Effects of a single-dose pretreatment with captopril on the immediate response to nasal challenge with allergen.

We performed a double-blind, placebo-controlled, crossover study using 12 subjects to determine the effects of a single 50-mg dose of captopril on the response to nasal challenge with increasing doses of allergen. Levels of kinins, histamine and N-alpha-p-tosyl-L-arginine methyl ester (TAME)-esterase activity were measured in nasal lavages, and symptom scores and the number of sneezes were recorded. Captopril had no significant effects on histamine, TAME-esterase, sneezing or symptom scores. Peak postchallenge kinin levels, however, were significantly elevated (p less than 0.05) compared to pl

Pulmonary angiotensin-converting enzyme activity in the oxygen-toxic sheep.

Despite the potential utility of endothelial metabolic substrates for the early clinical detection of acute lung injury, the relationship between lung capillary injury and pulmonary endothelial metabolic function remains incompletely understood. Previous studies have shown that lung capillaries are damaged by oxygen toxicity in the sheep; however, metabolic functions of the pulmonary endothelium have not been examined in this otherwise well-characterized animal model of lung injury. Therefore, we studied the activity of pulmonary endothelial angiotensin-converting enzyme (ACE) in five unanesth

The renin angiotensin system--its physiology and role in disease states.

The renin-angiotensin system is one of a number of interlinked mechanisms regulating vascular resistance and blood volume. Under certain conditions it may become a predominant factor in maintaining vascular tone. Knowledge about these conditions (sodium depletion, mineralocorticoid deficiency, renovascular hypertension and iatrogenic hyperreninaemic states) is important for the safe and effective use of drugs which inhibit the renin-angiotensin system. Measurements of plasma renin activity are useful in the diagnostic assessment of hypertensive patients with hypokalaemia or evidence of renal a

Development and design of specific inhibitors of angiotensin-converting enzyme.

Captopril is a remarkably effective new antihypertensive drug designed and developed as a potent and specific inhibitor of angiotensin-converting enzyme, a zinc metallopeptidase that participates in the synthesis of a hypertensive peptide, angiotensin II, and in the degradation of a hypotensive peptide, bradykinin. Earlier studies with a snake venom peptide (teprotride or SQ 20881) that could be administered only by injection demonstrated that specific inhibitors of angiotensin-converting enzyme could be highly effective as antihypertensive drugs, and helped to clarify the specificity and mech

[The correlation between serum angiotensin I-converting enzyme activity and the renin-angiotensin-aldosterone system in hypertensive patients (author's transl)].

In order to investigate the role and origin of serum angiotensin I-converting enzyme activity (ACEA) in hypertension, the correlation between serum ACEA and the renin-angiotensin-aldosterone system was evaluated in hypertensive patients of whom 36 had essential hypertension, five had hypertension associated with chronic renal failure, three had renovascular hypertension, and one had primary aldosteronism. Serum ACEA was 17.0 +/- 3.3 U (mean +/- sd) in the normotensive control, 20.1 +/- 7.1 U in patients with essential hypertension, 11.6 +/- 3.4 U in patients with chronic renal failure, 31.7 +/

Neuro-renin-angiotensin-aldosterone system axis in alzheimer's disease: from molecular dysregulation to therapeutic redirection.

Alzheimer's disease (AD) is characterized by progressive neurodegeneration marked by tau hyperphosphorylation, amyloid-beta (Aβ) buildup, neuroinflammation, and blood-brain barrier (BBB) dysfunction. Although much attention is paid to understanding amyloid and tau pathologies, there are still no disease-modifying solutions. Recent evidence indicates that the brain-specific Renin-Angiotensin-Aldosterone System (RAAS), conventionally involved in the regulation of cardiovascular diseases, could be central in controlling the key neuropathological alterations in AD. This review explains the binary

Blood ACE Activity Is Significantly Lowered in Psoriasis Compared to Atopic Dermatitis: A Small Pilot Study in Young Males.

PURPOSE: Emerging evidence suggests the involvement of the renin-angiotensin system (RAS) in the pathogenesis and progression of autoimmune dermatological diseases. In a small exploratory study, we investigated angiotensin-converting enzyme (ACE) activity in blood obtained from male probands with psoriasis vulgaris (n = 4) and atopic dermatitis (AD, n = 5). METHODS: The degradation capacity of dabsylated synthetic bradykinin (DBK) with and without inhibition was determined using a thin-layer chromatography (TLC)-based neuropeptide reporter assay. RESULTS: We observed a significantly reduced

Impact of Vitamin D Replacement on the Vitamin D-Leptin-Prolidase Axis in Secondary Hyperparathyroidism.

Severe vitamin D deficiency commonly results in secondary hyperparathyroidism and increased bone turnover. Despite the well-established inverse relationship between vitamin D and parathyroid hormone, the impact of vitamin D repletion on collagen matrix remodeling and adipose-bone signaling remains unclear, and the combined response of prolidase and leptin has not been previously examined. This prospective observational study included 36 female patients with severe vitamin D deficiency and secondary hyperparathyroidism who received oral vitamin D supplementation (50,000 IU/wk) for 8 weeks. Investigation of the toxic mechanism of aspartame on female infertility using network toxicology and molecular docking approaches.

Aspartame is a nonnutritive sweetener derived from phenylalanine and is widely used in food and beverages globally. In recent years, its safety, particularly the potential carcinogenic risks, has garnered significant attention; however, there has been relatively less focus on its potential infertility risks. This study employed network toxicology methods to construct an interaction network of aspartame and infertility-related targets and identify key targets and pathways. Subsequently, molecular docking technology was employed to further investigate the binding affinity and mechanism of action

Exploiting the angiotensin-converting enzyme pathway to augment endogenous opioid signaling.

Angiotensin Converting Enzyme (ACE) impacts hemodynamics by regulating the conversion of angiotensin I to the vasoconstricting angiotensin II. We recently identified a non-canonical central role of ACE in the degradation of enkephalin heptapeptide, Met-enkephalin-Arg-Phe (MERF). Enkephalins are short-lived, endogenous opioid peptides that mediate the body's intrinsic analgesic response. Here we identify chemically diverse ACE inhibitors using an optimized high throughput screening assay to boost endogenous opioid signaling. Our primary hits (thiorphan, D609, and raloxifene) were selected for d

Perilla frutescens Seeds as a Source of Antihypertensive Peptides: Discovery of Novel Tetrapeptides with ACE Inhibitory Activity.

Hypertension, a major risk factor for cardiovascular diseases, remains a leading global health concern and has prompted growing interest in angiotensin-converting enzyme (ACE) inhibitory peptides from plant seeds as promising nutraceutical ingredients. To our knowledge, this is the first study to identify antihypertensive peptides from Perilla frutescens seed protein, with thermolysin hydrolysate demonstrated the favorable ACE-inhibitory (ACEI) activity (IC50 = 141 µg/mL). Two parallel bioassay-guided fractionations (SCX and RP-HPLC) were used to screen the fractions with the best ACEI activit

Isolated renal sarcoidosis and concurrent secondary membranous nephropathy: a case report.

BACKGROUND: Sarcoidosis is a chronic inflammatory disease characterized by abnormal T-cell responses to unknown antigens. It is a multisystem disorder, affecting the lungs and intrathoracic lymph nodes in over 90% of cases. Isolated renal sarcoidosis, a rare presentation, is confined to the kidneys without systemic involvement. CASE PRESENTATION: A 46-year-old African American female was evaluated for persistently abnormal serum creatinine and pyuria. The only abnormality on her noninvasive workup was an elevated angiotensin converting enzyme level for which she was evaluated previously and s

Effects of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and coronavirus (COVID-19) vaccination on male fertility.

Coronavirus disease 2019, which is caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), remains an ongoing global public health challenge. This disease causes damage not only to the respiratory system, affecting the normal physiological function of the lungs, but also to other vital organs, such as the heart and testicles. Existing studies have shown that co-expression of angiotensin-converting enzyme 2 and transmembrane serine protease 2 is the main mechanism by which SARS-CoV-2 invades host cells. Angiotensin-converting enzyme 2-expressing cells are widespread in the corpu

Biodirected Screening and Preparation of Larimichthys crocea Angiotensin-I-Converting Enzyme-Inhibitory Peptides by a Combined In Vitro and In Silico Approach.

Food-derived angiotensin-I-converting enzyme (ACE)-inhibitory peptides have gained attention for their potent and safe treatment of hypertensive disorders. However, there are some limitations of conventional methods for preparing ACE-inhibitory peptides. In this study, in silico hydrolysis, the quantitative structure-activity relationship (QSAR) model, LC-MS/MS, inhibition kinetics, and molecular docking were used to investigate the stability, hydrolyzability, in vitro activity, and inhibition mechanism of bioactive peptides during the actual hydrolysis process. Six novel ACE-inhibitory peptid

Role of angiotensin converting enzyme in pathogenesis associated with immunity in cardiovascular diseases.

Angiotensin converting enzyme (ACE) is not only a critical component in the renin-angiotensin system (RAS), but also suggested as an important mediator for immune response and activity, such as immune cell mobilization, metabolism, biogenesis of immunoregulatory molecules, etc. The chronic duration of cardiovascular diseases (CVD) has been increasingly considered to be triggered by uncontrolled pathologic immune reactions from myeloid cells and lymphocytes. Considering the potential anti-inflammatory effect of the traditional antihypertensive ACE inhibitor (ACEi), we attempt to elucidate wheth

Spatial Distribution of SARS-CoV-2 Receptors and Proteases in Testicular Cells.

Coronavirus disease (COVID-19) is caused by severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2). SARS-CoV-2 RNA has been found in the human testis on occasion, but subgenomic SARS-CoV-2 and infectious SARS-CoV-2 virions have not been found. There is no direct evidence of SARS-CoV-2 infection of testicular cells. To better understand this, it is necessary to determine whether SARS-CoV-2 receptors and proteases are present in testicular cells. To overcome this limitation, we focused on elucidating with immunohistochemistry the spatial distribution of the SARS-CoV-2 receptors angiotensin
