancer is rare, it remains understudied. This limited research has led to a limited understanding of the tumor's heterogeneity and molecular pathogenesis, resulting in fewer treatment options. Surgical resection remains the most significant contributor to patient su…

Choroid plexus is a major site for cerebrospinal fluid (CSF) production, characterized by a multiciliated epithelial monolayer that regulates CSF production. We demonstrate that defective choroid plexus ciliogenesis or Intraflagellar transport yields neonatal hydrocephalus, at least in part, due to increased water channel Aqp1 and ion transporter Atp1a2 expression. We demonstrate choroid plexus multicilia as sensory cilia, transducing both canonical and non-canonical Shh signaling. Interestingly, it is the non-canonical Shh signaling that represses Aqp1 and Atp1a2 expression by Smo/Gαi/cAMP pa…

BACKGROUND: Choroid plexus tumors (CPTs) are rare epithelial brain tumors. Recent studies have found changes in the cells and the presence of amyloid, but these tiny details are still not well understood. Limited information is available regarding the biology of oncocytic changes (OCs) in CPTs.
MATERIAL AND METHODS: This study included 42 primary and recurrent CPTs. Out of these, 28 cases (67%) were grade I choroid plexus papilloma (CPP), eight cases (19%) were grade II atypical choroid plexus papilloma (ACPP), and six cases (14%) were grade III choroid plexus carcinomas (CPCs), based on the …

BACKGROUND: The choroid plexus plays an important role in brain homeostasis, including the active secretion of cerebrospinal fluid. Its function and structure have been reported to be affected by normal ageing. However, existing measures of choroid plexus volume may be complicated by partial volume (in vivo MRI) and tissue fixation artefacts (histology). In this study, we investigate possible changes in choroid plexus volume within the lateral ventricles of aged mice utilising two structural MRI protocols explicitly designed for time-efficient, high-resolution in vivo imaging of the choroid pl…

Investigation of the choroid plexus in schizophrenia has seen growing interest due to its role in the interaction between neuroinflammation and brain dysfunction. Most previous studies included treated and long-term ill patients, while antipsychotics and illness course might both affect the choroid plexus. Here, we recruited first-episode antipsychotic-naïve schizophrenia patients, performed high-resolution structural brain scan and manually extracted choroid plexus volume. Choroid plexus volume was compared between patients and healthy controls after controlling for age, sex and intracranial …

Paclitaxel is a widely used chemotherapeutic agent for treating various solid tumors. However, resulting neuropathic pain, often a lifelong side effect of paclitaxel, can limit dosing and compromise optimal treatment. The choroid plexus, located in the brain ventricles, spreads peripheral inflammatory reactions into the brain. Our study is the first to analyze the effects of paclitaxel on inflammatory alterations in the choroid plexus. We hypothesized that the choroid plexus could respond directly to paclitaxel and simultaneously be indirectly altered via circulating damage-associated molecula…
