The clinical impact of glucose-6-phosphate dehydrogenase deficiency in patients with sickle cell disease.

PURPOSE OF REVIEW: Glucose-6-phosphate dehydrogenase (G6PD) deficiency and sickle cell disease (SCD) cause hemolysis, often occurring in individuals of African descent. These disorders co-occur frequently, and possibly interact, altering clinical outcomes in SCD. However, epidemiological investigations of SCD with G6PD deficiency have produced variable results. This contribution reviews the available data about the interaction of G6PD deficiency and SCD. RECENT FINDINGS: Overall, G6PD deficiency contributes few, if any, effects to laboratory values and clinical outcomes in SCD patients, but m

Melanoma: epidemiology, risk factors, pathogenesis, diagnosis and classification.

This article reviews epidemiology, risk factors, pathogenesis and diagnosis of melanoma. Data on melanoma from the majority of countries show a rapid increase of the incidence of this cancer, with a slowing of the rate of incidence in the period 1990-2000. Males are approximately 1.5-times more likely to develop melanoma than females, while according to other studies, the different prevalence in both sexes must be analyzed in relation with age: the incidence rate of melanoma is grater in women than men until they reach the age of 40 years, however, by 75 years of age, the incidence is almost 3

"Toward a clearer definition of confounding" revisited with directed acyclic graphs.

In a 1993 paper (Am J Epidemiol. 1993;137(1):1-8), Weinberg considered whether a variable that is associated with the outcome and is affected by exposure but is not an intermediate variable between exposure and outcome should be considered a confounder in etiologic studies. As an example, she examined the common practice of adjusting for history of spontaneous abortion when estimating the effect of an exposure on the risk of spontaneous abortion. She showed algebraically that such an adjustment could substantially bias the results even though history of spontaneous abortion would meet some def

Dietary folate deficiency and elevated homocysteine levels endanger dopaminergic neurons in models of Parkinson's disease.

Although the cause of Parkinson's disease (PD) is unknown, data suggest roles for environmental factors that may sensitize dopaminergic neurons to age-related dysfunction and death. Based upon epidemiological data suggesting roles for dietary factors in PD and other age-related neurodegenerative disorders, we tested the hypothesis that dietary folate can modify vulnerability of dopaminergic neurons to dysfunction and death in a mouse model of PD. We report that dietary folate deficiency sensitizes mice to MPTP-induced PD-like pathology and motor dysfunction. Mice on a folate-deficient diet exh

Classification of grossly detectable abnormalities and conditions seen at postmortem in Canadian poultry abattoirs according to a hazard identification decision tree.

This study was designed to review all grossly detectable abnormalities and conditions (GDACs) encountered in poultry in Canadian abattoirs to determine which have potential to cause adverse health effects for the consumer. Review of the literature and consultation with scientists in the field of microbiology, epidemiology, poultry pathology, chemistry, and meat inspection served to generate an inventory of GDACs, and a decision tree containing algorithms was developed to identify GDACs potentially representing a health hazard to consumers. Through the use of the decision tree, GDACs were class

Risk factors for low back pain and sciatica.

So far, eight prospective studies and 50 cross-sectional or retrospective studies have focused on risk factors for low back syndromes. Half of these have been published during the 1980s. Hard physical work and, in particular, frequent lifting and postural stress are likely to result in disc degeneration, low back pain and sciatica. Physical strain may also have prophylactic effects, as physical leisure activity and muscular strength are negatively associated with the risk of low back pain. Much evidence points to driving motor vehicles being causally associated with low back pain and sciatica.

Time trends in exposure of cattle to bovine spongiform encephalopathy and cohort effect in France and Italy: value of the classical Age-Period-Cohort approach.

BACKGROUND: The Age-Period-Cohort (APC) analysis is routinely used for time trend analysis of cancer incidence or mortality rates, but in veterinary epidemiology, there are still only a few examples of this application. APC models were recently used to model the French epidemic assuming that the time trend for BSE was mainly due to a cohort effect in relation to the control measures that may have modified the BSE exposure of cohorts over time. We used a categorical APC analysis which did not require any functional form for the effect of the variables, and examined second differences to estimat

Endocrine-disrupting compounds and mammary gland development: early exposure and later life consequences.

Breast cancer is the most common non-skin cancer among women in this country. Breast cancer risk is significantly influenced by genetics, but over 70% of the women that are diagnosed have noninherited or sporadic cancer. The risk of breast cancer is thought to be modified by lifestyle and environment. Exposures to certain chemicals and hormone-mimicking or endocrine-disrupting compounds (EDCs) are suspected of contributing to increased breast cancer incidence as well as precocious puberty in the United States. Studies of EDC effects in rodents indicate that multiple toxicants can alter mammary

A review of probability of causation and its use in a compensation scheme for nuclear industry workers in the United Kingdom.

The assumption that any additional exposure to ionizing radiation leads to an increase in the risk of stochastic health effects implies that some cases of these effects will be caused by exposure incurred occupationally. The main health effect expected to arise in the exposed individuals is cancer. Such radiation-induced cancers cannot be distinguished from the far larger number of background cancers, and, therefore, causation must be assessed statistically. The probability of causation methodology has been developed to ascertain the likelihood that a particular cancer may be attributed to a p

An Appraisal of the Potential Downstream Benefits of RSV Vaccination in Older Adults.

Respiratory syncytial virus (RSV) has long been underrecognized as a significant pathogen in older adults, despite accumulating evidence of its substantial morbidity and mortality burden in this population. While RSV vaccination is now well established as an effective intervention for preventing lower respiratory tract disease, this narrative review examines whether its public health value may extend beyond these primary clinical endpoints. A targeted search of Scopus and Web of Science from database inception to May 2026 was conducted to identify biological, epidemiological, observational, in

Effects of Quinoa (Chenopodium quinoa Willd.) Consumption on Markers of CVD Risk.

A number of epidemiological studies have suggested that diets rich in whole grains are linked to lower cardiovascular disease (CVD) risk and mortality. Quinoa, a pseudo-cereal, is included in the “whole grain” category but the effects of quinoa consumption in humans is not widely studied. Our aim was to undertake a dietary intervention study to investigate the effects of daily consumption of quinoa-enriched bread (providing 20 g quinoa flour) on CVD risk markers compared with a 100% refined wheat bread control. Thirty-seven healthy overweight men (35⁻70 years, body mass index >25 k

Effects of dietary antioxidants on the immune function of middle-aged adults.

The immune system is highly reliant on accurate cell-cell communication for optimal function, and any damage to the signalling systems involved will result in an impaired immune responsiveness. Oxidant-mediated tissue injury is a particular hazard to the immune system, since phagocytic cells produce reactive oxygen species as part of the body's defence against infection. Adequate amounts of neutralizing antioxidants are required, therefore, to prevent damage to the immune cells themselves. Many antioxidants can be obtained directly from the diet (e.g. ascorbic acid, alpha-tocopherol, carotenoi

[Hypercholesterolemia: therapeutic approach].

Epidemiological observations, experimental and clinical researches have laid special stress on the importance of hypercholesterolemia in the natural development of the atherosclerotic disease and its cardiovascular complications. Primary and secondary trials have demonstrated the benefits of cholesterol-lowering therapy to modify the evolution of atherosclerotic disease. In particular, it was observed a significant reduction of incidence and mortality due to coronary heart disease, that is the most common complication of atherosclerosis. At the present time, we have a new class of cholesterol-

Sex-specific gut microbial and metabolic responses to inhaled diesel exhaust particle exposure are modified by probiotic treatment in C57BL/6 mice.

BACKGROUND: Epidemiological and experimental studies indicate that susceptibility to traffic-generated particulate matter (PM)-induced metabolic dysfunction varies by sex, with females frequently exhibiting greater vulnerability. Recent evidence suggests that disruption of the lung-gut axis and subsequent gut-derived inflammatory signaling may contribute to these outcomes. However, the effects of inhaled PM on gut microbiome signaling and the metabolic milieu across sexes remain inadequately characterized. This study examined whether diesel exhaust particulate (DEP) exposure induces sex-specif

Light Exposure as a Modifiable Determinant of Mental Health.

PURPOSE OF REVIEW: Light is a fundamental environmental signal that shapes human physiology, behaviour, and mental health. Beyond vision, light exposure regulates circadian rhythms, sleep, neuroendocrine function, arousal, and brain circuits implicated in emotional regulation. This review synthesizes recent evidence linking light exposure to mental health and argues that light should be conceptualized as a core, modifiable component of the mental health exposome – the cumulative, dynamic set of environmental influences shaping mental health across the lifespan. RECENT FINDINGS: Recent randomiz

Diet quality and depressive symptoms in older adults, assessing the effect modification by genetic predisposition and low-grade inflammation: a target trial emulation.

Longitudinal studies have shown an association between diet quality and depression. However, reverse causality, unmeasured confounding, and selection bias remained important limitations. We aim to examine the relationship between diet quality and depression in older adults while addressing these issues and explore modification role of genetic predisposition to depression and low-grade inflammation. We emulated a target trial of dietary interventions using data from the ASPREE cohort. An ultra-processed food (UPF) index and an anti-inflammatory diet measure were extracted from a food frequency

Alcohol-Induced Dysregulation of Hydrogen Sulfide Signaling in Alzheimer's Disease-Narrative Mechanistic Synthesis Review.

Alcohol use disorder (AUD) is highly comorbid with psychiatric conditions and is increasingly recognized as a modifiable factor associated with cognitive decline and dementia, including Alzheimer's disease (AD). While epidemiological and experimental studies consistently demonstrate that chronic alcohol exposure exacerbates neurodegenerative vulnerability rather than implying a single dominant causal pathway, accumulating evidence supports a multifactorial and context-dependent framework in which alcohol acts as a disease-modifying stressor that perturbs endogenous adaptive and resilience mech

Global trends and socioeconomic disparities in cancer burden among adolescents and young adults, 1990-2021.

BACKGROUND: Adolescents and young adults (AYAs, 10-24 years) constitute a distinct cancer population in whom malignancy arises during critical life transitions yet leads to substantial long-term morbidity and socioeconomic consequences. However, their global cancer burden and risk-factor profile remain incompletely characterized, especially in low- and middle-income settings. METHODS: We conducted a secondary analysis of the Global Burden of Disease 2021 dataset, synthesizing estimates of cancer incidence, mortality, and disability-adjusted life years (DALYs) for 32 cancer types among AYAs ac
