INTRODUCTION: The use of intensified chemotherapy has significantly improved overall survival (OS) and relapse-free survival (RFS) in cancer patients. However, it also increases the risk of chemotherapy-induced neutropenia (CIN). Granulocyte colony-stimulating factor (G-CSF) is commonly used to hasten neutrophil recovery, but limited research has focused on the superiority of different routes of G-CSF administration.
MATERIALS AND METHODS: This was a randomized, prospective, single-institution study. Twenty-eight participants were enrolled and randomly assigned to experimental and control arm

BACKGROUND: Positron emission tomography is a useful tool for quantitative imaging of neuroinflammation in human subjects; however, more suitable radiotracers are sought. One of the promising targets for imaging neuroinflammation is macrophage colony-stimulating factor 1 receptor (CSF1R) because of its expression on activated microglia. Several PET radiotracers for imaging CSF1R have been developed, however, a suitable CSF1R radiotracer radiolabeled with 18F is not yet available. We synthesized and performed preclinical evaluation of a new series of high-affinity, 18F-labeled PET radiotracers

Immunoisolation devices containing therapeutic protein-secreting cells offer potential for long-term therapy without immune suppression. However, scar tissue formation driven by the foreign body response (FBR) hinders nutritional exchange and ultimately leads to graft failure. We previously showed that inhibiting the colony-stimulating factor-1 receptor (CSF1R) pathway in monocytes and macrophages can block the FBR to implanted materials. Here, we demonstrate that coencapsulation of slow-releasing CSF1R inhibitor (GW2580) crystals with human stem cell-derived β cells (SC-β) in alginate spheres

BACKGROUND: Bronchopulmonary dysplasia (BPD) remains a leading cause of morbidity and long-term respiratory complications in preterm neonates. Effective strategies for moderate-severe BPD are currently lacking.
METHODS: Tracheal aspirate (TA) samples were collected from 62 preterm neonates, and the immune cell composition was analyzed using multicolor flow cytometry. Subsequently, CD14+ monocytes were isolated and purified from the TA samples for transcriptomic sequencing analysis. Cytokine arrays and enzyme-linked immunosorbent assays were utilized in the cytokine level detection from TA sam

Microglia, the resident macrophages of the brain, are derived from the yolk sac and colonize the brain before the blood-brain barrier forms. Once established, they expand locally and require Colony-stimulating-factor-1 receptor (CSF1R) signaling for their development and maintenance. CSF1R inhibitors have been used extensively to deplete microglia in the healthy and diseased brain. In this study, we demonstrated sex-dependent differences in the microglial response to the CSF1R inhibitor PLX3397. Male mice exhibited greater microglial depletion compared to females. Transcriptomic and flow cytom

BACKGROUND: With the rising prevalence of osteoarthritis (OA), this disease places a substantial economic burden on society. Nevertheless, there is still a lack of effective therapeutic agents to completely cure OA. This study thus aims to identify potential genetic targets that might influence the progression of OA.
METHODS: Using Mendelian randomization (MR), the correlation between the levels of macrophage colony-stimulating factor (CSF-1) and its associated genetic loci with OA at different anatomical sites was investigated. Data were primarily obtained from the arcOGEN Consortium, the UK

Colony-stimulating factors (CSFs) play critical roles in the host response to tissue injury and pathogenic infection. In this study, we conducted a comprehensive genome-wide analysis and identified seven csf genes in rainbow trout (Oncorhynchus mykiss). Comparative analyses of gene structure, conserved synteny, and protein sequence alignment further supported their annotation and orthology with other vertebrate csf genes. Although csf3 has been identified in several teleost species, its biological functions remain largely uncharacterized. Here, we reported that rainbow trout csf3 (Rtcsf3) gene…

PURPOSE: This study aimed to assess the predictive value of macrophage colony-stimulating factor (M-CSF) in the first trimester for hypertensive disorders complicating pregnancy (HDCP) and its association with disease severity and adverse pregnancy outcomes. HDCP pose significant risks to both maternal health and fetal health. M-CSF is implicated in the pathogenesis of HDCP by promoting inflammation and endothelial damage.
METHODS: Serum levels of M-CSF were measured using an enzyme-linked immunosorbent assay, and clinical characteristics and pregnancy outcomes were compared between groups.

Granulocyte colony-stimulating factor (G-CSF), a pivotal hematopoietic cytokine, has been noted for its potential to bolster embryo implantation and augment endometrial receptivity. The present meta-analysis endeavors to evaluate the therapeutic efficacy of G-CSF in mitigating the incidence of recurrent miscarriages, thereby enriching the clinical evidence supporting its use in treatment protocols. Our exhaustive literature search, concluded on August 25, 2024, spanned across various databases including PubMed, Medline, Cochrane Library, Web of Science, ClinicalTrials, China National Knowledge

We report a rare case of adenosquamous carcinoma of the gallbladder which simultaneously produces granulocyte-colony-stimulating factor (G-CSF) and parathyroid hormone-related protein (PTHrP), confirmed serologically and histologically. A 71-year-old man was examined for a gallbladder tumor with multiple lymph nodes and liver metastases. Histopathological evaluation by endoscopic ultrasound fine-needle aspiration revealed adenosquamous carcinoma of the gallbladder. Laboratory data showed markedly elevated white blood cell (WBC) count of 34,700 µL and corrected serum calcium level of 14.9 mg/dL

Most of embryos fail to produce live offspring during In Vitro Fertilization-Embryo Transfer (IVF-ET) procedure. There is a dearth of research activity addressing this problem despite the significant population of women suffering from repeated implantation failure after transfer of high-quality of embryos. As a clinically accessible option, granulocyte colony stimulating factor (G-CSF) is often used for the treatment to improve the rates of embryo implantation. However, there are currently no evidence-based standardized protocol for the clinical use of G-CSF. G-CSF was administered into one si

Intrauterine adhesion (IUA) caused by endometrial injury is a common cause of female infertility and is challenging to treat. Macrophages play a critical role in tissue repair and cyclical endometrial regeneration. Granulocyte-macrophage colony-stimulating factor (GM-CSF) has significant reparative and anti-fibrotic effects in various tissues. However, there is limited research on the role of GM-CSF in the repair of endometrial injury and the involvement of macrophages in GM-CSF-mediated endometrial repair. In this study, using a mouse model of endometrial scratching injury, we found that GM-C

We present the case of a 62-year-old man with rheumatoid arthritis who developed a leukaemoid reaction and acute respiratory distress syndrome (ARDS) following granulocyte colony-stimulating factor (G-CSF) administration that had been given to treat neutropenia secondary to methotrexate and leflunomide toxicity. Later it was established that he had Pneumocystis jirovecii pneumonia, which was treated to complete resolution with a course of corticosteroids and antibiotics. This case highlights the potential risk of G-CSF administration in an immune compromised individual in the midst of bone mar

Granulocyte colony-stimulating factor (G-CSF) administration is associated with a diverse range of cutaneous sequelae. Serious dermatological side effects of G-CSF include the development of Sweet's syndrome and exacerbations of pre-existing inflammatory disorders such as psoriasis. Here, we describe a report of acute leucocytoclastic vasculitis caused by G-CSF therapy associated with anti-Ro and anti-La antibodies in a patient with multiple myeloma. This case highlights the importance of having a high index of suspicion for acute leucocytoclastic vasculitis in patients with haematological mal

Granulocyte colony-stimulating factor (GCSF) is a growth factor that drives the proliferation and differentiation of granulocytes and monocytes/macrophages. Currently, two copies of GCSF, named GCSFa and GCSFb, have been identified in teleost fish, but data on the functions and signal pathways of these fish GCSFs are still limited. In the present study, a GCSFa homologue (LcGCSFa) was identified from large yellow croaker (Larimichthys crocea). The open reading frame (ORF) of LcGCSFa is 636 bp long and encodes a protein of 211 amino acids (aa), with a 19-aa signal peptide and a typical IL-6 dom

The functionality and durability of implanted biomaterials are often compromised by an exaggerated foreign body reaction (FBR). M1/M2 polarization of macrophages is a critical regulator of scaffold-induced FBR. Macrophage colony-stimulating factor (M-CSF), a hematopoietic growth factor, induces macrophages into an M2-like polarized state, leading to immunoregulation and promoting tissue repair. In the present study, we explored the immunomodulatory effects of surface bound M-CSF on poly-l-lactic acid (PLLA)-induced FBR. M-CSF was immobilized on the surface of PLLA via plasma immersion ion impl
