Cutaneous graft-versus-host disease (GVHD) is a frequent manifestation after allogeneic hematopoietic stem cell transplantation. This review synthesizes classification, pathophysiology, risk factors, and diagnostic frameworks for acute and chronic cutaneous GVHD. Acute cutaneous GVHD typically presents as a morbilliform eruption, with severe cases presenting with bullae and full-thickness desquamation. Chronic cutaneous GVHD presents in more protean ways, including both nonsclerotic and sclerotic phenotypes. We review current diagnostic criteria, staging, clinical and histopathologic features,

The efficacy of chimeric antigen receptor (CAR)-T cell therapy in relapsed/refractory acute myeloid leukemia (R/R AML) is limited by tumor heterogeneity, antigen evasion, and treatment-related toxicities. Gamma delta (γδ) T cells mediate antitumor activity independent of MHC by sensing stress-induced ligands. A prominent mechanism involves NKG2D ligand (NKG2DL) recognition, which is highly upregulated in malignancies but generally low or restricted expression in healthy tissues under homeostatic conditions. In human peripheral blood, the Vδ2 subset represents the predominant population. Vδ2 T

Extracorporeal photochemotherapy (ECP) was introduced by Edelson and co-workers in 1985. Since then the therapy, though still controversial in specific indications, has received wide acceptance and is presently been used in over 160 centers in Europe and the U.S.A. for a number of important dermatological and non-dermatological indications. Convincing data in the past few years has documented that ECP is associated with a very low side-effect profile. As of 2000 ECP continues to be used in the treatment of cutaneous T-cell lymphoma, alone or in combination with other treatment modalities, and

PURPOSE: Resistance to anti-PD1-based immune checkpoint blockade (ICB) remains a problem for the treatment of metastatic melanoma. Tumor cells as well as host myeloid cells can express the immune checkpoint ligand CD155 to regulate immune cell function. However, the effect of tumor CD155 on the immune context of human melanoma has not been well described. This observational study characterizes tumor CD155 ligand expression by metastatic melanoma tumors and correlates results with differences in immune cell features and response to ICB.

Follicular lymphoma, the most common indolent lymphoma, though highly responsive to therapy is coupled with multiple relapses for the majority of patients. Advances in biologic understanding of molecular events in lymphoma cells and the tumor microenvironment, along with novel cellular and targeted therapies, suggest this may soon change. Here we first review the development of the molecular concepts and classification of follicular lymphoma, along with therapeutic development of treatments based on chemotherapy plus monoclonal antibodies targeting CD20. We then focus on developments over the

BACKGROUND: Lower respiratory tract infections developing during invasive mechanical ventilation in patients with severe COVID-19 acute respiratory distress syndrome (ARDS) present important diagnostic and therapeutic challenges. Streptomyces spp., an uncommon respiratory isolate, frequently raises uncertainty regarding contamination, colonization, or clinically relevant infection because standardized criteria for clinical interpretation are lacking.
CASE PRESENTATION: A 65-year-old woman with hypertension and obesity was admitted with severe COVID-19 ARDS requiring invasive mechanical ventil

Cancer arises from malignant cells that exist in dynamic multilevel interactions with the host tissue. Cancer therapies aiming to directly kill cancer cells, including oncogene-targeted therapy and immune-checkpoint therapy that revives tumour-reactive cytotoxic T lymphocytes, are effective in some patients1,2, but acquired resistance frequently develops3,4. An alternative therapeutic strategy aims to rectify the host tissue pathology, including abnormalities in the vasculature that foster cancer progression5,6; however, neutralization of proangiogenic factors such as vascular endothelial grow

Malaria remains a heavy public health and socioeconomic burden in tropical and subtropical regions. Increasing resistance against front-line treatments implies that novel targets for antimalarial intervention are urgently required. Protein kinases of both the parasites and their host cells possess strong potential in this respect. We present an overview of the updated kinome of Plasmodium falciparum, the species that is the largest contributor to malaria mortality, and of current knowledge pertaining to the function of parasite-encoded protein kinases during the parasite's life cycle. Furtherm

Alternative splicing (AS) significantly increases the diversity of the eukaryotic proteome, and alterations in AS induced by viruses have emerged as a novel approach to studying virus-host interactions. Human metapneumovirus (HMPV) interacts with host cells through multiple mechanisms, directly or indirectly utilising various host systems to facilitate infection and replication. In this study, the BEAS-2B human normal lung epithelial cell line was used as the cell model for HMPV infection. The host gene alternative splicing events following HMPV infection were then characterised using RNA sequ

Cell therapies are potential alternatives to organ transplantation for liver failure or dysfunction but are compromised by inefficient engraftment, cell dispersal to ectopic sites, and emboli formation. Grafting strategies have been devised for transplantation of human hepatic stem cells (hHpSCs) embedded into a mix of soluble signals and extracellular matrix biomaterials (hyaluronans, type III collagen, laminin) found in stem cell niches. The hHpSCs maintain a stable stem cell phenotype under the graft conditions. The grafts were transplanted into the livers of immunocompromised murine hosts

While considerable advances have been made in the understanding of the tissue-destructive mechanisms involved in etiology of periodontitis, the adoption and application of this knowledge has not been generally incorporated into the daily management of patients with chronic periodontitis. A number of adjunctive pharmacotherapeutic agents have been investigated in clinical trials and several agents are used in everyday practice as adjuncts to mechanical nonsurgical periodontal therapy. The only systemic, host modulatory agent approved for the adjunctive use with nonsurgical periodontal procedure

We present a highly unusual case of pulmonary Pseudallescheria boydii infection in a nonimmunocompromised host with a cavitating mass lesion. The diagnosis was confirmed by open lung biopsy. The patient was treated at another institution with course of amphotericin B, considered an ineffective therapy for this infection, and presented to us with direct extension and invasion of the left atrial appendage and the pulmonary artery, followed by massive pulmonary embolization and hematogenous dissemination to the liver, spleen, kidney, pancreas, and brain.

INTRODUCTION: Hand, foot, and mouth disease (HFMD) is a common enterovirus-induced illness primarily affecting children under 5 years of age. Enterovirus 71 (EV71) is a major causative agent associated with severe HFMD that can lead to fatal neurological complications. Despite available vaccines, no antiviral therapy with a clearly defined molecular mechanism has received approval. While most antiviral agents target viral components, host-targeting antiviral approaches remain underexplored. We first identified voxtalisib, a dual PI3K/mTOR inhibitor, through compound screening as a potential in

The unquestionable clinical success of left ventricular assist device (LVAD) implantation has, nevertheless, been accompanied by complications arising from interactions between the implanted biomaterial and the host immune system. The aberrant state of monocyte and T-cell activation resulting from these host/device interactions is accompanied by two parallel processes: (1) selective loss of Th1 cytokine producing CD4 T-cells through activation-induced cell death; and (2) unopposed activation of Th2 cytokine producing CD4 T-cells resulting in B-cell hyperreactivity and dysregulated immunoglobul

The human gastrointestinal tract harbors trillions of microorganisms that collectively exert profound influences on host physiology, particularly immune function. Emerging evidence reveals that gut microbiota modulates immune responses through intricate interactions with host microRNAs (miRNAs), small non-coding RNAs that post-transcriptionally regulate gene expression. This review synthesizes current understanding of how gut bacteria influence host immunity via miRNA-dependent mechanisms. We examine two primary pathways: first, the capacity of gut microbes to regulate host miRNA expression pr

UNLABELLED: Post-translational modifications orchestrate virus-host interactions, yet their roles in the hepatitis B virus (HBV) life cycle remain incompletely defined. The small hepatitis B surface antigen (SHBs)-the most abundant HBV protein-drives immune evasion, pathogenesis, and virion/subviral particle (SVP) production. Here, we identify UFL1, the E3 ligase of UFMylation, as a host factor that modifies and stabilizes SHBs by antagonizing its ubiquitination to potentiate HBV output and SHBs-driven pro-oncogenic activities. Using affinity purification-mass spectrometry, we discovered UFL1

Non-coding RNAs have emerged as critical regulators of the immune response to infection. MicroRNAs (miRNAs) are small non-coding RNAs which regulate host defense mechanisms against viruses, bacteria and fungi. They are involved in the delicate interplay between Mycobacterium tuberculosis, the causative agent of tuberculosis (TB), and its host, which dictates the course of infection. Differential expression of miRNAs upon infection with M. tuberculosis, regulates host signaling pathways linked to inflammation, autophagy, apoptosis and polarization of macrophages. Experimental evidence suggests

The gut microbiota is a critical regulator of systemic immune homeostasis; accumulating evidence implicates specific commensal bacteria, termed "pathobionts," in autoimmune disease pathogenesis. However, the definition of pathobionts remains context-dependent, as their effects are influenced by host genetics and host-microbe interactions. In this review, we summarize representative pathobionts supported by functional evidence in selected extraintestinal autoimmune diseases and discuss how these mechanisms may inform phage-based microbiome-targeted interventions. Mechanistically, pathobionts co

P. gingivalis, a keystone periodontal pathogen in periodontitis, is recognized for its capacity to evade host immune clearance. Its intracellular survival leads to both local and systemic infection, thereby increasing the risk of periodontitis and its associated systemic diseases. However, macrophages have limited capacity to clear P. gingivalis, which contributes to disease establishment and progression. Our findings confirmed P. gingivalis presence in periodontitis tissue, with intracellular P. gingivalis mainly located in macrophages. However, P. gingivalis cannot be completely cleared by m
