Nowadays, tissue and organ failures resulted from injury, aging accounts, diseases or other type of damages is one of the most important health problems with an increasing incidence worldwide. Current treatments have limitations including, low graft efficiency, shortage of donor organs, as well as immunological problems. In this context, tissue engineering (TE) was introduced as a novel and versatile approach for restoring tissue/organ function using living cells, scaffold and bioactive (macro-)molecules. Among these, scaffold as a three-dimensional (3D) support material, provide physical and

UNLABELLED: Anterior cruciate ligament (ACL) is one of the most difficult tissues to heal once injured. Ligament regeneration and tendon-bone junction healing are two major goals of ACL reconstruction. This study aimed to investigate the synergistic therapeutic effects of Stromal cell-derived factor 1 (SDF-1)-releasing collagen-silk (CSF) scaffold combined with intra-articular injection of ligament-derived stem/progenitor cells (LSPCs) for ACL regeneration and the amelioration in the long-term complication of osteoarthritis (OA). The stem cell recruitment ability of CSF scaffold and the multip

Implant-based breast reconstruction is a common standard of care for breast cancer patients following mastectomy. To support implant weight and placement, surgeons utilize autologous tissues, acellular dermal matrices (ADMs), or synthetic meshes. While ADMs provide structural support, they present significant risks, including infection transmission and seroma. Conversely, synthetic meshes exhibit poor cellular adhesion, leading to inadequate tissue integration. To address these challenges, there is a need for a matrix that enhances tissue integration while minimizing infection risks and other

Spider silks are the toughest known biological materials, yet are lightweight and virtually invisible to the human immune system, and they thus have revolutionary potential for medicine and industry. Spider silks are largely composed of spidroins, a unique family of structural proteins. To investigate spidroin genes systematically, we constructed the first genome of an orb-weaving spider: the golden orb-weaver (Nephila clavipes), which builds large webs using an extensive repertoire of silks with diverse physical properties. We cataloged 28 Nephila spidroins, representing all known orb-weaver

Metal-free molecular magnets differ from conventional magnetic devices that predominantly rely on metallic magnets as these metal-free materials may offer biocompatibility and bioresorbability. However, existing molecular materials lack the ferromagnetic strength required for high-fidelity sensing at clinically relevant tissue depths. Here we introduce a bioresorbable metal-free molecular magnet derived from graphitic carbon nitride, combining metallic-level ferromagnetism with full bioresorbability. De-nitrogenation creates spin centres coupled through a reconstructed π-σ conjugated framework

Irregular bone defects demand biomaterials that can be delivered into complex shapes while offering gradually developing mechanical strength and bioactive electrical stimulation-two critical properties rarely obtained simultaneously. Here, we develop an injectable, polymer-induced self-reinforcing and piezoelectric hydrogel engineered from regenerated silk fibroin, tyramine-modified CMC, and MXene-cellulose nanofiber nanosheets through enzymatically dynamic Cross-linking. This material is designed to overcome two major limitations of current injectable hydrogels: inadequate mechanical maturati

OBJECTIVE: Chronic tympanic membrane perforations generally require surgical repair, most often via tympanoplasty. While effective, these procedures involve anesthesia, carry significant costs, and may not always be accessible. The objective of this review is to explore biomaterials and biological therapies that could promote tympanic membrane regeneration and potentially reduce the need for surgery.
DATA SOURCES: PubMed and Google Scholar between September 1958 and December 2025.
REVIEW METHODS: A PRISMA-guided systematic review of clinical studies about biomaterials or biological therapies

The clinical intractability of diabetic foot ulcers stems from a profound uncoupling of cutaneous neurovascular networks, rendering standard metabolic and topical interventions largely palliative. Paradoxically, remote orthopedic trauma robustly accelerates distal skin repair, yet the systemic molecular mediators driving this "bone-skin crosstalk" remain undefined, precluding its translation into non-invasive therapies. Here, we establish that macroscopic bone fracture expedites diabetic wound healing through the systemic release of exosomal miR-130b-3p, a potent orchestrator of coupled angiog

Advancements in drug delivery systems (DDSs) have revolutionized pharmaceutical development by enhancing therapeutic precision, minimizing off-target effects, and improving patient compliance. Modern DDS technologies, such as lipid nanoparticles (LNPs), liquid-liquid phase separated (LLPS) systems, inorganic nanoparticles, GalNac systems, small molecules, dendrimers, polymers, peptides, and silk fibroin (SF) hydrogels, have significantly expanded the range of anti-cancer therapeutics. These advancements extend beyond conventional chemotherapeutics to include biologics such as nucleic acids (NA

Spinal cord injury (SCI) is a devastating trauma to the central nervous system, causing permanent functional nerve defects. A key therapeutic challenge is the inhibition of the secondary injury cascade, specifically the progressive neural damage from iron overload-induced ferroptosis and oxidative stress. To target these dual mechanisms, we developed a dual-functional, iron-scavenging, and hydrogen-releasing microneedle patch (MN/MON@AB) composed of ammonia borane (AB)-loaded, amino-functionalized mesoporous organosilica nanoparticles (MON-NH2) embedded in a biodegradable silk fibroin array. Drug-coated balloon catheters are widely used to dilate narrowed arteries and deliver therapeutic agents to atherosclerotic plaques; however, uncontrolled passive drug leakage during catheter navigation remains a challenge. Herein, we developed a mechanoresponsive drug-releasing silk balloon catheter that actively releases drugs upon balloon inflation while minimizing passive drug leakage. To this end, we designed a molecular template that induces densely packed hydrophobic domains within the silk fibroin network. These domains enable strong binding and mechanoresponsive release of hydrophobic

Photothermal therapy (PTT) is widely recognized for the treatment of infected wounds. However, the wound hyperpathia caused by infection and heat from PTT is often overlooked, which might affect wound healing significantly. In this study, we systematically investigate the management of pain in bacteria-infected wounds using PTT, and develop a photothermal antibacterial hydrogel capable of sustained and controlled analgesia system (STB-Lid Gel), aiming to simultaneously achieve efficient anti-infection and precise pain management. STB-Lid Gel is constructed from a silk fibroin (SF) hydrogel mat

The in vivo performance of tissue-engineered heart valves remains constrained by a persistent early failure triad at the blood-material interface, namely thrombosis, unresolved inflammation, and slow endothelialization. Here, a bioinstructive multilayer valve scaffold is engineered to address these interfacial barriers through spatially integrated structural and biological functions. The scaffold comprises a digitally programmable 3D-printed framework, a silk fibroin wrapping layer, and a hydrogel biointerface incorporating Arg-Gly-Asp/GelMA adhesive cues together with H2S-releasing microgels.

Critical-sized maxillofacial bone defects are frequently exposed to pathological mechanical overload (MO), which disrupts cellular mechanosensing and compromises bone regeneration. However, strategies that protect osteogenic cells from overload-induced mechanobiological dysfunction remain limited. Here, we develop a biomimetic scaffold incorporating liposomal quercetin (LQ) into a collagen/silk fibroin/nano-hydroxyapatite matrix (LQ-CSH) to restore osteoblast function under pathological loading conditions. MO induces cytoskeletal disorganization and oxidative stress, leading to inactivation of

Medication-related osteonecrosis of the jaw (MRONJ), a severe complication of antiresorptive agents, such as zoledronate (ZOL), significantly affects patients' quality of life. Impaired angiogenesis and vascular endothelial cell (EC) function are core features of MRONJ, although the underlying mechanisms remain largely unknown. In this study, we show that ZOL induces human umbilical vein endothelial cell (HUVEC) dysfunction, which is characterized by suppressed cell viability, migration, tube formation, and angiogenesis-osteogenesis uncoupling. ZOL triggers reactive oxygen species (ROS) accumu

Infected bone defects (IBDs) represent a major clinical challenge in which persistent infection, microenvironmental dysregulation, and impaired regeneration reinforce a vicious cycle that is difficult to resolve. Here, we report carbon dot-functionalized Haversian-mimetic scaffolds designed to combine antibacterial activity with regenerative support in IBDs. A 3D-printed scaffold fabricated by digital light processing is constructed by integrating human hair-derived carbon dots (CrCi-CDs) into a silk fibroin methacrylate (SilMA)/gelatin methacrylate (GelMA) network, yielding the CrCi-CDs/SilMA

Traumatic brain injury (TBI) results in severe neurological dysfunction, and effective morphological and functional recovery remains a challenge. Currently, safe and efficient strategies for neural regeneration and repair are lacking. Both human umbilical cord mesenchymal stem cell (hUCMSC)-derived exosomes and three-dimensional bioprinted scaffolds have exhibited application potential during neural repair. Nevertheless, the combined function of 3D-printed collagen/silk fibroin (3D-CS) scaffolds with injury-preconditioned exosomes obtained from hUCMSCs in the repair of traumatic brain injury r

BACKGROUND: With an in-depth understanding of cardiac cell differentiation, cell therapy derived from stem cells has shown promising therapeutic effects in the treatment of myocardial infarction (MI). Although many types of cardiac or noncardiac cells have been found to play protective roles in MI, the specific role of endocardial cells (ECCs) in MI has not been reported.
METHODS: The current study was designed to determine whether human embryonic stem cell (hESC)-derived endocardial cells (hESC-ECCs) could be protective against MI. We first developed a cell delivery system by constructing a

Uterine diverticulum (UD) is characterized by localized thinning of the uterine myometrium, leading to the formation of recessed structures. While its mechanisms remain undefined, UD, often an aftermath of cesarean sections, mandates subsequent surgical intervention, highlighting the critical need for intraoperative prevention. Drawing inspiration from the natural anatomical structure of the uterus, we developed biomimetic, anisotropic, multi-channel silk fibroin-based uterine bioscaffolds (BSBs). The BSBs, characterized by aligned fibrous structures and hollow channels, significantly promoted
