Morphological and flow-cytometric analysis of haemin-induced human neutrophil activation: implications for transfusion-related acute lung injury.

BACKGROUND: Transfusion-related acute lung injury (TRALI) is associated with vascular endothelial cell injury following neutrophil activation. Recently, it has been suggested that haem-related molecules induce activation of neutrophils and that erythrocyte-derived substances contained in blood preparations are involved in TRALI. We observed the morphological effects and reactive oxygen species (ROS) production of haem-related molecules and investigated the effects of signal transduction inhibitors on haem-induced neutrophil activation. MATERIALS AND METHODS: The polymorphonuclear cell fractio

Heme oxygenase attenuates allergen-induced airway inflammation and hyperreactivity in guinea pigs.

Heme oxygenase (HO), the heme-degrading enzyme, has shown anti-inflammatory effects in several models of pulmonary diseases. HO is induced in airways during asthma; however, its functional role is unclear. Therefore, we evaluated the role of HO on airway inflammation [evaluated by bronchoalveolar lavage (BAL) cellularity and BAL levels of eotaxin, PGE(2), and proteins], mucus secretion (evaluated by analysis of MUC5AC gene expression and periodic acid-Schiff staining), oxidative stress (evaluated by quantification of 4-hydroxynonenal adducts and carbonylated protein levels in lung homogenates)

Alcohol-water as a novel medium for beta-hematin preparation.

It is shown that alcohol-water mixtures (e.g., 43 wt% ethanol) provide a suitable medium for the efficient production in high yield of beta-hematin from ferriprotoporphyrin IX (FP), as measured by infrared spectroscopy. Previous and present light-absorption data of FP, obtained under specific acid conditions in an ethanol-water medium, suggest the presence of FP monomers, which are considered to promote the reaction leading to beta-hematin. Other aspects of the mechanism of reaction are discussed.

Thiolated gamma-cyclodextrin-polymer-functionalized CeFe3O4 magnetic nanocomposite as an intrinsic nanocatalyst for the selective and ultrasensitive colorimetric detection of triacetone triperoxide.

Explosives are powerful destructive weapons used by criminals and terrorists across the globe and their use within military installation sites poses serious environmental health problems. Existing colorimetric sensors for triacetone triperoxide (TATP) relies on detecting its hydrolysed H2O2 form. However, such detection strategy limits the practicability for on-site TATP sensing. In this work, we have developed a novel peroxidase mimic catalytic colorimetric sensor for direct recognition of TATP. Ceria (Ce)-doped Fe3O4 nanoparticles (CeFe3O4) were synthesized via the hot-injection organic synt

Synthesis, β-haematin inhibition, and in vitro antimalarial testing of isocryptolepine analogues: SAR study of indolo[3,2-c]quinolines with various substituents at C2, C6, and N11.

A series of indolo[3,2-c]quinolines were synthesized by modifying the side chains of the ω-aminoalkylamines at the C6 position and introducing substituents at the C2 position, such as F, Cl, Br, Me, MeO and NO2, and a methyl group at the N11 position for an SAR study. The in vitro antiplasmodial activities of the derivative agents against two different strains (CQS: NF54 and CQR: K1) and the cytotoxic activity against normal L6 cells were evaluated. The test results showed that compounds 6k and 6l containing the branched methyl groups of 3-aminopropylamino at C6 with a Cl atom at C2 exhibited

Characterization of Staphylococcus epidermidis and Staphyloccocus warneri small-colony variants associated with prosthetic-joint infections.

We determined the frequency of isolation of staphylococcal small-colony variants (SCVs) from 31 culture-positive patients undergoing revision of total hip prosthesis for aseptic loosening or presumed prosthetic-joint infection (PJI). We analysed auxotrophy of cultured SCVs, their antimicrobial susceptibility profiles and their biofilm-forming capacity. Eight SCV strains were cultivated from six (19 %) patients. All SCVs were coagulase-negative staphylococci (CNS) with Staphylococcus epidermidis as the predominant species; there was also one Staphylococcus warneri SCV. The SCVs were auxotrophic

Steinernema feltiae (= Neoaplectana carpocapsae): effect of sterols and hypolipidemic agents on development.

The structure and concentration of sterol in a lipid-defined artificial medium affected the development of the entomogenous nematode, Steinernema feltiae (= Neoaplectana carpocapsae). The nematode grew normally in vitro when the medium was supplemented with delta 5-desalkylsterol (cholesterol) or delta 5-desalkylsteryl ester (cholesterol oleate). The minimum amount of cholesterol in the medium that was necessary to support the development of S. feltiae to the climax population (i.e., dauer stage) was 0.0025%. The nematode also completed its life cycle normally when delta 0- or delta 7-desalkyl

The Role of Cytochrome P450 Holoenzyme Metabolism in the Origin of Neuropathologies.

The increased activity of δ-aminolevulinic acid synthase-1 (ALAS1) leads to the accumulation of δ-aminolevulinic acid (ALA), which may reduce the iron(III) retention ability of ferritin, resulting in iron(III) overload. The accumulation of ALA occurs in the condition of attenuation of the heme negative feedback loop over ALAS1 activity in concert with the induction of ALAS1 expression. Attenuation of the heme negative feedback loop is maintained by elevated quantities of the heme catabolizing enzyme, heme oxygenase-1 (HO-1), which, in turn, may be induced by highly increased heme concentration

Genomic characterization of a vancomycin-intermediate Staphylococcus aureus (VISA) small colony variant after long-term antibiotic therapy.

OBJECTIVES: Vancomycin (VCM)-intermediate Staphylococcus aureus (VISA) and small-colony variants (SCVs) are clinically important phenotypes that are frequently difficult to treat and commonly encountered in persistent or recurrent infections. We previously reported a VISA strain, HV2019-1, isolated during a relapse episode of pacemaker-associated bloodstream infection. In the present study, we further investigated its phenotypic characteristics and genomic background. METHODS: Colony size, hemolytic activity, and pigmentation were quantitatively evaluated. To elucidate the genetic basis under

Vitamin D alleviates intracerebral hemorrhage symptoms by modulating pro-inflammatory/anti-inflammatory phenotypic transformation of microglia through P2X7R activation of NLRP3.

OBJECTIVE: The primary objective of this study was to analyze the potential influence of vitamin D on microglial phenotypic transformation in cerebral hemorrhage. METHODS: To simulate cerebral hemorrhage conditions, a hemin-induced brain hemorrhage cell model was established, and cell viability was subsequently measured by CCK-8 assay. Western blotting was then performed to detect the expression of P2X7R, NLRP3, and microglial phenotypic transformation marker proteins. The expression of CD16/32 and CD206 was assessed by immunofluorescence, and ELISA was used to measure levels of inflammatory

Identification of two Cryptococcus neoformans heme transporters involved in Fhb1-mediated nitrosative stress protection in a fission yeast model.

In Schizosaccharomyces pombe and Cryptococcus neoformans, the flavohemoglobins (flavoHbs) that protect these yeasts against nitrosative stress are Yhb1 and Fhb1, respectively. Although heme is an essential cofactor for flavoHb activity, the molecular circuitry governing the acquisition of exogenous heme by flavoHbs remains largely unknown. Because S. pombe is a powerful model organism for studying heme biology, Fhb1 was heterologously expressed in S. pombe mutant cells lacking Yhb1. Fhb1 expression functionally complements yhb1Δ cells that are defective in nitric oxide (˙NO) detoxification. Mo

Ascorbate-catalyzed reductive nitrosylation of ferric myoglobin.

Myoglobin plays a key role in oxygen storage and delivery through oxygen binding and release from the ferrous (Fe+2) heme. However, when the heme is oxidized to its ferric (Fe+3) form it can no longer bind oxygen and further oxidation leads to lipid peroxidation which can lead to a variety of pathological consequences. Ferric heme can be reduced to the ferrous form by nitric oxide (NO•) through classical reductive nitrosylation but this reaction, where a second NO• molecule binds to the reduced heme to form iron nitrosyl myoglobin, is very slow and inefficient. We have recently demonstrated th

Iron-doped carbon dots with multi-enzyme activities for rapid determination of phenolic compounds.

Multi-enzymic nanozymes have attracted growing attention due to their distinct advantages over single enzyme-like nanozymes, particularly their synergistic effects and cascaded reactions. Herein, iron-doped carbon dots (FeCDs) were prepared by a one-step calcination method using hemin chloride, histidine, and potassium citrate as precursors. The resultant FeCDs exhibit a monodispersed spherical structure with an average particle size of 1.1 nm, where iron acts as a key catalytic active center. Enzyme activity experiments demonstrate that FeCDs exhibit peroxidase-like, catalase-like, and photo-

Inhibition of Complement Will Substantially Reduce the Anemia of Malaria.

There is compelling evidence that complement plays a major role in the elimination of nonparasitized erythrocytes (np-Es) in the anemia of malaria. In fact, for every erythrocyte destroyed after infection by Plasmodium falciparum (Pf) or Plasmodium vivax, at least 10 np-Es are cleared, most likely due to C3b opsonization. In malaria, complement is activated by three breakdown products released from lysed parasitized erythrocytes (p-Es): hemin/hematin, the digestive vacuole (DV), and hemozoin. Both childhood anemia in malaria and post-artesunate delayed-hemolysis in individuals with malaria tre

C-type lectin-like receptor-2 in platelets mediates ferric chloride-induced platelet activation and attenuates ferroptosis of endothelial cells.

BACKGROUND: An iron overload status induces ferroptosis, an iron-dependent nonapoptotic cell death, in various pathological conditions. We previously reported that hemin (heme), protoporphyrin-IX with ferric iron, activates platelets via C-type lectin-like receptor-2 (CLEC-2) and glycoprotein VI/FcRγ, but protoporphyrin-IX alone blocks CLEC-2-dependent platelet activation. Therefore, we hypothesized that free iron has the ability to activate platelets. OBJECTIVES: This study aimed to elucidate platelet activation mechanisms of iron (ferric chloride), including the identification of signaling

Escherichia coli with increased aminoglycoside resistance due to an amino acid substitution at position 85 of HemC.

OBJECTIVE: The mechanism of aminoglycoside resistance due to abnormal hemin synthesis remains unclear. We investigate an Escherichia coli strain with a single amino acid substitution at position 85 of HemC. METHODS: An aminoglycoside-resistant Escherichia coli DH5α was selected by passaging in Lysogeny Broth (LB) medium containing amikacin. Whole genome sequencing was performed to determine the genetic profile of the strain. An isogenic strain of E. coli DH5α was created. Growth rates, drug susceptibilities and expressions of the heme synthetic genes were compared between the original strain

Development and Characterization of Novel Chitosan-Coated Curcumin Nanophytosomes for Treating Drug-Resistant Malaria.

This study aimed at enhancing the efficacy of curcumin (CR) by formulating and coating it with chitosan. In silico molecular docking studies revealed that CR exhibited almost similar and low binding energies when compared to artemisinin, indicating high stability at the target site. It can be confirmed that CR is effective in treating and reducing Plasmodium falciparum parasites. Fourier transform infrared studies confirmed that there was a shift and disappearance of some drug peaks in the formulation which revealed complexation with phospholipids. The F2EXT3-developed formulation exhibited gr

Free haem levels in gingival crevicular fluid and their relationship to periodontal clinical parameters, smoking and subgingival microbial composition.

BACKGROUND AND OBJECTIVES: Periodontitis caused by multifactorial polymicrobial infection results in a destructive inflammatory process and loss of tooth supporting tissues. Many putative bacterial virulence factors that cause host destruction are regulated by iron and haem. Therefore, this study investigated the free haem levels in the gingival crevicular fluid (GCF) at periodontitis sites in smokers and nonsmokers and their relationship to subgingival microbial composition. MATERIALS AND METHODS: A cross-sectional study was carried out on 78 patients with a split-mouth design who were divid
