Epigallocatechin-3-gallate functions as a physiological regulator by modulating the jasmonic acid pathway.

Flavonoids, a class of plant polyphenols derived from plant secondary metabolism, play important roles in plant development and have beneficial effects on human health. Epigallocatechin-3-gallate (EGCG) is the most abundant polyphenol, and its molecular and biochemical mechanism have been followed with interest. The shared signaling heritage or convergence of organisms has allowed us to extend this research into the model plant, Arabidopsis thaliana. Here, we showed that EGCG could promote jasmonic acid (JA) signaling in A. thaliana. EGCG not only inhibited seed germination but also elevated t

Cotton cytochrome P450 CYP82D regulates systemic cell death by modulating the octadecanoid pathway.

Plant oxylipins are derived from unsaturated fatty acids and play roles in plant growth and development as well as defence. Although recent studies have revealed that fatty acid metabolism is involved in systemic acquired resistance, the precise function of oxylipins in plant defence remains unknown. Here we report a cotton P450 gene SILENCE-INDUCED STEM NECROSIS (SSN), RNAi suppression of which causes a lesion mimic phenotype. SSN is also involved in jasmonate metabolism and the response to wounding. Fatty acid and oxylipin metabolite analysis showed that SSN overexpression causes hyperaccumu

Signaling cross-talk in plant disease resistance.

Hormone signaling crosstalk plays a major role in plant defense against a wide range of both biotic and abiotic stresses. While many reviews on plant-microbe interactions have well described the general trends of signaling pathways in shaping host responses to pathogens, few discussions have considered a synthesis of positive versus negative interactions among such pathways, or variations in the signaling molecules themselves. This review deals with the interaction trends between salicylic, jasmonic, and abscisic acids in the signaling pathways, as well as exceptions to such trends. Here we fo

The epiphytic fungus Pseudozyma aphidis induces jasmonic acid- and salicylic acid/nonexpressor of PR1-independent local and systemic resistance.

Pseudozyma spp. are yeast-like fungi, classified in the Ustilaginales, which are mostly epiphytic or saprophytic and are not pathogenic to plants. Several Pseudozyma species have been reported to exhibit biological activity against powdery mildews. However, previous studies have reported that Pseudozyma aphidis, which can colonize plant surfaces, is not associated with the collapse of powdery mildew colonies. In this report, we describe a novel P. aphidis strain and study its interactions with its plant host and the plant pathogen Botrytis cinerea. This isolate was found to secrete extracellul

Pithy protection: Nicotiana attenuata's jasmonic acid-mediated defenses are required to resist stem-boring weevil larvae.

Folivory is the best studied plant-herbivore interaction, but it is unclear whether the signaling and resistance traits important for the defense of leaves are also important for other plant parts. Larvae of the tobacco stem weevil, Trichobaris mucorea, burrow into stems of Nicotiana attenuata and feed on the pith. Transgenic N. attenuata lines silenced in signaling and foliar defense traits were evaluated in a 2-year field study for resistance against attack by naturally occurring T. mucorea larva. Plants silenced in early jasmonic acid (JA) biosynthesis (antisense [as]-lipoxygenase3 [lox3];

Endogenous peptide defense signals in Arabidopsis differentially amplify signaling for the innate immune response.

AtPep1, a 23-aa peptide encoded by Arabidopsis PROPEP1, a member of a small, six-member gene family, activates expression of the defense gene PDF1.2 (encoding defensin) and its own precursor gene, PROPEP1, through the jasmonate/ethylene signaling pathway, mediated by a cell-surface receptor, PEPR1. Overexpression of two family members, PROPEP1 and PROPEP2, enhances resistance of Arabidopsis plants against the pathogen Pythium irregulare, and PROPEP2 and PROPEP3 are expressed at highly elevated levels in Arabidopsis in response to pathogen infections and to several pathogen-associated molecules

Plant growth-promoting rhizobacteria systemically protect Arabidopsis thaliana against Cucumber mosaic virus by a salicylic acid and NPR1-independent and jasmonic acid-dependent signaling pathway.

Arabidopsis thaliana ecotype Columbia plants (Col-0) treated with plant growth-promoting rhizobacteria (PGPR) Serattia marcescens strain 90-166 and Bacillus pumilus strain SE34 had significantly reduced symptom severity by Cucumber mosaic virus (CMV). In some cases, CMV accumulation was also significantly reduced in systemically infected leaves. The signal transduction pathway(s) associated with induced resistance against CMV by strain 90-166 was determined using mutant strains and transgenic and mutant Arabidopsis lines. NahG plants treated with strains 90-166 and SE34 had reduced symptom sev

Mycorrhiza-induced resistance against Botrytis cinerea in tomato operates through the systemin signalling cascade.

Arbuscular mycorrhizal fungi enhance plant resistance against necrotrophic fungi by priming jasmonic acid-dependent defences. Systemin is a solanaceous peptide hormone that mediates resistance against Botrytis cinerea, and JA signalling is critical for this systemin-mediated defence. Mycorrhizal symbiosis primes prosystemin expression in the tomato (Solanum lycopersicum)-B. cinerea pathosystem. Whether prosystemin or systemin mediates mycorrhiza-induced resistance (MIR) against B. cinerea remains unclear. We hypothesise that systemin mediates JA priming during MIR. We integrated transcriptiona

Levan-derived oligosaccharides (LOS) prime barley against fungal pathogens through asparagine metabolism.

Overuse of chemical pesticides motivates green disease control strategy. Levan-derived oligosaccharides (LOS) are immune elicitors, but its structure information and underlying regulatory mechanisms remain unclear. In this study, LOS produced from sucrose using LevB1SacB fusion enzyme, was purified by CSR-1Na resin, and characterized by HPLC, ATR-FTIR and 1D/2D NMR. Foliar LOS spraying (0.5‰) enhanced barley resistance to Rhizoctonia solani and Pyrenophora graminea, reducing necrotic area by 60.5% and 22.4%, but had little effect on the hemibiotrophic pathogen Bipolaris sorokiniana of spot blo

Lipoxygenase 2 (LOX2) enhances cold tolerance through modulating jasmonate biosynthesis in Nicotiana tabacum.

BACKGROUND: Chilling stress severely compromises tobacco production by reducing leaf biomass accumulation and deteriorating leaf quality. Lipoxygenase (LOX) plays a key role in the oxidative metabolism of linoleic and linolenic acids, which serves as the initial step for jasmonic acid (JA) biosynthesis. However, its specific function in mediating cold-induced JA accumulation and cold tolerance in Nicotiana tabacum remains elusive. RESULTS: Our results found that cold stress significantly induces NtLOX2 expression in N. tabacum. Transgenic tobacco plants overexpressing NtLOX2 (OE#NtLOX2-1 and O

JA-CmMYC2 and SA-CmWRKY40 signaling networks relieve the inhibition of CmTLP15/22 on the cold tolerance of melon seedlings.

Cold stress is a major environmental factor that restricts plant growth and development. Previous studies report that thaumatin-like proteins (TLPs) contribute to plant cold responses, but the underlying molecular mechanism remains unclear. Using melon, a warm-adapted vegetable crop, we performed transcriptome analysis and identified CmTLP15/22 as key cold-responsive genes. Silencing CmTLP15/22 increased CmCBF1/2/4 expression and antioxidant capacity and improved cold tolerance in melon seedlings. Protein-DNA interaction assays identified CmMYC2 and CmWRKY40 as transcription factors that bind

G protein-coupled estrogen receptor (GPER) modulates epithelial-mesenchymal transition-like phenotype in human glioblastoma cells.

Glioblastomas (GBs) are the most frequent and aggressive central nervous system tumors, whose progression is influenced by sex hormones. 17β-estradiol (E2), promotes an epithelial-to-mesenchymal transition (EMT)-like process and enhances proliferative, migratory, and invasive capacities in GB through intracellular estrogen receptors. However, the role of G protein-coupled estrogen receptor (GPER) in GB remains poorly understood. We evaluated the effects of E2, the GPER-agonist G-1, and the GPER-antagonist G-15 on EMT-related processes, migration, and invasion in U251 and U87 GB-derived cells.

Suppression of ZmJAZ12 Expression Modulates Jasmonic Acid and Salicylic Acid Signalling and Enhances Maize Resistance to Southern Corn Rust.

Southern corn rust (SCR), caused by the biotrophic fungus Puccinia polysora, severely threatens maize yield worldwide. In this study, we identified ZmJAZ12, a member of the JASMONATE ZIM-domain (JAZ) protein family, as a negative regulator of SCR resistance in maize. Genome-wide association study (GWAS) and expression analyses revealed that the expression level of ZmJAZ12 was negatively correlated with SCR resistance, and the resistant haplotypes exhibited significantly lower transcript abundance of ZmJAZ12. The ZmJAZ12 overexpression (OE) lines showed enhanced susceptibility to SCR, while ZmJ

Global update on the susceptibilities of influenza viruses to neuraminidase inhibitors and the cap-dependent endonuclease inhibitor baloxavir, 2020-2023.

Antiviral susceptibility of influenza viruses is monitored by the World Health Organization Global Influenza Surveillance and Response System. This study describes a global analysis of the susceptibility of influenza viruses to neuraminidase (NA) inhibitors (NAIs, oseltamivir, zanamivir, peramivir, laninamivir) and the cap-dependent endonuclease inhibitor (CENI, baloxavir) for three periods (May to May for 2020-2021, 2021-2022 and 2022-2023). In particular, global influenza activity declined significantly in 2020-2021 and 2021-2022 when compared to the pre-pandemic period of COVID-19. Combined

Cyclopentanoid monoterpene glycosides from the aerial parts of Paederia foetida L.

Eight previously undescribed cyclopentanoid monoterpene glycosides, paedosides A-H (1-8), and 19 known analogues were isolated from the aerial parts of Paederia foetida L. The structures of these compounds, especially their absolute configurations, were unequivocally established by comprehensive spectroscopic analysis, various quantum chemical calculations, and single-crystal X-ray diffraction analysis. Compound 1 contains a thiocarbonate group, with the aglycone C-1 linked via O-glycosidic bonds to both sugar C-1' and C-6', thereby forming a 7-membered ring. Compounds 2-4 feature a skeleton w

Genome-wide Characterization of Small Secreted Peptides in Nicotiana tabacum and Functional Assessment of NtLTP25 in Plant Immunity.

Small secreted peptides (SSPs), serving as signaling molecules for intercellular communication, play significant regulatory roles in plant growth, development, pathogen immunity, and responses to abiotic stress. Despite several SSPs, such as PIP, PSK, and PSY having been identified to participate in plant immunity, the majority of SSPs remain understudied, necessitating the exploration and identification of SSPs regulating plant immunity from vast genomic resources. Here we systematically characterized 756 putative SSPs across the genome of Nicotiana tabacum. 173 SSPs were further annotated as

Functional characterization of AcWRKY94 in response to Pseudomonas syringae pv. actinidiae in kiwifruit.

WRKY transcription factors are essential for coping with various biotic stresses. Pseudomonas syringae pv. actinidiae (Psa)-induced kiwifruit canker is a major problem restricting kiwifruit yield. Nevertheless, it's unclear how the kiwifruit WRKY genes respond to Psa. Through genome-wide identification, 112 WRKY members were found in 'Hongyang' genome in this work. Promoter analysis revealed that there were many cis-acting elements associated with stress responses in the AcWRKY gene's promoter region. According to transcriptomic analysis, 90 of the AcWRKY genes were differently expressed follo
