Patient tolerance to carbonic anhydrase inhibitors.

We evaluated carbonic anhydrase inhibitors in a crossover study with a placebo and random allocation of treatment administration. Drugs evaluated included acetazolamide tablets and Sequels, dichlorphenamide, ethoxzolamide, methazolamide, and an ascorbic acid placebo. The best tolerated drug was acetazolamide Sequels. Methazolamide was next. The least tolerated drugs were ethoxzolamide, acetazolamide tablets, and dichlorphenamide.

[Medicine hypersensitivity--case report].

In this paper a case of good tolerance to benzathine benzylpenicillin, but benzylpenicillin potassium allergy in subject with history of multidrug hypersensitivity was described. The same therapeutic agent is contained in both commercial drugs and dosage formulation can cause the unpredictable adverse drug reactions (ADRs). This thesis was confirmed by positive intradermal test with the same, but negative with other commercial product of benzylpenicillin potassium. Later chromatographic analysis shows drug contaminations. Thus an exclusion of allergy to drug impurities (and additives) is neces

T394A Mutation at the μ Opioid Receptor Blocks Opioid Tolerance and Increases Vulnerability to Heroin Self-Administration in Mice.

UNLABELLED: The etiology and pathophysiology underlying opioid tolerance and dependence are still unknown. Because mu opioid receptor (MOR) plays an essential role in opioid action, many vulnerability-related studies have focused on single nucleotide polymorphisms of MOR, particularly on A118G. In this study, we found that a single-point mutation at the MOR T394 phosphorylation site could be another important susceptive factor in the development of opioid tolerance and dependence in mice. T394A mutation, in which a threonine at 394 was replaced by an alanine, did not alter agonist binding to M

Benzalkonium chloride and heavy-metal tolerance in Listeria monocytogenes from retail foods.

Phenotypic and genotypic tolerance in 71 Listeria monocytogenes isolates from different varieties of foods to benzalkonium chloride (BC) and cadmium were investigated by susceptibility test and molecular methods. To investigate the role of efflux pumps in BC tolerance, reserpine, an efflux pump inhibitor, was added to the BC tolerant strains. Tolerance to BC and cadmium were 26.8% (19/71) and 49.3% (35/71) respectively. Strains with BC tolerance were significantly more frequent among those of serotype 4b (100%, 6/6) than among those of serotype 1/2a (or 3a) (13.5%, 5/37), which represent the p

Dominant roles of BRCA1 in cellular tolerance to a chain-terminating nucleoside analog, alovudine.

Alovudine is a chain-terminating nucleoside analog (CTNA) that is frequently used as an antiviral and anticancer agent. Generally, CTNAs inhibit DNA replication after their incorporation into nascent DNA during DNA synthesis by suppressing subsequent polymerization, which restricts the proliferation of viruses and cancer cells. Alovudine is a thymidine analog used as an antiviral drug. However, the mechanisms underlying the removal of alovudine and DNA damage tolerance pathways involved in cellular resistance to alovudine remain unclear. Here, we explored the DNA damage tolerance pathways resp

The Ptk2-Pma1 pathway enhances tolerance to terbinafine in Trichophyton rubrum.

The increasing prevalence of dermatophyte resistance to terbinafine, a key drug in the treatment of dermatophytosis, represents a significant obstacle to treatment. Trichophyton rubrum is the most commonly isolated fungus in dermatophytosis. In T. rubrum, we identified TERG_07844, a gene encoding a previously uncharacterized putative protein kinase, as an ortholog of budding yeast Saccharomyces cerevisiae polyamine transport kinase 2 (Ptk2), and found that T. rubrum Ptk2 (TrPtk2) is involved in terbinafine tolerance. In both T. rubrum and S. cerevisiae, Ptk2 knockout strains were more sensitiv

Discovery of a Caffeic Acid Thioamide with Potent Activity against Pathogenic Fungi and Fluconazole Tolerance-Eliminating Effects in Candida albicans.

BACKGROUND: The increasing incidence of invasive fungal infections, coupled with the rising threat of antifungal resistance and tolerance, has created an urgent demand for novel antifungal agents with distinct mechanisms of action. METHODS: In this study, a series of caffeic acid amide derivatives (59 compounds) from our in‑house library was systematically screened for in vitro antifungal activity against clinically relevant pathogenic fungi. Structure-activity relationship (SAR) analysis was performed to identify key pharmacophoric features. The lead compound F4 was further characterized via

Stochastic Pharmacokinetic-Pharmacodynamic Analysis of the Effect of Transdermal Buprenorphine on Electroencephalogram and Analgesia.

BACKGROUND: The analgesic effect of opioids is often based on subjective one dimensional measurements. Electroencephalography (EEG) offers a possibility to objectively quantify the brain's activity before and after the administration of opioids. The aim of this study was to investigate the pharmacokinetic-pharmacodynamic (PKPD) properties of the buprenorphine transdermal patch on resting EEG and pain tolerance. METHOD: Twenty-two healthy male subjects (mean age 23.1 ± 3.8 years) were studied. They received a 144-hour buprenorphine (20 μg/h) or placebo transdermal patch in this experimental, r

Discovery of novel dual μOR/σ1R ligands as potent analgesics with reduced tolerance liability and improved safety profiles.

Opioid tolerance remains a widespread challenge in pain management, as prolonged μOR activation progressively diminishes analgesic efficacy, necessitating dose escalation and thereby increasing the risk of adverse effects. σ1R antagonism has been shown to potentiate opioid antinociception and mitigate the development of opioid tolerance. However, whether these complementary pharmacological actions can be effectively integrated into a single dual-target ligand remains underexplored. Our previous work yielded 4x, a promising dual μOR agonist/σ1R antagonist with potent in vitro and in vivo activi

D-penicillamine-induced autoimmunity in the Brown Norway rat: role for both T and non-T splenocytes in adoptive transfer of tolerance.

The D-penicillamine autoimmune syndrome observed in Brown Norway (BN) rats is similar to an idiosyncratic reaction seen in some patients. We have previously shown that 2 weeks of low dose (5 mg/day) D-penicillamine pretreatment completely prevented all clinical signs of autoimmunity normally observed in 60-80% of rats treated with high dose (20 mg/day) D-penicillamine. We demonstrated that this tolerance is immune-mediated; tolerance to D-penicillamine is long lasting, we can adoptively transfer splenocytes from tolerant rats into slightly irradiated naive syngeneic recipients, and rats expose

Impaired Glucose Tolerance in Healthy Men Treated with St. John's Wort.

The purpose of this study was to examine whether the over-the-counter herbal medicinal plant St. John's wort affects glucose tolerance in healthy men. To do this, we included 10 healthy men who were examined by a 2-hr oral glucose tolerance test on three occasions: A: baseline; B: after 21 days of treatment with St. John's wort; and C: at least 6 weeks after the last capsule of St. John's wort was ingested. Plasma glucose, serum insulin and C-peptide levels were measured during an oral glucose tolerance test and used for estimation of area under the concentration-time curve (AUC) as well as in

mRNA therapeutics for restoring immune tolerance in autoimmune diseases.

Autoimmune diseases arise from breakdown of immune tolerance and are commonly treated with broad immunosuppression that limits efficacy and increases toxicity. For drug discovery, mRNA therapeutics offer a programmable, modular platform to develop antigen-specific and mechanism-driven autoimmune therapies. mRNA-based approaches enable transient, controllable expression of self-antigens, immunoregulatory proteins and transcription factors to induce tolerance, reshape local immune microenvironments and enhance regulatory immune pathways. Preclinical studies across models of multiple sclerosis, t

Ceftazidime, as a single antibiotic, in the treatment of multi-resistant Gram-negative infections in intensive care.

In this study we have evaluated the efficacy and tolerance of ceftazidime used as the only antibiotic in the treatment of 19 critically ill patients, with serious infections caused by multiply resistant Gram-negative bacilli in an intensive care unit. The bacteria isolated showed a high susceptibility to ceftazidime and a moderate resistance to seven other antibiotics. Clinical improvement occurred in 42.1% of the patients treated. Tolerance, both local and general, to the drug was considered good. Ceftazidime appeared to be capable of replacing beta-lactam and aminoglycoside combinations in t

Lysophosphatidylserine Nanoparticles Induce Oral Tolerance Independent of TIM-4 and Canonical PS Processing.

Protein replacement therapy is highly effective for treating many protein-deficiency disorders, but its long-term clinical benefit is often limited by anti-drug antibody responses that neutralize therapeutic proteins, compromising safety and efficacy. To address these unwanted immune responses, a lipid nanoparticle-based immunotherapy platform has been developed to induce antigen-specific tolerance. Mice treated with antigen associated with either double-chain phosphatidylserine (PS) or lysophosphatidylserine (LysoPS), a single-acyl-chain phospholipid, showed reduced antibody titers following

The role of nucleus accumbens dopamine signaling in the development of morphine tolerance in neuropathic pain.

BACKGROUND: There is converging evidence that indicates that the nucleus accumbens (NAc) plays a substantial role in pain modulation and analgesic drug responses. Here, the role of the NAc dopaminergic signaling in the development of morphine tolerance in a rat neuropathic pain model was explored. METHODS: Morphine tolerance was induced by twice daily administration of intrathecal morphine in spinal nerve-ligated animals. The extracellular dopamine level in the NAc was measured by microdialysis study and the effects of dopaminergic receptor agonists microinjected into the NAc on morphine anal

Tracking donor-reactive T cells: Evidence for clonal deletion in tolerant kidney transplant patients.

T cell responses to allogeneic major histocompatibility complex antigens present a formidable barrier to organ transplantation, necessitating long-term immunosuppression to minimize rejection. Chronic rejection and drug-induced morbidities are major limitations that could be overcome by allograft tolerance induction. Tolerance was first intentionally induced in humans via combined kidney and bone marrow transplantation (CKBMT), but the mechanisms of tolerance in these patients are incompletely understood. We now establish an assay to identify donor-reactive T cells and test the role of deletio

[Informative value of rerespiration as a method for evaluating functional reserves of the cardiorespiratory system in modeling of some flight factors].

This article presents a study of the informative value of the recurrent breathing (rerespiration) method developed by the authors, for evaluating the functional status of the human body during recovery after exposure to noise and vibration, to piloting accelerations and head-down hypokinesis. It is shown that a 1.5-h exposure to noise and vibration increases tolerance to rerespiration, suggesting a stimulatory effect. A 5-h exposure in half of cases either does not change tolerance to the test or lowers it. Piloting head-to-seat accelerations of 7 to 9 G are associated with a lowering in toler

Regulatory T Cells for Control of Autoimmunity.

Regulatory T (Treg) cells, which specifically express the master transcription factor FoxP3, are indispensable for the maintenance of immunological self-tolerance and homeostasis. Their functional or numerical anomalies can be causative of autoimmune and other inflammatory diseases. Recent advances in the research of the cellular and molecular basis of how Treg cells develop, exert suppression, and maintain their function have enabled devising various ways for controlling physiological and pathological immune responses by targeting Treg cells. It is now envisaged that Treg cells as a "living d
