A practical guide for nephrologist peer reviewers: understanding and appraising Mendelian randomization studies.

Identifying risk factors for disease onset and progression has been a core focus in nephrology research. Mendelian Randomization (MR) has emerged as a powerful genetic epidemiological approach, utilizing genome-wide association studies (GWAS) to establish causal relationships between modifiable risk factors and kidney disease outcomes. MR uses genetic variants as instrumental variables to infer causal relationships between exposures and disease outcomes. This method leverages the natural randomization of genetic variants to balance confounders, akin to matched cohorts in observational research

Genetic insights into the therapeutic potential of hydrogen sulfide.

Hydrogen sulfide (H2S) is a chalcogen-hydrogen gas that can be endogenously synthesized and is the third known bioactive gaseous signalling molecule (gasotransmitter). H2S has diverse physiological roles, such as with cell division, neurotransmission and inflammation. Its involvement with a variety of disease-related processes has spurred growing interest in its therapeutic potential. We propose that variants in genes implicit in regulating cellular H2S availability could help prioritise conditions for exploration of H2S as a therapeutic target. Here, we present the first systematic genetic in

Causal associations of COVID-19 on neurosurgical diseases risk: a Mendelian randomization study.

Many researchers have explored the potential association between one neurosurgical disease and coronavirus disease 2019 (COVID-19), but few systematically analyzed the association and causality between COVID-19 and various neurosurgical diseases. A Mendelian randomization analysis was conducted to evaluate the causal association between COVID-19 (including critically ill COVID-19, hospitalized COVID-19, and respiratory syndrome coronavirus 2 (SARS-CoV-2) infection) and 30 neurosurgical diseases within European populations. The consequences of inverse variance weighted models suggest that genet

Overlapping Genetic Architecture Between Schizophrenia and Neurodegenerative Disorders.

Epidemiological and clinical studies have suggested comorbidity between schizophrenia and several neurodegenerative disorders. However, little is known whether there exists shared genetic architecture. To explore their relationship from a genetic and transcriptomic perspective, we applied polygenic and linkage disequilibrium-informed methods to examine the genetic correlation between schizophrenia and amyotrophic lateral sclerosis (ALS), Parkinson's disease, Alzheimer's disease and frontotemporal dementia. We further combined genome-wide association summary statistics with large-scale transcri

Causal relationship between diabetes and urinary tract infections: A Mendelian randomization study.

Diabetes mellitus (DM) is associated with many infectious diseases, but the relationship between diabetes and urinary tract infections remains controversial. Therefore, we investigated, by Mendelian randomization (MR), the causal relationship between type 1 DM (T1DM), type 2 DM (T2DM), and 3 common urinary tract infections at different sites. We evaluated the causal relationship between T1DM and T2DM and pyelonephritis, cystitis, and urethritis in a two-sample MR study, using the inverse variance weighted (IVW) method for the primary analysis and MR-Egger and weighted median estimation methods

Causal associations between dietary habits and age at natural menopause: A two-sample diet-wide Mendelian randomization study.

BACKGROUND: Menopause, the natural cessation of menstrual cycles, marks a significant phase in a woman's life and is associated with various health outcomes. While previous observational studies have suggested associations between dietary habits and the age at natural menopause (ANM), their findings have been inconsistent and often confounded, making it difficult to infer causality. This study aims to assess the causal relationships between dietary habits and ANM using a two-sample diet-wide Mendelian randomization (MR) approach. METHODS: The study utilized data from 445,779 participants in t

From infection to tumor: genetic evidence of viral antibody immune response' role in urologic cancer development.

BACKGROUND: Urologic tumors are among the most common malignancies worldwide, and the association between chronic infections and the risk of developing these tumors has garnered significant attention. However, traditional observational studies are prone to confounding factors, making it challenging to establish a clear causal relationship. METHOD: This study employs a two-sample bidirectional Mendelian randomization analysis, utilizing genetic data on antibody levels and urologic tumors obtained from GWAS databases. The inverse variance weighted (IVW) method was used to estimate causal relati

The crucial role of immune factors in susceptibility to SARS-CoV-2 infection and severity of COVID-19.

Susceptibility to SARS-CoV-2 infection and severity of COVID-19 have high heterogeneity, but the underlying factors for such heterogeneity were largely unknown. This study aimed to investigate whether genetically determined immune phenotypes contribute to the variability in COVID-19 outcomes, by integrating large-scale GWAS data to assess genetic correlations, causal associations, and pleiotropic gene functions. Based on the summary statistics from GWAS analyses for immune phenotypes and COVID-19 outcomes, comprehensive analyses were performed to elucidate the associations between immune pheno

Complex effects of dyslexia risk factors account for ADHD traits: evidence from two independent samples.

BACKGROUND: Developmental dyslexia (DD) and attention deficit/hyperactivity disorder (ADHD) are among the most common neurodevelopmental disorders, whose etiology involves multiple risk factors. DD and ADHD co-occur in the same individuals much more often than would be expected by chance. Several studies have found significant bivariate heritability, and specific genes associated with either DD or ADHD have been investigated for association in the other disorder. Moreover, there are likely to be gene-by-gene and gene-by-environment interaction effects (G × G and G × E, respectively) underlying
