Lack of genetic correlation and causal association between specific gut microbiota and osteoporosis.

Several observational studies have reported the change in gut microbiota (GM) composition in individuals with osteoporosis (OP); however, the genetic correlation and causal association between GM and OP remain undetermined. Large-scale genome-wide association study summary analysis of GM from the MiBio-Gen consortium and OP from the UK Biobank were used for analysis. Bivariate linkage disequilibrium score regression was conducted to explore the genetic correlation between GM and OP. Bidirectional 2-sample Mendelian randomization (MR) studies were then used to determine the causal association b

Association of polymorphisms of NOD2, TLR4 and CD14 genes with susceptibility to gastric mucosa-associated lymphoid tissue lymphoma.

BACKGROUND: The clinical course of Helicobacter pylori infection is highly variable and is influenced by both microbial and host factors, including the genetic composition of the infecting strains and variations in the host immune responses. A genetic risk profile for gastric cancer has been identified, but genetic susceptibility to develop gastric mucosa-associated lymphoid tissue (MALT) lymphoma is unclear. The aim of this study was to evaluate the relationship between NOD2, TLR4 and CD14 genetic polymorphisms and the development of gastric MALT-lymphoma. MATERIALS AND METHODS: Fifty-six pa

Intratumoral microbiota-host gene associations characterize stage-associated microecological patterns in colorectal cancer.

INTRODUCTION: Intratumoral microbiota are emerging as important ecological components of the colorectal cancer (CRC) microenvironment. However, their stage-associated patterns and coordination with host gene expression remain poorly understood. This study aimed to characterize stage-associated associations between intratumoral microbiota and the host transcriptome in CRC. METHODS: We profiled 73 regional tumor specimens from 30 independent, treatment-naïve CRC patients using full-length 16S rRNA gene sequencing and mRNA sequencing. Microbial diversity, predicted functional profiles, co-occurr

Microbial metabolic profiling reshapes NF-κB-mediated immune metabolic network: a new mechanism for CRC development.

BACKGROUND: The progression of colorectal cancer (CRC) is driven by multiple factors, including genetic mutations, metabolic reprogramming, immune escape, and dysbiosis of the intestinal microbiota. NF-κB acts as a central signaling hub within the tumor microenvironment that integrates inflammatory, metabolic, and immune cues to promote CRC progression. METHODS: This review systematically summarizes the roles of key immune pathways and metabolic reprogramming in CRC pathogenesis and outlines CRC-associated alterations in gut microbial profiles. The synthesis of functional microbial metabolites

Vitamin D and Microbiome: Molecular Interaction in Inflammatory Bowel Disease Pathogenesis.

Studies of systemic autoimmune diseases point to characteristic microbial patterns in various diseases, including inflammatory bowel disease (IBD). Autoimmune diseases, and IBD in particular, show a predisposition to vitamin D deficiency, leading to alterations in the microbiome and disruption of intestinal epithelial barrier integrity. This review examines the role of the gut microbiome in IBD and discusses how vitamin D-vitamin D receptor (VDR)-associated molecular signaling pathways contribute to the development and progression of IBD through their effects on gut barrier function, the micro

Elucidating applied voltage on the fate of antibiotic resistance genes in microbial electrolysis cell: Focusing on its transmission between anolyte and biofilm microbes.

Microbial electrolysis cell (MEC) system to treat wastewater containing antibiotics has been researched actively in past years. However, the fate of antibiotic resistant genes (ARGs) in MEC is not fully revealed. The effect of applied voltage on the migration of ARGs between anolyte and biofilm microbes via examining the microbial physiology and abundances of macrolide resistance genes (MRGs) and mobile genetic elements (MGEs) was elucidated in this research. Results showed that the abundance of MRGs and MGEs was decreased in the anolyte, but their abundances were increased on the electrode bi

Gut microbiome markers in subgroups of HLA class II genotyped infants signal future celiac disease in the general population: ABIS study.

Although gut microbiome dysbiosis has been illustrated in celiac disease (CD), there are disagreements about what constitutes these microbial signatures and the timeline by which they precede diagnosis is largely unknown. The study of high-genetic-risk patients or those already with CD limits our knowledge of dysbiosis that may occur early in life in a generalized population. To explore early gut microbial imbalances correlated with future celiac disease (fCD), we analyzed the stool of 1478 infants aged one year, 26 of whom later acquired CD, with a mean age of diagnosis of 10.96 ± 5.6 years.

Understanding the physiological functions of the host xenobiotic-sensing nuclear receptors PXR and CAR on the gut microbiome using genetically modified mice.

Pharmacological activation of the xenobiotic-sensing nuclear receptors pregnane X receptor (PXR) and constitutive androstane receptor (CAR) is well-known to increase drug metabolism and reduce inflammation. Little is known regarding their physiological functions on the gut microbiome. In this study, we discovered bivalent hormetic functions of PXR/CAR modulating the richness of the gut microbiome using genetically engineered mice. The absence of PXR or CAR increased microbial richness, and absence of both receptors synergistically increased microbial richness. PXR and CAR deficiency increased

Immune-mediated genesis of multiple sclerosis.

Multiple sclerosis (MS) is widely acknowledged to be an autoimmune disease affecting the neuronal myelin structure of the CNS. Autoantigens recognized as the target of this autoimmune process are: myelin basal protein, anti-proteolipid protein, antimyelin-associated glycoprotein and antimyelin-based oligodendrocytic basic protein. Ample evidence supports the idea of a dysregulation of immunological tolerance towards self-antigens of neuronal myelin structure triggered by one or more viral or bacterial microbial agents in predisposed HLA gene subjects. Genetic predisposition to MS has been high

Functional activity and functional gene diversity of a Cu-contaminated soil remediated by aided phytostabilization using compost, dolomitic limestone and a mixed tree stand.

Trace elements (TEs) availability, biochemical activity and functional gene diversity was studied in a Cu-contaminated soil, revegetated for six years with a mixed stand of willow, black poplar, and false indigo-bush, and amended or not with compost plus dolomitic limestone (OMDL). The OMDL amendment significantly reduced Cu and As availability and soil toxicity, and increased the biochemical activity and microbial functional diversity assessed with the GEOCHIP technique, as compared to the unamended soil (Unt). The OMDL soil showed significantly higher abundance of 25 functional genes involve

Primary biliary cirrhosis: environmental risk factors.

Primary biliary cirrhosis (PBC) is an autoimmune disease of unclear etiology. It is a chronic, progressive condition that causes intrahepatic ductal destruction ultimately leading to symptoms of cholestasis, cirrhosis and liver failure. The disease predominantly affects middle aged Caucasian women. It has a predilection to certain regions and is found in higher incidences in North America and Northern Europe. It also has a genetic predisposition with a concordance rate of 60% among monozygotic twins. Combinations of genetic and environmental factors are proposed in the pathogenesis of this dis

Microbial Profile Alteration as a Potential Biomarker and Therapeutic Target in Children with Celiac Disease: A Narrative Review.

Celiac disease (CD) is an autoimmune disorder triggered by gluten ingestion in genetically susceptible individuals. While gluten exposure is a necessary environmental trigger, its interaction with the intestinal microbiota has recently been recognized as a critical factor in modulating the onset and progression of the disease. Emerging evidence suggests that compositional and functional alterations in the microbial community may contribute to CD's pathogenesis by influencing intestinal barrier integrity, antigen presentation, and the balance between pro- and anti-inflammatory immune responses.

Enantiomer-specific effects of tebuconazole on antibiotic resistance gene and potential spread across the soil-vegetable continuum.

Although pesticides can exert enantioselective effects on microbial communities in agroecosystems, their roles in shaping antibiotic resistance gene (ARG) dissemination across the soil-vegetation continuum remains poorly understood. Here, we investigated the effects of the R- and S-enantiomers of tebuconazole, one of the most widely used fungicides in the global market, on ARGs, mobile genetic elements (MGEs), bacterial communities, and potential pathogens across a soil-pak choi continuum. S-tebuconazole consistently had stronger effects than R-tebuconazole, increasing the ARG abundance across

Preemptive biofilm colonization blocks microbial metal corrosion.

Sustainable materials are needed to address the serious economic and safety risks of microbial metal corrosion. Colonizing metal surfaces with biofilms of noncorrosive microbes was previously shown to reduce aerobic, abiotic corrosion. However, the ability of biofilms to thwart highly corrosive anaerobic microbes is untested. Here we report on a strain of Escherichia coli genetically modified for enhanced metal adherence and adaptively evolved to tolerate sulfide. The E. coli biofilms effectively inhibited all known major routes for anaerobic microbial iron corrosion, including proton and sulf

Integrative Approaches to Gut Microbiota: Biomarkers, Dysbiosis, and Therapeutic Potential.

The human gut contains bacteria, viruses, and archaea that influence health depending on age, diet, and location. An individual harbours about 150 bacterial species. An imbalance of these microorganisms, known as dysbiosis, can negatively impact health. Commensal bacteria maintain microbial homeostasis by building short chain fatty acids. The 2-way communication channel that connects the gut and the brain is referred to as the gut-brain axis. Dysbiosis of the gut leads to some autoimmune diseases. A decline of beneficial bacteria causes the progression of rheumatoid arthritis. Gut dysbiosis in

Therapeutic Applications of Antimicrobial Silver-Based Biomaterials in Dentistry.

Microbial infection accounts for many dental diseases and treatment failure. Therefore, the antibacterial properties of dental biomaterials are of great importance to the long-term results of treatment. Silver-based biomaterials (AgBMs) have been widely researched as antimicrobial materials with high efficiency and relatively low toxicity. AgBMs have a broad spectrum of antimicrobial properties, including penetration of microbial cell membranes, damage to genetic material, contact killing, and dysfunction of bacterial proteins and enzymes. In particular, advances in nanotechnology have improve

Gut Microbiota in Psoriasis: Evidence for Dysbiosis, Pathological Mechanisms, Causality, and Therapeutic Implications.

Psoriasis is a chronic inflammatory skin disease driven by genetic susceptibility, environmental triggers, and immune dysregulation, with a global prevalence of approximately 2%-3%. In recent years, the gut microbiota has garnered increasing attention as a key interface linking environmental factors to host immunity in the pathogenesis of psoriasis. A growing body of research has revealed significant gut dysbiosis in patients with psoriasis, characterized by reduced microbial diversity, depletion of specific beneficial bacteria, and enrichment of opportunistic pathogens. Mechanistically, the g

Amphipods associate with epibiotic filamentous bacteria at a shallow-water hydrothermal vent.

In the deep ocean, hydrothermal vents sustain remarkable animal-microbial symbioses, in which bacteria colonizing animal external or internal tissues use reduced chemicals from venting fluids to perform chemosynthesis and nourish their host. Hydrothermal vents also occasionally occur within the photic zone, but the prevalence of photosynthesis within these settings is considered to override the development of chemosymbiotic interactions; animal-microbial associations such as those observed at deep-sea vents have rarely been recorded at shallow vents. During 2019 sampling of the Strýtan hydroth

The role of the microbiome in gynecological cancers: implications for diagnosis and treatment.

Gynecological malignancies such as cancer of the cervix, ovary, endometrium, vulva, and vagina pose a severe global health burden. Although conventionally attributed to genetic mutation, hormonal imbalance, and chronic viral infection, including high-risk human papillomavirus, recent evidence suggests that the human microbiome plays a central role in their pathogenesis and development. This review summarizes existing evidence that microbial dysbiosis, specifically the depletion of beneficial Lactobacillus species and overrepresentation of anaerobic organisms such as Fusobacterium, Atopobium, a
