Immunohistochemical expression of growth factors in subacute thyroiditis and their effects on thyroid folliculogenesis and angiogenesis in collagen gel matrix culture.

The inflammatory-mechanistic basis of subacute thyroiditis remains unclear. To elucidate the roles of vascular endothelial cell growth factor (VEGF), basic fibroblast growth factor (bFGF), platelet-derived growth factor-BB (PDGF), transforming growth factor-beta1 (TGF-beta1) and epidermal growth factor (EGF) in the inflammatory process, their immunoexpression was examined in biopsy specimens of ten cases. At the granulomatous stage, all cases expressed VEGF, bFGF, PDGF, and TGF-beta1 in monocytes/macrophages infiltrating into follicle lumina, and in both epithelioid histiocytes and multinuclea

Novel growth factors involved in the pathogenesis of proliferative vitreoretinopathy.

AIMS: To determine whether hepatocyte growth factor (HGF) and connective tissue growth factor (CTGF) are expressed in human specimens of proliferative vitreoretinopathy (PVR) and to propose a model of PVR pathogenesis based upon the known activities of these growth factors. Methods Immunohistochemical methods (ABC Elite) were used to demonstrate the presence of HGF and CTGF in cryostat sections of five human PVR membranes. RESULTS: In each of the five PVR membranes, stromal cells were immunohistochemically positive for both HGF and CTGF. Based upon this information and the known actions of th

Glypican-3 modulates BMP- and FGF-mediated effects during renal branching morphogenesis.

The kidney of the Gpc3-/ mouse, a novel model of human renal dysplasia, is characterized by selective degeneration of medullary collecting ducts preceded by enhanced cell proliferation and overgrowth during branching morphogenesis. Here, we identify cellular and molecular mechanisms underlying this renal dysplasia. Glypican-3 (GPC3) deficiency was associated with abnormal and contrasting rates of proliferation and apoptosis in cortical (CCD) and medullary collecting duct (MCD) cells. In CCD, cell proliferation was increased threefold. In MCD, apoptosis was increased 16-fold. Expression of Gpc3

The response of small vessel endothelial cells from fetal rat lung to growth factors.

Small vessel pulmonary endothelial cells were obtained from rat fetal lung at day 20 of gestation, and were maintained in culture to passage three for study. Endothelial cells grown on a collagen matrix with Dulbecco's minimal essential medium: Ham's F12 medium (1:1, v/v) supplemented with 20 ml/l fetal bovine serum, bovine pituitary extract (50 mg/l), endothelial cell growth supplement (100 mg/l), hydrocortisone (1 mg/l) and an increased (10 mmol/l) magnesium concentration retained the characteristic endothelial cell marker factor VIII antigen during the third passage in culture. The factors

Alterations in growth plate and articular cartilage morphology are associated with reduced SOX9 localization in the magnesium-deficient rat.

Insufficient dietary magnesium (Mg) intake has been associated with low bone mass in humans, and recent basic science studies have indicated that this bone loss may be secondary to increased release of substance P and TNFc Much less is known about the effects of low Mg intake on cartilage. We have evaluated growth plate and articular cartilage in rats following a 6 month dietary Mg restriction. Histomorphometry demonstrated significantly decreased distal femur articular cartilage chondrocyte density and decreased tibial growth plate width in experimental animals compared to controls. Growth pla

The zinc-reversible antimicrobial activity of neutrophil lysates and abscess fluid supernatants.

There is some evidence to suggest that microbial growth inhibition may occur in chronic abscesses. A substance perhaps responsible for this phenomenon is calprotectin, a neutrophil cytoplasmic protein that inhibits microbial growth and that belongs to a class of proteins often having specific binding sites for zinc. In the present study, the suppressive effects of either human or mouse neutrophil lysates on Candida albicans growth were found to be completely reversed by micromolar quantities of zinc but not by iron or other trace elements. Similarly, supernatants of exudates from experimental

The Relation of Change in Hostility and Sociability During Childhood to Substance Use in Mid Adolescence.

In a cohort-sequential longitudinal study (N = 1,075), we related change in children's hostility and sociability assessed from 1(st)-8(th) grade to their use of cigarettes, alcohol, and marijuana assessed from 9(th)-12(th) grade. Children who were more hostile at 1(st) grade, and had higher rates of growth of hostility, used more of all three substances at 9(th) grade, and those with higher initial levels of hostility increased their use of cigarettes and marijuana from 9(th) to 12(th) grade. Children who were more sociable at 1(st) grade used more alcohol at 9(th) grade. These findings demons

Perinatal substance abuse and the drug-exposed neonate.

Tobacco and alcohol are the substances most abused during pregnancy. Alcohol (ethanol) is the human teratogen that produces the most serious neurobehavioral effects on the fetus. Cocaine is associated with spontaneous abortions, premature labor, precipitous labor, stillbirths, meconium staining and abruptio placentae. Heroin use during pregnancy has been associated with low birth weight, miscarriage, prematurity, microcephaly and intrauterine growth retardation. Marijuana is not scientifically linked to significant teratogenic effects. Since most substance abusers use multiple drugs, a positiv

Influence of gestational salt restriction in fetal growth and in development of diseases in adulthood.

Recent studies reported the critical role of the intrauterine environment of a fetus in growth or the development of disease in adulthood. In this article we discussed the implications of salt restriction in growth of a fetus and the development of growth-related disease in adulthood. Salt restriction causes retardation of fatal growth or intrauterine death thereby leading to low birth weight or decreased birth rate. Such retardation of growth along with the upregulation of the renin angiotensin system due to salt restriction results in the underdevelopment of cardiovascular organs or decrease

Expression of proinflammatory cytokines and growth factors at the injured growth plate cartilage in young rats.

Growth plate, a cartilage-like tissue responsible for bone lengthening in children, has limited abilities to regenerate. After injury, a bony repair often occurs at the injury site, which may lead to growth arrest or angulation of the involved long bone. Our previous study has demonstrated inflammatory, fibrogenic, and osteogenic responses at the injured growth plate; however, the underlying mechanisms remain unclear. In this study, mRNA expression patterns of pro-inflammatory cytokines, growth factors, transcription factors, and matrix proteins were examined by quantitative RT-PCR over a 35-d

Significance of serum human hepatocyte growth factor levels in patients with hepatic failure.

Serum human hepatocyte growth factor levels were measured using a newly developed enzyme-linked immunosorbent assay kit in patients with liver diseases. Serum human hepatocyte growth factor levels were increased in correlation with derangements of prothrombin time, total bilirubin and other parameters reflecting hepatocellular dysfunction in 112 patients with chronic liver disease. The levels were positively correlated with serum AST and ALT levels in 59 of these patients whose prothrombin times were within the normal range. Abnormally increased serum human hepatocyte growth factor levels were

Human breast epithelial cells in serum-free collagen gel primary culture: growth, morphological, and immunocytochemical analysis.

Human breast epithelial cells derived from various sources (fibroadenoma, reduction mammoplasty, and mastectomy tissues from premenopausal patients) have been cultured in collagen gel matrix using serum-free medium. Response to various additives has been analyzed for growth-promoting effect when added to a basal medium containing insulin, cholera toxin, and BSA. A consistent observation has been the effect of EGF and cortisol in growth stimulation of human breast epithelial cells, while separately, each additive elicited only a small response. Under this condition, employing EGF and cortisol c

Removal of metabolites, cytokines and hepatic growth factors by extracorporeal liver support in children.

BACKGROUND: Molecular Adsorbents Recirculating System (MARS)-mini has recently been approved and applied in children with hepatic failure. However, its indication, efficacy and capability to induce liver regeneration remain unclear. The aim of our pilot study in children was to analyse the impact of MARS on markers of detoxification and regeneration. METHODS: In children with fulminant Wilson's disease and bridged with MARSmini for liver transplantation, we analyzed toxic metabolites (bile acids, bilirubin, lactate, ammonia, tryptophan and copper), regulators of the inflammatory cascade [nitr

Neurokinin-1 receptor and substance P messenger RNA levels increase during intraabdominal adhesion formation.

BACKGROUND: Intraabdominal peritoneal adhesions are a significant cause of postoperative morbidity and remain one of the major long-term complications associated with abdominal surgery. Adhesion formation at the molecular level involves a complex interaction of cytokines, growth factors, cell adhesion molecules, and neuropeptides, as well as many other factors secreted by cells proximate to the traumatized area. Limited studies exist which investigate the molecular processes involved in adhesion formation. Therefore, the aim of the present study was to determine the pattern of gene expression

Natural Cell-Inspired Integration of Redox Reactant Molecules in Hierarchical MOFs Harvested for Reaction Pathway-Regulated Glycoprotein Assay.

Natural cell systems with multiple isolated compartments possess controllable signal transduction pathways, which can segregate a series of complex substances and confine them from each other to allow for feedback-driven reactions. By biomimicking the ingenuity of cell systems, we herein designed spatially compact metal-organic frameworks (MOFs) as a hierarchical host for complex substances at isolated locations for the development of reaction pathway-regulated glycoprotein assay. The well-defined zeolitic imidazolate framework-8 (ZIF-8) matrices were fabricated via epitaxial growth to organiz

Expression Profiling of Candidate Genes in Sugar Beet Leaves Treated with Leonardite-Based Biostimulant.

Leonardite-based biostimulants are a large class of compounds, including humic acid substances. Foliar application of biostimulants at field level improves plant growth, yield and quality through metabolic changes and stimulation of plant proton pumps. The present study aimed at identifying optimum dosage of BLACKJAK, a humic acid-based substance, which is able to modify genes involved in sugar beet growth. Thirty-three genes belonging to various biochemical pathway categories were tested in leaves of treated sugar beet (Beta vulgaris L.) samples to assess gene expression profiling in response
