DESIGN: a framework for generative AI-enabled crop molecular design-driven breeding.

Crop breeding is shifting from selecting naturally occurring variants to actively designing genetic features. Generative artificial intelligence (GenAI) provides a computational foundation for this transition; however, its application in crop systems remains limited. This review bridges this gap by systematically evaluating the translational potential of GenAI for crop molecular design breeding. We examine four major computational architectures and their applications across six key areas relevant to crop breeding: gene regulatory element design, protein engineering, small-molecule generation,

A semiparametric Gaussian mixture model for chest CT-based 3D blood vessel reconstruction.

Computed tomography (CT) has been a powerful diagnostic tool since its emergence in the 1970s. Using CT data, 3D structures of human internal organs and tissues, such as blood vessels, can be reconstructed using professional software. This 3D reconstruction is crucial for surgical operations and can serve as a vivid medical teaching example. However, traditional 3D reconstruction heavily relies on manual operations, which are time-consuming, subjective, and require substantial experience. To address this problem, we develop a novel semiparametric Gaussian mixture model tailored for the 3D reco

How does computed tomography inform our understanding of shoulder kinematics? A structured review.

The objective of this structured review was to review how computed tomography (CT) scanning has been used to measure the kinematics of the shoulder. A literature search was conducted using Evidence-based Medicine Reviews (Embase) and PubMed. In total, 29 articles were included in the data extraction process. Forty percent of the studies evaluated healthy participants' shoulder kinematics. The glenohumeral joint was the most studied, followed by the scapulothoracic, acromioclavicular, and sternoclavicular joints. Three-dimensional computed tomography (3DCT) and 3DCT with biplane fluoroscopy are

Multiscale In Silico Modeling of Cartilage Injuries.

Injurious loading of the joint can be accompanied by articular cartilage damage and trigger inflammation. However, it is not well-known which mechanism controls further cartilage degradation, ultimately leading to post-traumatic osteoarthritis. For personalized prognostics, there should also be a method that can predict tissue alterations following joint and cartilage injury. This chapter gives an overview of experimental and computational methods to characterize and predict cartilage degradation following joint injury. Two mechanisms for cartilage degradation are proposed. In (1) biomechanica

Measurement of acute tolerance to alcohol in human subjects.

Acute tolerance can be defined as a decrease in response to alcohol within a single exposure to the drug, which occurs independently of changes in blood alcohol concentrations (BACs). BACs change over time in most human alcohol administration studies, and computational techniques that account for these changes must be used to measure the rate of acute tolerance development. The most widely used acute tolerance measure in human research is often called the Mellanby effect, and involves the comparison of responses at the same BAC on the ascending and descending limbs of the blood alcohol curve.

Harnessing AI to fuse phenotypic signatures for drug target identification: progress in computational modeling.

Computational models integrating large-scale gene expression profiles provide a powerful approach for predicting multi-target drug interactions (DTIs). Unlike traditional experimental and computational methods that often require detailed structural or target-specific information, gene expression-based models leverage reference transcriptional signatures. This enables functional inference of interactions without explicit structural data, offering a valuable strategy in data-limited scenarios. By incorporating phenotypic information, these models bridge phenotype screening and target prediction,

Computational Intelligence-Based Disease Severity Identification: A Review of Multidisciplinary Domains.

Disease severity identification using computational intelligence-based approaches is gaining popularity nowadays. Artificial intelligence and deep-learning-assisted approaches are proving to be significant in the rapid and accurate diagnosis of several diseases. In addition to disease identification, these approaches have the potential to identify the severity of a disease. The problem of disease severity identification can be considered multi-class classification, where the class labels are the severity levels of the disease. Plenty of computational intelligence-based solutions have been pres

Viewpoint on Crossmodal Neural Plasticity.

Crossmodal neural plasticity, the brain's ability to adapt in response to sensory deprivation, serves as a primary model for understanding the multifaceted interplay between intrinsic neurobiological constraints and environmental experience in brain development. In this viewpoint, Olivier Collignon and Ella Striem-Amit engage in a collaborative dialogue exploring the fundamental principles of brain organization in individuals born with blindness, deafness, or without hands. The discussion evaluates current shifts in the field, including the move toward high-resolution neuroimaging, deep data a

Formaldehyde hydrazone as a methyl reagent for nickel-catalyzed cross-coupling methylation of aryl and heteroaryl electrophiles.

Methyl groups play a vital role in pharmaceuticals, where their installation onto aryl and heteroaryl moieties of drug candidates can often enhance drug-target interactions. This phenomenon, termed the "magic methyl effect", has become an important tool for improving therapeutic potency during the lead optimization stages of drug development. Herein, we report a methodology for the methylation of various phenols (as tosylates) and aryl halides via a hydrazone-mediated Ni-catalyzed cross-coupling reaction, employing formaldehyde hydrazone as the methyl reagent. The reaction delivers moderate to

DTREEv2, a computer-based support system for the risk assessment of genetically modified plants.

Risk assessment of genetically modified organisms (GMOs) remains a contentious area and a major factor influencing the adoption of agricultural biotech. Methodologically, in many countries, risk assessment is conducted by expert committees with little or no recourse to databases and expert systems that can facilitate the risk assessment process. In this paper we describe DTREEv2, a computer-based decision support system for the identification of hazards related to the introduction of GM-crops into the environment. DTREEv2 structures hazard identification and evaluation by means of an Event-Tre

[X-ray diagnosis of chronic traumatic lesions of the diaphragm].

The authors analyze the clinical and x-ray findings in 24 patients with chronic traumatic injuries of the diaphragm. The clinical findings did not hint at a chronic traumatic injury of the diaphragm. The x-ray method was the principal in the diagnosis of the condition. A correct methodologic approach to the examination of a patient helped detect a chronic injury to the diaphragm virtually in all the cases and the type of the injury could be recognized by routine x-ray methods. Indications for the use of invasive methods of examinations were defined: pneumoperitoneum and bronchography. Computer

From Mood Episodes to Digital Signatures: Passive and Active Phenotyping of Bipolar Disorder Over Time.

BACKGROUND: Digital phenotyping has emerged as a promising approach to capture real-time behavioral and physiological data in individuals with bipolar disorder (BD). By integrating passive and active data streams, this approach may enable the identification of dynamic patterns associated with mood instability. However, the conceptual integration of these data into clinically meaningful digital signatures remains insufficiently defined and lacks standardized operational frameworks. METHODS: This narrative review synthesizes current evidence on digital phenotyping in BD and proposes a conceptua

Tailoring transition metal complexes for nonlinear optics applications. 2. A theoretical investigation of the second-order nonlinear optical properties of M(CO)(5)L complexes (M = Cr, W; L = Py, PyCHO, Pyz, PyzBF(3), BPE, BPEBF(3)).

In this work, we report an ab initio investigation of second-order nonlinear optical (NLO) properties and absorption electronic spectra of push-pull transition metal chromophores of the formula [M(CO)(5)L] (M = Cr, W; L = pyridine (Py), 4-formyl-pyridine (PyCHO), pyrazine (Pyz), trans-1,2-bis(4-pyridyl)ethylene (BPE)). Pyz and BPE are considered either with one nitrogen atom free or interacting with the strong acceptor BF(3). All of the molecular properties have been calculated using two different and methodologically independent approaches: the time dependent and coupled perturbed density fun
