The natural history of periodontal disease in man. The rate of periodontal destruction before 40 years of age.

This longitudinal study of randomly selected Norwegian students and academicians has shown that 50% of the 17-year olds have lost no periodontal support, and the other 50% exhibited slight localized loss of attachment primarily on buccal surfaces of first molars and first bicuspids of both jaws. At 21 all students show one or more of these lesions as well as loss of attachment on interproximal surfaces. At 30 years of age the mean cumulative loss is still less than 1 mm. As they approach 40 years of age the mean individual loss of attachment is slightly above 1.5 mm or 10% of the total periodo

Can Human Oral Mucosa Stem Cells Differentiate to Corneal Epithelia?

Human oral mucosa stem cells (hOMSCs) arise from the neural crest, they can self-renew, proliferate, and differentiate to several cell lines and could represent a good source for application in tissue engineering. Because of their anatomical location, hOMSCs are easy to isolate, have multilineage differentiation capacity and express embryonic stem cells markers such as-Sox2, Oct3/4 and Nanog. We have used SHEM (supplemented hormonal epithelial medium) media and cultured hOMSCs over human amniotic membrane and determined the cell's capacity to differentiate to an epithelial-like phenotype and t

Fine structure of bacterial adhesion to the epithelial cell membranes of the filiform papillae of tongue and palatine mucosa of rodents: a morphometric, TEM, and HRSEM study.

The palatine mucosa and filiform papillae of the dorsal tongue mucosae of rodents were examined using transmission electron microscopy (TEM) and high resolution scanning electron microscopy (HRSEM). In the HRSEM method, the samples were fixed in 2% osmium tetroxide, dehydrated in alcohol, critical point-dried, and coated with gold-palladium. In addition, the HRSEM technique was used for morphometric analysis (length, width, and length/width ratio of cocci and bacilli). For the TEM method, the tissues were fixed in modified Karnovsky solution (2.5% glutaraldehyde, 2% formalin in 0.1M sodium pho

Meningococcal biofilm growth on an abiotic surface - a model for epithelial colonization?

Neisseria meningitidis colonizes the human nasopharynx asymptomatically, often for prolonged periods, but occasionally invades from this site to cause life-threatening infection. In the nasopharynx aggregated organisms are closely attached to the epithelial surface, in a state in which the expression of components of the bacterial envelope differs significantly from that found in organisms multiplying exponentially in liquid phase culture or in the blood. We and others have hypothesized that here they are in the biofilm state, and to explore this we have investigated biofilm formation by the s

Optisol vs Dexsol as storage media for preservation of human corneal epithelium.

PURPOSE: To compare the efficacy of two storage media, Optisol GS and Dexsol, in preservation of donor corneal epithelium. METHODS: A total of 12 pairs of corneas not suitable for transplantation, all with intact epithelium, were used in this study, with one cornea of the pair stored in Optisol GS and its other counterpart in Dexsol. At each of three durations of storage--1, 2, and 4 days--four of these paired corneas were prepared for light microscopy and scanning and transmission electron microscopy. Another four pairs of control cornea were prepared in the same way and placed in universal

Characterization of the molecular interplay between Moraxella catarrhalis and human respiratory tract epithelial cells.

Moraxella catarrhalis is a mucosal pathogen that causes childhood otitis media and exacerbations of chronic obstructive pulmonary disease in adults. During the course of infection, M. catarrhalis needs to adhere to epithelial cells of different host niches such as the nasopharynx and lungs, and consequently, efficient adhesion to epithelial cells is considered an important virulence trait of M. catarrhalis. By using Tn-seq, a genome-wide negative selection screenings technology, we identified 15 genes potentially required for adherence of M. catarrhalis BBH18 to pharyngeal epithelial Detroit 5

Three Amino Acid Changes in Avian Coronavirus Spike Protein Allow Binding to Kidney Tissue.

Infectious bronchitis virus (IBV) infects ciliated epithelial cells in the chicken respiratory tract. While some IBV strains replicate locally, others can disseminate to various organs, including the kidney. Here, we elucidate the determinants for kidney tropism by studying interactions between the receptor-binding domain (RBD) of the viral attachment protein spike from two IBV strains with different tropisms. Recombinantly produced RBDs from the nephropathogenic IBV strain QX and from the nonnephropathogenic strain M41 bound to the epithelial cells of the trachea. In contrast, only QX-RBD bin

Antiviral potential of the arctigenin derivative COA in reducing viral adhesion to the epithelial cell surface against IHNV infection.

Infectious hematopoietic necrosis virus (IHNV) is a major aquaculture threat, highlighting the need for effective antiviral agents. This study evaluates the arctigenin derivative COA for its antiviral potential against IHNV. COA demonstrated no cytotoxicity to EPC cells at concentrations up to 10 μM and inhibited over 90 % of IHNV gene expression in vitro. It preserved normal nuclear morphology in infected cells, reduced viral titers, and completely blocked IHNV infection at 10 μM. Time-gradient assays identified COA's antiviral action during early infection, specifically inhibiting viral adso

The regeneration of gingiva: its potential value for the recession of healthy gingiva.

The partial withdrawal of healthy gingiva not only affects the appearance but also can bring about some complaints when the healthy gingiva is stimulated for some reasons. The junctional epithelium of gingiva moves to the root with aging, and compared with the tooth crown, the tooth root which has lower mineral content is prone to decay. Thus, gingival recession could lead to the root surface decay and make the tooth sensitive. Gingival recession is not reversible. Once the healthy gingiva shrinked, the teeth could feel uncomfortable, food impaction appeared and the original restorations have

Immunohistochemical study of nerve fibres with substance P- or calcitonin gene-related peptide-like immunoreactivity in the junctional epithelium of developing rats.

The beginning of innervation in the junctional epithelium of maxillary first molars was examined in gingival tissues from 19 to 32-day-old rats. Substance P- or calcitonin gene-related peptide (CGRP)-like immunoreactivity was demonstrated by the avidin-biotin peroxidase complex method. In 19-day-old rats, nerve fibres with substance P- or CGRP-like immunoreactivity were seen in the connective tissue and oral epithelium, but not in the reduced enamel epithelium, which would be transformed into the junctional epithelium. In 21-day-old rats, the fibres with substance P- or CGRP-like immunoreactiv

Pattern of periodontal breakdown in adult Tanzanians.

A total of 170 adult Tanzanians aged 30-69 yr were examined for loss of attachment, gingival recession, plaque, calculus and gingival bleeding on all surfaces of all teeth. The severity of loss of attachment varied considerably between tooth types. Irrespective of age, mandibular incisors and first and second molars were the teeth most affected by loss of attachment. In all age groups heavy plaque deposits and gingival bleeding occurred more frequently in posterior than in anterior teeth. Dental calculus was most frequently observed in the maxillary posterior and mandibular anterior teeth. The

Tooth eruption: evidence for the central role of the dental follicle.

The roles of the gubernaculum dentis, root formation, tooth crown and dental follicle in prefunctional eruption of a mandibular premolar have been studied in nine beagle dogs by radiographic and histologic evaluations of the effects of surgical ablation or removal of these structures on tooth eruption. The dental follicle was the only one of these structures required for the coordinated enlargement of the eruption pathway and formation of bone in the base of the bony crypt, the radiographic and histologic hallmarks of tooth eruption. These data, together with the topographic relationships of t

Drug susceptibility testing of circulating lung cancer cells for personalized treatment.

The presence of Circulating tumor cells (CTCs) has been proven to be correlated with disease progression and the patient's response to treatment. However, the culture of CTCs for clinical utility is still a big challenge. We have developed a short-term method that enables CTCs culture and provides an opportunity to monitor drug susceptibility testing in individual patients. In a proof-of-concept study, we established a unique method using Matrigel® coated in 96 well plate to enable cancer cell clusters to attach and proliferate. The culture method using Matrigel® provides in vitro conditions a

Leishmania development in sand flies: parasite-vector interactions overview.

Leishmaniases are vector-borne parasitic diseases with 0.9 - 1.4 million new human cases each year worldwide. In the vectorial part of the life-cycle, Leishmania development is confined to the digestive tract. During the first few days after blood feeding, natural barriers to Leishmania development include secreted proteolytic enzymes, the peritrophic matrix surrounding the ingested blood meal and sand fly immune reactions. As the blood digestion proceeds, parasites need to bind to the midgut epithelium to avoid being excreted with the blood remnant. This binding is strictly stage-dependent as
