[Antineutrophil cytoplasmic antibodies in patients with sickle cell anemia].

PURPOSE: To determine whether anti neutrophil cytoplasmic antibodies (ANCA): anti myeloperoxidase (anti MPO) and anti proteinase 3 (anti PR3), are present in patients with sickle cell anaemia (SCA) during painful crisis. PATIENTS AND METHODS: 55 patients with SCA were included in this study, 35 of them had painful crisis and the rest were in steady state. We determined levels of anti MPO and anti PR3 by two enzyme immunoassays commercially available (Progen Biotechnik GMBH). RESULTS: A significant increase of anti MPO in the group of patients with painful crisis was found in comparison with

[Screening tests for malignant hyperthermia susceptibility].

The ideal screening test for malignant hyperthermia susceptibility (MHS) has yet to be discovered. It should be simple noninvasive, yet totally specific and sensitive. Until such an ideal test becomes available, allowing simple routine preoperative screening, tests should only be used in certain specific situations. These include: patients in whom a clinical crisis was suspected; the members of the family of a subject labeled MHS because of a fatal, or otherwise, crisis, or in whom tests were positive; patients with other pathological conditions which could be linked to malignant hyperthermia

[Improvement of the flow properties of the blood of patients by low-molecular hydroxyethyl starch (Expafusin)].

10 patients with various diseases causing elevated ESR-values and impaired flow properties of blood received infusions of 500 ml of low molecular weight hydroxyethylstarch (HES 6%, Expafusin) within approximately 30 minutes. Measurements of rheological parameters for several hours after infusion showed a statistically significant decrease of apparent whole blood viscosity and of plasma viscosity. RBC aggregation was slightly, but statistically significantly reduced, whereas RBC filterability remained unchanged. These data obtained with blood from patients with altered flow properties of blood

The Red Blood Cell-Inflammation Vicious Circle in Sickle Cell Disease.

Sickle cell disease (SCD) is a genetic disease caused by a single mutation in the β-globin gene, leading to the production of an abnormal hemoglobin called hemoglobin S (HbS), which polymerizes under deoxygenation, and induces the sickling of red blood cells (RBCs). Sickled RBCs are very fragile and rigid, and patients consequently become anemic and develop frequent and recurrent vaso-occlusive crises. However, it is now evident that SCD is not only a RBC rheological disease. Accumulating evidence shows that SCD is also characterized by the presence of chronic inflammation and oxidative stress

No-reflow in acute ischemic stroke: mechanisms, measurements, and molecular markers.

BACKGROUND: No-reflow is a critical microvascular phenomenon in acute ischemic stroke in which patients fail to achieve adequate tissue reperfusion despite successful recanalization of a large vessel occlusion. It affects at least 25% of patients after reperfusion and results from persistent microvascular dysfunction that prevents restoration of normal blood flow to ischemic tissue, leading to ongoing injury. OBJECTIVE: This paper synthesizes historical, mechanistic, diagnostic, and therapeutic perspectives on the no-reflow phenomenon to clarify its pathophysiology and highlight gaps limiting

Structural and functional characteristics of the pulmonary hemomicrocirculatory bed in induced systemic sclerosis: an experimental study.

The objective of this study was to investigate the effects of prolonged exposure to the oxidative agent NaClO on histopathological changes in the lung tissues of laboratory animals. Specifically, the study aimed to examine morphological changes in the pulmonary microcirculation and the level of vascular cell adhesion molecule-1 (VCAM-1) as a functional activity indicator of endothelial cells in animals with induced systemic sclerosis (SSc). A laboratory animal model was used to assess the impact of long-term exposure to NaClO on lung tissues. The animals were divided into three groups: the exp

Haem oxygenase protects against thrombocytopaenia and malaria-associated lung injury.

BACKGROUND: Malaria-triggered lung injury can occur in both severe and non-severe cases. Platelets may interact with parasitized erythrocytes, leukocytes and endothelium. These interactions can lead to microvessel obstructions and induce release of inflammatory mediators. Induction of the haem oxygenase enzyme is important in the host's response to free haem and to several other molecules generated by infectious or non-infectious diseases. In addition, an important role for the haem oxygenase-1 isotype has been demonstrated in experimental cerebral malaria and in clinical cases. Therefore, the

Haematopoietic effects induced in mice by the snake venom toxin jararhagin.

Venom toxins have been tested as anti-thrombotic, and anti-metastatic drugs in experimental models. The jararhagin toxin, from Bothrops jararaca snake venom, acts upon several biological processes, as inflammation, pain, platelet aggregation, etc. In this article, the systemic effects of intra-peritoneal injections of different jararhagin doses were determined in mice. About 50% significant decrease was observed in total blood leukocytes in the first (48 ng), and second (24 ng) weeks. The reduction of lymphocytes, monocytes and neutrophils accounted for this leucopoenia up to the sixth week. S

Myeloperoxidase in the pathogenesis of sickle cell disease.

Sickle cell disease (SCD) is an inherited disorder characterized by abnormal hemoglobin molecules that cause the sickling of red blood cells (RBCs) which occlude blood vessels, increase hemolysis, and leads to chronic inflammation. Accumulating evidence has strongly linked SCD and other inflammatory conditions to the persistent activation of neutrophils which generate the antimicrobial enzyme myeloperoxidase (MPO) that produces hypohalous acids such as hypochlorous acid (HOCl) and hypothiocyanous acid. The MPO-HOCl system can induce oxidative damage of erythrocytes and vasculature through the

Huntington's disease: the current state of research with peripheral tissues.

Huntington's disease (HD) is a genetically dominant condition caused by expanded CAG repeats. These repeats code for a glutamine tract in the HD gene product huntingtin (htt), which is a protein expressed in almost all tissues. Although most HD symptoms reflect preferential neuronal death in specific brain regions, even before the HD gene was identified numerous reports had described additional abnormalities in the peripheral tissues of HD patients, including weight loss, altered glucose homeostasis, and sub-cellular abnormalities in fibroblasts, lymphocytes and erythrocytes. Several years hav

Effective prevention of microbial biofilm formation on medical devices by low-energy surface acoustic waves.

Low-energy surface acoustic waves generated from electrically activated piezo elements are shown to effectively prevent microbial biofilm formation on indwelling medical devices. The development of biofilms by four different bacteria and Candida species is prevented when such elastic waves with amplitudes in the nanometer range are applied. Acoustic-wave-activated Foley catheters have all their surfaces vibrating with longitudinal and transversal dispersion vectors homogeneously surrounding the catheter surfaces. The acoustic waves at the surface are repulsive to bacteria and interfere with th

Gastric intramural hematoma and hemoperitoneum in a captive northern fur seal.

A 16-yr-old adult male northern fur seal (Callorhinus ursinus) was found dead in its outdoor pool in November 1995. The animal was maintained at Mystic Aquarium (Mystic, Connecticut, USA) from March 1980 to November 1995. Gross necropsy findings included hemoperitoneum and locally extensive gastric intramural hemorrhage that involved the posterior fundic, antral, and pyloric regions and extended into the duodenum. The gastric mural thickening grossly resembled hemangioma, and the gastric serosa was ruptured at the site of maximal mural expansion. In histologic sections of the stomach, a cribif

C3 receptors on granulocytes from patients with rheumatoid arthritis and Felty's syndrome.

The expression of receptors for the third component of complement (C3R) on polymorphonuclear cells (PMN) from patients with rheumatoid arthritis (RA) or Felty's syndrome (FS) and healthy controls (HC) was studied by means of a rosetting technique using ox erythrocytes coated with rabbit IgM and mouse complement. The percentage of rosette forming PMN (RF-PMN) was found to be increased in patients with FS (59%) and patients with RA (47%) compared to HC (18%), when measured directly after cold isolation of PMN. Incubation at 37 degrees C of cold isolated cells increased the percentage of RF-PMN i
