Genome-Wide Identification and Bioinformatics Analysis of Thaumatin-like Proteins in Aegiceras corniculatum.

BACKGROUND: The mangrove ecosystem serves as a vital coastal ecotone, providing essential ecological services, such as water purification, shoreline protection, and biodiversity maintenance. Fungal pathogens threaten mangrove health and contribute to ecosystem degradation, but the molecular mechanisms underlying disease resistance in mangroves remain poorly explored. Introdustion: Thaumatin-like proteins (TLPs), belonging to the pathogenesis-related-5 (PR-5) family, play a crucial role in antifungal defense in plants. METHOD AND RESULTS: In this study, we identified 23 TLP family members in t

Arabidopsis AtERF014 acts as a dual regulator that differentially modulates immunity against Pseudomonas syringae pv. tomato and Botrytis cinerea.

ERF transcription factors play critical roles in plant immune responses. Here, we report the function of AtERF014, a nucleus-localized transcriptional activator, in Arabidopsis immunity. Expression of AtERF014 was induced by Pseudomonas syringae pv. tomato (Pst) and Botrytis cinerea (Bc). AtERF014-overexpressing (OE) plants displayed increased Pst resistance but decreased Bc resistance, whereas AtERF014-RNAi plants exhibited decreased Pst resistance but increased Bc resistance. After Pst infection, expression of salicylic acid (SA)-responsive genes AtPR1 and AtPR5 in AtERF014-OE plants and of

Integrated physiological, biochemical, and gene expression responses of tomato (Solanum lycopersicum L.) to time-dependent salinity stress.

BACKGROUND: Salinity is a major abiotic stress factor limiting tomato production by affecting plant growth, physiology, and metabolism. This study investigated the time-dependent physiological, biochemical, and gene expression responses of tomato (Solanum lycopersicum L.) cultivar H2274 under different salinity levels (control, 1.5, 3.0, and 4.5 dS m⁻¹). Plants were sampled on the 15th and 45th days of stress exposure. Physiological parameters, including chlorophyll content and leaf color, biochemical responses associated with antioxidant activity and lipid peroxidation, and the expression of

VvGAMYB Orchestrates Multi-Signaling Networks to Modulate GA-Induced Parthenocarpic Seedless Berry Development in Grapes.

Exogenous gibberellin (GA) efficiently induces seeded grape parthenocarpy and improves berry quality, with VvGAMYB serving as a key response factor. However, the VvGAMYB-mediated regulatory networks underlying this process remain unclear. In this study, GA3 treatment induced ovule abortion in 'Wink' grape, leading to the formation of parthenocarpic seedless berries. Exogenous GA drastically repressed VvGAMYB levels, thereby promoting parthenocarpy in grape ovaries. Through protein interaction screening, we identified 181 predicted VvGAMYB-associated proteins, which include differentially expre

Phyto-Oxylipins as Key Regulators of Plant Resilience to Abiotic and Biotic Stress.

Phyto-oxylipins, oxidized derivatives of unsaturated fatty acids, serve as crucial mediators in plant responses to biotic and abiotic stresses, which are becoming increasingly frequent and severe under changing climatic conditions. These signaling molecules, produced enzymatically or spontaneously, orchestrate a wide range of physiological and molecular responses that enhance plant resilience. By scavenging reactive oxygen species (ROS) and upregulating antioxidant enzymes, phyto-oxylipins help mitigate oxidative damage, thus preserving cellular integrity and sustaining growth under environmen

Auxin signaling.

Following its discovery nearly a century ago, the signaling molecule auxin has captured the interest of plant scientists. Mostly through genetics, key components of the auxin response were identified, and these were rapidly connected in a topologically simple nuclear auxin pathway (NAP). While the identification of the NAP established a coherent framework for transcriptional auxin response, recent years have seen a remarkable series of advances in the evolution, mechanisms, and regulation of NAP components that shed new light on auxin response. Furthermore, it has long been known that auxin tr

Investigation of the Jasmonate ZIM-Domain Family Reveals PavJAZ8 Regulates Fruit Aroma Traits in Sweet Cherry (Prunus avium L.).

Jasmonate ZIM-domain (JAZ) family genes, which belong to TIFY family, are plant-specific transcriptional repressors. As key regulators of the jasmonic acid signaling pathway, JAZ proteins play crucial roles in various aspects of plant biology. However, the identification and functional characterization of JAZ genes in sweet cherry (Prunus avium L.) fruit remain unknown. In the present study, we identified nine JAZ members in the sweet cherry genome. We systematically analyzed the gene structures, protein domains, evolutionary relationships, and physicochemical properties of these members and a

Jasmonic acid improves cadmium tolerance in rice (Oryza sativa) by reducing the production of nitric oxide.

The involvement of jasmonic acid (JA) in the rice's response to cadmium (Cd) stress is well recognized, though the underlying mechanisms remain unclear. In this study, exposure to Cd stress rapidly elevated endogenous JA concentrations in rice roots, meanwhile, a mutant coleoptile photomorphogenesis 2 (cpm2) which produces less JA, was more sensitive to Cd stress than its wild type (WT). JA mitigated Cd toxicity by decreasing Cd absorption in root cell wall and shoot, which was achieved by up-regulating the expression of the Cd-chelation and efflux-related genes such as OsHMA3, OsABCG36 and Os

Hydrogen sulfide interferes with ethylene biosynthesis and signaling pathway in tomato by the mediation of SlERF.D2 persulfidation.

Hydrogen sulfide (H2S), as a signaling molecule, is found to delay fruit ripening and senescence by antagonizing the biosynthesis and signaling of ethylene, whereas the mechanism remains unclear. In the current work, exogenous H2S fumigation could alleviate tomato fruit ripening and an ethylene response factor SlERF.D2 was found to be persulfidated at Cys35 by mass spectrometry analysis. Meanwhile, ethylene biosynthesis related genes SlACS1 and SlACO3 were significantly downregulated at gene expression level in H2S-treated fruit. By CRISPR/Cas9 and gene overexpression, we showed that overexpre

Signaling Crosstalk between Salicylic Acid and Ethylene/Jasmonate in Plant Defense: Do We Understand What They Are Whispering?

During their lifetime, plants encounter numerous biotic and abiotic stresses with diverse modes of attack. Phytohormones, including salicylic acid (SA), ethylene (ET), jasmonate (JA), abscisic acid (ABA), auxin (AUX), brassinosteroid (BR), gibberellic acid (GA), cytokinin (CK) and the recently identified strigolactones (SLs), orchestrate effective defense responses by activating defense gene expression. Genetic analysis of the model plant Arabidopsis thaliana has advanced our understanding of the function of these hormones. The SA- and ET/JA-mediated signaling pathways were thought to be the b

Identification of olivetolic acid cyclase from Cannabis sativa reveals a unique catalytic route to plant polyketides.

Δ(9)-Tetrahydrocannabinol (THC) and other cannabinoids are responsible for the psychoactive and medicinal properties of Cannabis sativa L. (marijuana). The first intermediate in the cannabinoid biosynthetic pathway is proposed to be olivetolic acid (OA), an alkylresorcinolic acid that forms the polyketide nucleus of the cannabinoids. OA has been postulated to be synthesized by a type III polyketide synthase (PKS) enzyme, but so far type III PKSs from cannabis have been shown to produce catalytic byproducts instead of OA. We analyzed the transcriptome of glandular trichomes from female cannabis

Plant-to-plant defence induction in cotton is mediated by delayed release of volatiles upon herbivory.

Caterpillar feeding immediately triggers the release of volatile compounds stored in the leaves of cotton plants. Additionally, after 1 d of herbivory, the leaves release other newly synthesised volatiles. We investigated whether these volatiles affect chemical defences in neighbouring plants and whether such temporal shifts in emissions matter for signalling between plants. Undamaged receiver plants were exposed to volatiles from plants infested with Spodoptera caterpillars. For receiver plants, we measured changes in defence-related traits such as volatile emissions, secondary metabolites, p

Hulless barley is a cold-resistant crop widely planted in the northwest plateau of China. It is also the main food crop in this region. Phosphorus (P), as one of the important essential nutrient elements, regulates plant growth and defense. This study aimed to analyze the development and related molecular mechanisms of hulless barley under P deficiency and explore the regulatory genes so as to provide a basis for subsequent molecular breeding research. Transcriptome analysis was performed on the root and leaf samples of hulless barley cultured with different concentrations of KH2PO4 (1 mM and

Single-cell RNA-sequencing profiles reveal the developmental landscape of hawthorn leaves.

Hawthorn (Crataegus spp.) is an economically and medicinally important genus in Rosaceae. Its leaves accumulate valuable secondary metabolites, including flavonoids, and have long been used in traditional Chinese medicine. Despite available chromosome-level genome assemblies and bulk transcriptome data, the cellular composition of hawthorn leaves, their developmental trajectories, and the cell-type-specific distribution of biosynthetic gene expression remain uncharacterized. Here we report the first single-cell transcriptomic atlas of hawthorn leaves, generated by integrating protoplast-based

The Compact Root Architecture 2 systemic pathway is required for the repression of cytokinins and miR399 accumulation in Medicago truncatula N-limited plants.

Legume plants can acquire mineral nitrogen (N) either through their roots or via a symbiotic interaction with N-fixing rhizobia bacteria housed in root nodules. To identify shoot-to-root systemic signals acting in Medicago truncatula plants at N deficit or N satiety, plants were grown in a split-root experimental design in which either high or low N was provided to half of the root system, allowing the analysis of systemic pathways independently of any local N response. Among the plant hormone families analyzed, the cytokinin trans-zeatin accumulated in plants at N satiety. Cytokinin applicati

CircRNAs responsive to winter dormancy and spring flushing conditions of tea leaf buds.

Circular RNAs (circRNAs) are important regulators of diverse biological processes of plants. However, the evolution and potential functions of circRNAs during winter dormancy and spring bud flushing of tea plant is largely unknown. Using RNA-seq data, a total of 1184 circRNAs were identified in the winter dormant and spring bud flushing leaf samples of tea plants in two different cultivars exhibiting different duration of winter dormancy. A total of 156 circRNAs are found to be differentially expressed and the weighted gene co-expression network (WGCNA) analysis revealed that 22 and 20 differe

Benzothiadiazole, a plant defense inducer, negatively regulates sheath blight resistance in Brachypodium distachyon.

Plant defense inducers that mimic functions of the plant immune hormone salicylic acid (SA) often affect plant growth. Although benzothiadiazole (BTH), a synthetic analog of SA, has been widely used to protect crops from diseases by inducing plant defense responses, we recently demonstrated that SA, but not BTH, confers resistance against Rhizoctonia solani, the causal agent of sheath blight disease, in Brachypodium distachyon. Here, we demonstrated that BTH compromised the resistance of Bd3-1 and Gaz4, the two sheath blight-resistant accessions of B. distachyon, which activate SA-dependent si

Genome‑wide analysis of the CaWRKY transcription factors reveals CaWRKY28 as a candidate gene for cold tolerance breeding in Coffea arabica L.

BACKGROUND: In China, over 98% of coffee is produced in Yunnan Province, where Coffea arabica L. predominates. Cold stress represents a major genetic constraint limiting coffee production, especially during the fruiting stage. WRKY transcription factors (TFs) are key regulators of stress responses, yet their systematic characterization and breeding value in C. arabica L. are remain largely unexplored. RESULTS: Here, we conducted a genome-wide analysis of WRKY genes across four C. arabica L. genomes, identifying 400 CaWRKY members phylogenetically classified into three groups. Segmental duplic

The R2R3MYB VvMYBPA1 from grape reprograms the phenylpropanoid pathway in tobacco flowers.

This work shows that, in tobacco, the ectopic expression of VvMYBPA1 , a grape regulator of proanthocyanidin biosynthesis, up- or down-regulates different branches of the phenylproanoid pathway, in a structure-specific fashion. Proanthocyanidins are flavonoids of paramount importance for animal and human diet. Research interest increasingly tilts towards generating crops enriched with these health-promoting compounds. Flavonoids synthesis is regulated by the MBW transcriptional complex, made of R2R3MYB, bHLH and WD40 proteins, with the MYB components liable for channeling the complex towards s

Salicylic Acid Is Required for Broad-Spectrum Disease Resistance in Rice.

Rice plants contain high basal levels of salicylic acid (SA), but some of their functions remain elusive. To elucidate the importance of SA homeostasis in rice immunity, we characterized four rice SA hydroxylase genes (OsSAHs) and verified their roles in SA metabolism and disease resistance. Recombinant OsSAH proteins catalyzed SA in vitro, while OsSAH3 protein showed only SA 5-hydroxylase (SA5H) activity, which was remarkably higher than that of other OsSAHs that presented both SA3H and SA5H activities. Amino acid substitutions revealed that three amino acids in the binding pocket affected SA
