Chandlerella quiscali is a filarial nematode parasite of the common grackle (Quiscalus quiscula), a widespread bird species found throughout most of North America. Worms collected from wild-caught birds were morphologically identified as C. quiscali and tested for the presence of Wolbachia, an alphaproteobacterial endosymbiont required for reproduction and maturation by many filarial species. Although various methods, including polymerase chain reaction, in situ hybridization, and immunohistology, were used, we were unable to detect evidence of colonization with Wolbachia. Due to the widesprea

Fasciola gigantica and Fasciola hepatica are causative pathogens of fascioliasis, with the widest latitudinal, longitudinal, and altitudinal distribution; however, among parasites, they have the largest sequenced genomes, hindering genomic research. In the present study, we used various sequencing and assembly technologies to generate a new high-quality Fasciola gigantica reference genome. We improved the integration of gene structure prediction, and identified two independent transposable element expansion events contributing to (1) the speciation between Fasciola and Fasciolopsis during the

Prophylaxis with macrocyclic lactone (ML) endectocides is the primary strategy for heartworm control. Recent evidence has confirmed that ML-resistant Dirofilaria immitis isolates have evolved. Comparison of genomes of ML-resistant isolates show they are genetically distinct from wild-type populations. Previously, we identified single nucleotide polymorphisms (SNPs) that are correlated with phenotypic ML resistance. Since reliable in vitro assays are not available to detect ML resistance in L3 or microfilarial stages, the failure to reduce microfilaraemia in infected dogs treated with an ML has

Trichinella spiralis, the causative agent of trichinellosis, is able to infect a wide range of carnivores and omnivores including human beings. In the past 30 years, a mass of vaccination efforts has been performed to control T. spiralis infection with the purpose of reduction in worm fecundity or decrease in muscle larval and adult burdens. Here, we summarize the development of veterinary vaccines against T. spiralis infection. During recent years, increasing numbers of new vaccine candidates have been developed on the protective immunity against T. spiralis infection in murine model. The vac

Human cases of gastric anisakiasis caused by the zoonotic parasite Anisakis pegreffii are increasing in Italy. The disease is caused by ingestion of larval nematodes in lightly cooked or raw seafood. Because symptoms are vague and serodiagnosis is difficult, the disease is often misdiagnosed and cases are understimated.

Entomologic and serologic surveys were performed in four sentinel communities in the Oaxaca focus in southern Mexico to assess the level of transmission and exposure incidence to Onchocerca volvulus. All communities have been receiving ivermectin mass treatment twice per year since 1997. In one community, parasite DNA was detected by polymerase chain reaction-enzyme-linked immunosorbent assay in 2004 in one pool of 50 vector heads of 170 such pools (8,500 flies) examined, which indicated an estimated transmission potential of 6.7 third-stage larvae/person/year. No evidence for transmission was

The temporal expression of defensin, cecropin and transferrin was assessed in Aedes aegypti naturally refractory to Wuchereria bancrofti upon infection with this worm, in parallel to analysis of filarial development in the insect. Compared to controls, transcription of defensin and cecropin was higher in infected mosquitoes as soon as 2h post infection and peaked before 48h. Transferrin transcription was higher in infected mosquitoes at 24h, and at 48h was almost leveled to controls. At 72h and 7 days post infection, levels of all transcripts in infected insects decreased gradually and were si

Schistosoma mansoni, an intravascular parasite, lives in a hostile environment in close contact with host humoral and cellular cytotoxic factors. To establish itself in the host, the parasite has evolved a number of immune evasion mechanisms, such as antioxidant enzymes. Our laboratory has demonstrated that the expression of antioxidant enzymes is developmentally regulated, with the highest levels present in the adult worm, the stage least susceptible to immune elimination, and the lowest levels in the larval stages, the most susceptible to immune elimination. Vaccination of mice with naked DN

We report on the rediscovery of a presumably lost type specimen of Echinococcus multilocularis (Leuckart, 1863). The study establishes the mitochondrial genome of E. multilocularis from a historical syntype specimen and explores modern genomic techniques to clarify its identity in European populations. Despite initial failure in nested PCR, high-throughput next-generation sequencing (NGS) successfully assembled the mitochondrial genome from post-capture reads, revealing a 13,738 bp sequence. This genome contained 12 protein-coding genes, 2 rRNA genes, and 22 tRNA genes. Phylogenetic analysis p

Echinococcosis is a severe parasitic zoonosis worldwide distribution, caused by the genus Echinococcus. One of its recently identified species, Echinococcus shiquicus, is believed to be endemic to the Qinghai-Tibet plateau in China. Research has confirmed that Tibetan foxes are the definitive hosts for E. shiquicus, while plateau pikas (Ochotona curzoniae) serve as intermediate hosts. In this study, cyst containing protoscolices was first observed in the lung of plateau zokor (Eospalax baileyi) in the Tibetan Autonomous Prefecture of Gannan, Gansu Province, China. This observation was confirme

We described Creptotrema cambeve n. sp. from the intestine of the Brazilian catfish Cambeva davisi (Haseman, 1911) (= Trichomycterus davisi) based on integrative analyses using morphological and molecular approaches. Creptotrema cambeve n. sp. closely resembles morphologically Creptotrema paraense Vicente, Santos & Souza, 1978, which was described from a siluriform fish in Northern Brazil; however, the two species differ mainly in the oral and ventral sucker sizes and the distribution of the vitellaria. The phylogenetic analyses of the 28S rDNA placed the sequences of the new species in a mono

Echinochasmids are a group of globally distributed digeneans, and the adults are found in the intestines of birds, mammals and reptiles. In the Neotropical region of Mexico, adult specimens were obtained from seven fish-eating bird species in six localities, whereas specimens of Stephanoprora aylacostoma were obtained experimentally in Argentina. Morphologically, the new specimens from the Neotropical region of Mexico were identified as Stephanoprora uruguayense and an undescribed species of Echinochasmus. OBJECTIVE: Cystic echinococcosis (CE) is a zoonotic infection that affects humans, livestock and wild animals through the larval form of Echinococcus granulosus sensu lato (s.l.). Molecular and taxonomic studies carried out in the recent years accept that Echinococcus granulosus s.l., a complex of 5 cryptic species, causes CE. In this study, we performed morphological and molecular characterisation of cyst isolates obtained from a wild boar and mule naturally infected with hydatid cyst.
METHODS: After gDNA isolation, the mitochondrial cytochrome oxidase subunit 1 (mt-CO1) gene region was ampl

BACKGROUND: Cystic echinococcosis (CE) in humans and livestock is caused by Echinococcus granulosus (sensu lato). In China where CE is endemic, a number of studies have shown that Echinococcus granulosus (sensu stricto) is majorly responsible for CE. However, E. canadensis (G6) which is the second leading cause of CE is now being detected in most parts of the country. In this study, the species diversity and genetic variation of Echinococcus granulosus (s.l.) in four counties in Tibet Autonomous Region of China were investigated.
METHODS: Infection with Echinococcus granulosus (s.s.) in yaks

There are limited reports of infectious agents affecting Australian cowtail stingrays. In the present study, a new species of ascaridoid nematode belonging to the genus Mawsonascaris is described. The most distinct characteristic features were observed in females (the presence of a polar spine in the eggs and a flap-like projection in the vulval area). An identification key for Mawsonascaris spp. is provided. Additionally, internal transcribed spacers (ITS) sequences were obtained for the new species. Alignment of the ITS sequence of the specimens in the present study with those deposited in G

The present study was performed to analyze molecularly the phylogenetic positions of human-infecting Trichostrongylus species in Mazandaran Province, Iran, which is an endemic area for trichostrongyliasis. DNA from 7 Trichostrongylus infected stool samples were extracted by using in-house (IH) method. PCR amplification of ITS2-rDNA region was performed, and products were sequenced. Phylogenetic analysis of the nucleotide sequence data was performed using MEGA 5.0 software. Six out of 7 isolates had high similarity with Trichostrongylus colubriformis, while the other one showed high homology wi

Echinococcus granulosus (sensu lato) is now recognized as an assemblage of cryptic species, which differ considerably in morphology, development, host specificity (including infectivity/pathogenicity for humans) and other aspects. One of these species, E. granulosus sensu stricto (s.s.), is now clearly identified as the principal agent causing cystic echinococcosis in humans. Previous studies of a small section of the cox1 and nadh1 genes identified two variants of E. granulosus s.s. to be present in Australia; however, no further work has been carried out to characterize the microdiversity of

A total of 1523 individuals of 34 species of chaetodontids from the Great Barrier Reef were examined for faustulid trematodes. Specimens resembling Paradiscogaster glebulae Bray, Cribb & Barker, 1994 were found in nine chaetodontid species at three localities. These specimens are shown, on the basis of combined morphological and molecular analyses, to comprise a complex of morphologically similar and partly cryptic species. The complex may comprise as many as six distinct species of which three are resolved here. The true P. glebulae is identified in Chaetodon ornatissimus Cuvier, 1831, Chaeto

In this study, we used sequences of mitochondrial and nuclear markers to test the hypothesis that Tabascotrema verai Lamothe-Argumedo and Pineda-López, 1990, a parasite of the cichlid Petenia splendida Günther, 1862 in Middle America, represents a single species, following a molecular prospecting approach. One-hundred and five individuals of T. verai were collected from the intestine of 43 specimens of P. splendida in eight localities of Mexico, Belize and Guatemala. A portion (n=53) of the collected specimens was used for molecular studies, whereas the remaining individuals were processed for
