PURPOSE: Rett syndrome (RTT) is a neurological disorder marked by the loss of developmental milestones. Classic RTT involves variants in the methyl-CpG-binding protein 2 (MECP2) gene. Our study examines the genetic basis of typical and atypical RTT, along with RTT-like phenotypes, using MECP2-targeted sequencing (TS) and exome sequencing (ES).
METHODS: MECP2 sequencing was conducted on 27 Bangladeshi female participants with RTT features. ES was subsequently conducted on the 13 participants who tested negative for MECP2 variants. Data were processed using the Genome Analysis Toolkit and Ameri

BACKGROUND: Inherited protein C deficiency is an established genetic risk factor for venous thromboembolism. The pathogenic relevance and clinical interpretation of exonic and near-splice PROC variants, however, remain poorly understood.
OBJECTIVES: We systematically investigated 15 PROC variants located at exon-intron boundaries (±20 bp) or within coding regions, integrating splicing, protein function, and computational prediction to refine variant classification and explore RNA-based correction.
METHODS: Variant effects on splicing and protein function were first evaluated using in silico

There are over 130 genetic counselor (GC) training programs documented in more than 30 countries. Some regions also have developed practice-based competencies and guidelines for accrediting GC training, often including lists of required curriculum items (for example: ACGC, CAGC, EBMG, HGSA). However, areas where genetic counseling is emerging or desired may struggle to create training approaches from these often-aspirational knowledge and skill development lists. Down through history, biological arguments have often been used to legitimize a social gender order characterized by male supremacy. In the 1990's, a lively debate on the biological grounds of gender differences once again emerged in various fields. In the present article, the biological models used for explaining cognitive and behavioral gender differences are scrutinized, and recent research is discussed in light of history. These biological models emanate from theories about sex hormones, genetics and brain anatomy. Regarding the cognitive effects of sex hormones, no consensus has been reac

Drug addiction, or substance use disorder (SUD), is a chronic, relapsing disorder in which compulsive drug-seeking and drug-taking behaviour persist despite serious negative consequences. Drug abuse represents a problem that deserves great attention from a social point of view, and focuses on the importance of genetic studies to help in understanding the genetic basis of addiction and its medical treatment. Despite the complexity of drug addiction disorders, and the high number of environmental variables playing a role in the onset, recurrence, and duration of the symptoms, several studies hav

A woman in her 20s with a past medical history of surgical debulking of a right neck mass presented to the hospital for persistent and worsening right shoulder pain. The shoulder pain was associated with trismus and back and neck pain. A CT scan of the neck with contrast revealed post-surgical changes with increased heterotopic ossification throughout the surgical site extending to the supraclavicular soft tissues and the left sternocleidomastoid muscle, suggesting muscle ossification. A biopsy was performed, and the patient was diagnosed with myositis ossificans (MO). Initial treatment began

Geneticization is a broad term referring to several related processes such as a spreading tendency to use a genetic model of disease explanation, a growing influence of genetics in medical practice, and the slow changing of individual and societal attitudes towards reproduction, prevention and control of disease. These processes can be demonstrated in medical literature on preventive genetic screening and counselling programs for beta-thalassaemia in Cyprus, the United Kingdom and Canada. The preventive possibilities of the new genetic and diagnostic technologies have been quickly understood a

Research in genetics and the collaboration of various medical disciplines are responsible for the discovery of many new syndromes. The association of ocular and skin malformations is not astonishing when one remembers that the embryonic development of the eye and the skin are of ectodermal origin; both the eye and skin may reflect a variety of systemic disturbances occurring during their formation, or represent genetic disorders. An ocular and a dermatological feature may be the first indication of a possible disturbance on other organs. The reality that juveniles may frequently present simult

We describe a patient with signs and symptoms of virilization caused by 21-hydroxylase deficiency. The patient, a Hispanic woman, first sought medical attention at age 24, when she presented to a medical clinic with an uncomplicated urinary tract infection. At that time several signs of virilization were noted and she was referred to the endocrinology clinic. Evaluation revealed temporal balding, hyperpigmentation, acne, absent breast development, a muscular habitus, and clitoromegaly. Radiological studies revealed bilaterally enlarged adrenal glands and ovaries. Laboratory evaluation revealed

The incidence of genetic abnormalities in our population is rather high. About one out of 13 liveborn children will sooner or later suffer from a fully or partially genetically determined malformation or disease. With the use of numerical models ist has been attempted to predict the dysgenic effects of modern medicine and medical genetics in the next few generations of the Swiss population. Modern medical treatment will result foremost in an increase of multifactorial diseases and malformations. However, the impact on society will probably be smaller than the actual burden of chronic genetic d

There is little causal evidence on factors that can protect individuals against Alzheimer's Disease and Related Dementias (ADRD) risk. We study the causal effect of education on ADRD, exploiting a regression discontinuity generated by a compulsory schooling reform. ADRD was ascertained based on medical history, hospital records, and death registries, addressing concerns about selective sample attrition. We find that education reduces incidence of ADRD. Using molecular genetic data, we show that the reform weakened the relationship between genetics and ADRD incidence, implying genetic risk of d

Adults with congenital heart disease make up most patients with congenital heart disease vastly surpassing the pediatric patients largely because of significant improvements in the medical, interventional, and surgical approaches. An increasing body of evidence highlights the impact of noncardiac morbidity and mortality in these patients. Malignancy is a known major cause of death in adult patients with congenital heart disease. The causes are multifactorial, including genetics, radiation, delayed age-appropriate screening, anatomy variations, and thymectomy. This article provides an overview

I am the Huntington Sheldon Professor of Medical Discovery in the Department of Molecular Biology and Genetics in the School of Medicine at the Johns Hopkins University, where I have been running a lab for 30 years. Our research focusses on the molecular control of embryonic polarity and germline development, with an emphasis on asymmetric cell division and biomolecular condensates. We have uncovered mechanisms that localize proteins and RNAs in the cytoplasm by controlling protein diffusion and RNA condensation. My lab has also characterized a repressive program that launches the germline by
